Propofol is an ultra-fast-acting intravenous anesthetic, which is rapidly metabolized primarily into inactive compounds in the live and then excreted in the urine. The purpose of this study is to explore the risk signals of propofol based on the US FDA Adverse Event Reporting System database. The risk signals of propofol-related adverse reactions in adverse event (AE) reports from 2004 to 2021 in the US FAERS were mined using ratio-report method (ROR) and the ratio-report ratio method (PRR) methods. We screened out 1651 pairs AE reports using propofol as primary suspect (PS) drugs. ROR, PRR, BCPNN and MGPS methods were used to calculate respectively, there are 363 positive preferred terms (PT) signals with 9549 cases. Among them, the top 3 adverse reactions associated with using propofol from the FAERS database were anaphylactic shock, hypotension and propofol infusion syndrome. The top 3 systems of the body associated with adverse reaction of propofol from the FAERS database were General disorders, Cardiac disorders and administration site conditions and Respiratory, thoracic and mediastinal disorders. The top 4 indication of using propofol from the FAERS database, including anaesthesia, induction of anaesthesia, sedation, general anaesthesia. There are many adverse reactions that are not included in the drug insert of propofol and involve a wide range of organs and/or systems. Caution should be exercised in the clinical use of propofol.

Leonurus japonicus Houtt is an important anti-skin pigmentation herb used in traditional Chinese medicine. However, the molecular basis for this activity is complex and not fully understood. In this study, water and ethanol extracts and polysaccharide extract from L. japonicus (LJPs) were analyzed by LC-MS/MS and HPLC-DAD separately. Cytotoxicity was analyzed by using CCK-8, antioxidant activity using flow cytometer, anti-MMPs, anti-tyrosinase and signalling pathway analysis using Western blotting to investigate their anti-melanogenesis function. The results showed that the water and ethanol extracts contained alkaloids, flavonoids, and phenolic acids. The LJPs mainly contain glucose, fucose, glucuronic acid, mannose, threonine and arginine, and structure characterization by FITR analyses indicated that LJPs have β- or α-D-glycosidic bonds and contain pyranose rings. The L. japonicus extracts displayed high cell viability at their maximum concentration. The water extract and polysaccharides significantly reduced lipopolysaccharide (LPS)-induced intracellular reactive oxygen species (ROS) content and exhibited a cytoprotective role. Also, these extracts displayed higher matrix metalloproteinase-2 (anti-MMP-2), anti-MMP-9 and anti-tyrosinase activities. Furthermore, the polysaccharides displayed significantly greater inhibitory effect on intracellular ROS and tyrosinase protein expression than α-arbutin and ursolic acid used for the clinical treatment of skin pigmentation. This study also investigated the polysaccharide inhibition of melanin synthesis by repressing the expression of melanocytic lineage-specific transcription factor (MITF) and melanogenic enzymes via modulation of the phosphoinositide 3-kinase (PI3K-Akt-mTOR) and β-catenin pathways. The overall results indicate that L. japonicus is a promising candidate for anti-pigmentation treatment.

The clinical value of magnetic resonance angiography (MRA), transcranial Doppler ultrasound (TCD) and electroencephalogram (EEG) in the diagnosis of intracranial artery stenosis in ischemic cerebrovascular disease (ICVD) was explored. The clinical data of TCD, MRA and EEG in 98 patients with ICVD were retrospectively analyzed. The clinical value of TCD, MRA combined with EEG in the diagnosis of intracranial artery stenosis in ICVD was analyzed based on the results of digital subtraction angiography (DSA) as the gold standard. A total of 98 patients were diagnosed with 27 cases of normal intracranial artery, 32 cases of mild stenosis, 25 cases of moderate stenosis, and 14 cases of severe stenosis DSA examination. TCD was detected in 23 cases of normal, 25 cases of mild stenosis, 23 cases of moderate stenosis and 13 cases of severe stenosis. MRA showed 24 cases of normal, 28 cases of mild stenosis, 21 cases of moderate stenosis and 12 cases of severe stenosis. EEG examination detected that there were 25 cases of normal, 24 cases of mild stenosis, 22 cases of moderate stenosis, and 14 cases of severe stenosis. Sensitivity, accuracy and negative predictive value had significant differences between combined diagnosis and single diagnosis. The combined detection of TCD, MRA and EEG is consistent with DSA detection in the diagnosis of ICVD intracranial artery stenosis.

Background: The clinical efficacy of cancellous screws (CS) and femoral neck system (FNS) internal fixation in the treatment of femoral neck fracture (FNF) was compared in this research.
Methods: Ninety-six patients with FNF were enrolled in this study. There were 47 cases in FNS group and 49 cases in CS group. The data of operation time, intraoperative bleeding, bed days and complication were recorded. Patients' joint function was evaluated by Harris score. At the last follow-up, the cost-effectiveness analysis of the two methods was analyzed according to the total cost of patients and hip function.
Results: The intraoperative blood loss of the patients in FNS group was significantly higher than that in the CS group, but the length of hospitalization was significantly lower. There was no significant difference in the incidence of complications between FNS and CS. The joint function of FNS group was significantly better than that of CS group at 3 months after operation. There was no significant difference in Harris score between the two groups at the last follow-up. The overall medical cost of FNS group in 1 year is high. Cost-effectiveness analysis showed that compared with CS group, FNS group needs to pay 5761.1 yuan more for each additional Harris score.
Conclusion: FNS and CS internal fixation can achieve satisfactory clinical results in the treatment of FNF. FNS treatment is helpful to the early functional recovery of patients, but the overall medical cost is high.

Maintaining erectile function is an important quality of life issue for patients with localized prostate cancer treated with robotic-assisted radical prostatectomy (RARP). However, most existing studies are retrospective and inherently weak and cannot conclude which NS approach is most effective in restoring function in patients. We therefore performed a consistent and objective assessment of sexual function outcomes in RARP using different nerve-sparing methods to optimize postoperative outcomes. A systematic review and meta-analysis was performed based on PRISMA and STROBE statement criteria. Statistical analysis was performed using StataMP software version 14. The Newcastle-Ottawa scale was used to assess the risk of bias. This single-arm meta-analysis included 3 randomized controlled trials and 14 cohort studies with a total of 3756 patients. Our meta-analysis found that patients had the highest efficiency rate of 0.86 (0.78, 0.93) after the NS technique using the retrograde method. Overall, there are significant differences between RARP NS techniques and outcomes, and the ideal technical strategy to optimize outcomes remains controversial. However, there is consensus on the importance of careful separation, dissection of the NVB, reduction of traction and thermal injury, and preservation of the fascia around the prostate. We still need more well-designed randomized controlled trials with videos describing the details of the different surgical techniques before they can be replicated.

To investigate the value of bracketless invisible orthodontic treatment combined with restoration in patients with anterior esthetic restoration. Sixty-two patients requiring esthetic restoration of anterior teeth admitted to our hospital from May 2019 to August 2022 were selected and divided into observation group and control group according to random number table, with 31 cases in each group. The patients in the control group were treated with repair, while patients in the observation group were treated with bracketless invisible correction treatment combined with repair. The patients in both groups were treated with repair. After 2 weeks, dental esthetics, periodontal index related parameters, patients' esthetic acceptance of restorations, and satisfaction were compared. The aesthetic restoration effect of teeth in the observation group was significantly better than that in the control group after treatment, and the difference was statistically significant (P < 0.05). There was no significant difference in GI, SPD, PLI, and SBI levels before and after treatment between the two groups (P > 0.05). The acceptance rate of prosthesis aesthetics in the observation group was 100.00%, which was significantly higher than that in the control group (83.87%), and the difference was statistically significant (P < 0.05). The satisfaction scores of restoration color, shape and coordination with adjacent teeth in the observation group were higher than those in the control group, and the differences were statistically significant (P < 0.05). Compared with simple restorative treatment, combined with bracketless invisible orthodontic treatment helps to further improve the esthetic restoration effect of anterior teeth, has less impact on the periodontal health of patients, and has higher patient acceptance and satisfaction.

Hepatocellular carcinoma (HCC) is identified as a common cancer type across the world and needs novel and efficient treatment. Tripterine, a well-known compound, exerts suppressive role in HCC development. However, the related molecular mechanism of tripterine in HCC remains unclear. The expression of MBNL1-AS1in HCC tissues and cells was measured via qRT-PCR assay. MTT assay was employed to estimate cell viability. Besides, cell migration as well as invasion was determined through transwell assay. Additionally, the binding ability of miR-708-5p and MBNL1-AS1or HK2 was proved by starBase database and luciferase reporter gene assay. Moreover, the HK2 level was detected by immunoblotting. MBNL1-AS1 was reduced in HCC tissues and cells. Overexpression of MBNL1-AS1 decreased the sensitivity of HCC cells to tripterine while MBNL1-AS1 silence played opposite effect. In addition, miR-708-5p was the target of MBNL1-AS1 and was down-regulated through MBNL1-AS1 in HCC cells. Moreover, miR-708-5p suppressed glycolysis rate and reduced the expression of vital glycolytic enzyme (HK2, LDHA and PKM2) in HCC cells. Furthermore, miR-708-5p reduced HK2 expression by binding to it directly. In this investigation, we proved that LncRNA MBNL1-AS1 increased the tripterine resistance of HCC cells at least partly by mediating miR-708-5p-related glycolysis. These findings revealed a potent therapeutic target for the treatment of HCC.

The long non-coding RNA (LncRNA) X-inactive specific transcript (XIST) regulates the biological process of osteoclasts and the process of related diseases. This study was attempted to investigate the mechanism of LncRNA XIST acting in osteoclast formation and orthodontic induced inflammatory root resorption (OIIRR). The compression force (CF) -induced cell model and the orthodontic tooth movement (OTM) rat model were designed and established in this study. The expression of LncRNA XIST, miR-130b-3p, phosphatase and tensin homolog deleted on chromosome 10 (PTEN) as well as osteoclast related marker genes and inflammatory factors level were measured in this study. The interaction among LncRNA XIST, microRNA-130b-3p (miR-130b-3p) and PTEN were researched through luciferase activity and western blot assay. Pathological sections were used to analyze root resorption and osteoclast formation. The OTM rat model was successfully constructed, which was characterized by increased tooth spacing and increased root resorption pits. PTEN and LncRNA XIST was overexpressed in OTM group. Mechanism analysis showed that the overexpression of LncRNA XIST enhanced the PTEN level by sponging miR-130b-3p. The overexpression of LncRNA XIST increased the secretion of inflammatory factors and positive osteoclasts number, but inhibited the differentiation of osteoclasts by sponging miR-130b-3p and promoting the level of PTEN. This finding demonstrates that LncRNA XIST regulates osteoclast formation and aggravated OIIRR through miR-130b-3p/PTEN axis, suggesting that LncRNA XIST may be used as potential targets for OIIRR therapy.

Object: To analyze the clinical effect of external application of self-made Chinese medicine liquid to prevent phlebitis caused by intravenous indwelling needle in our hospital.
Methods: According to different nursing strategies, 600 patients were divided into control group and observation group. The observation group received external application of self-made Chinese medicine liquid in our hospital; the control group was treated with wet compress with 50% magnesium sulfate solution. Then the success rate of indwelling needle puncture, the incidence of phlebitis, the incidence of pain during intravenous drip and the patients' satisfaction with the effect of phlebitis prevention and treatment were compared between the two groups. Then, the success rate of indwelling needle puncture, the incidence of phlebitis, the incidence of pain during intravenous drip and the patients' satisfaction with the effect of phlebitis prevention and treatment were compared between the two groups.
Results: The success rate of one-time puncture was 93.33% in the observation group and 81.67% in the control group. Observation group average indwelling catheter time for (3.89±2.37) days, significantly higher than that of control group (3.00±1.82) days; the incidence of phlebitis in the observation group was 27.33%, lower than that in the control group (43.00%). The satisfaction rate of observation group was 94.00%, significantly higher than that of control group (80.00%). All the differences were statistically significant (P<0.05).
Conclusion: Compared with magnesium sulfate wet compress, external application of self-made Chinese medicine liquid in our hospital can effectively prevent the occurrence of phlebitis caused by indwelling needle, especially relieve pain during intravenous drip in patients, with high patient satisfaction.

To investigate the image computer analysis of abnormally proliferating transformed cells of gastric mucosa and its clinical significance. The pathological pictures of gastric adenomatous polyp cells, abnormally proliferating altered cells, and tubular adenocarcinoma cells in the stomach mucosa were assessed by image computer method on a total of 96 gastroscopic biopsy and ESD resection specimens. The data of cytoplasmic area, nuclear area, nuclear-cytoplasmic ratio, nuclear factor and N-heterotypic index of gastric adenomatous polyps, abnormal proliferative transformation and gastric intramucosal tubular adenocarcinoma were collected, and the mean, standard deviation and variance were calculated respectively. Standard Error, Maximum, Minimum Parameters and Statistical Structure. There were substantial discrepancies between gastric mucosal gastric adenomatous polyp cells and gastric mucosal abnormally proliferating transformed cells, according to the five data in the abnormal cells in the stomach mucosal proliferation area (p < 0.01); There was no significant difference between cells (p > 0.05). Computer analysis of cell images can provide quantitative values for the pathological diagnosis of gastric adenomatous polyp cells, abnormally proliferating transformed cells and tubular adenocarcinoma cells in the gastric mucosa, especially the degree of atypical proliferation.
