A method for fast genomic DNA extraction from saliva and its application 唾液基因组DNA快速提取方法及其应用

Objective To explore a fast,effective and noninvasive method of genomic DNA extraction.Methods We compared the genomic DNAs extracted by two DNA extraction methods of potassium iodide(KI) and Kit from saliva(fresh and placed a week at room temperature),and PCR amplifications were done for TP53 and PRB-3 genes from their extracted DNA.Ninety-nine healthy children and adults were recruited and their saliva samples were collected to detect the stability of DNA extracted by KI.Results High quality genomic DNAs were extracted by both methods from the fresh saliva.The DNA yield based on KI was(1.91 ± 0.15) μg,with the D260/ D280 being 1.99 ± 0.05,and that of Kit was(2.64 ± 0.34) μg,with the D260/ D280 being 1.81 ± 0.02.Although the extracted DNAs had obvious degradation from saliva samples placed for a week at room temperature,the expectant DNA fragments were successfully amplified by PCR for TP53 and PRB-3 genes using these extracted DNAs.Although the genomic DNAs from saliva samples of 99 healthy volunteers by the method of KI showed individual difference(DNA yield was [1.89 ± 0.46] μg),the DNA quality was stable and reliable(D260/ D280 was 1.96 ± 0.10).Conclusion KI method for extracting genomic DNA from saliva is not only cheap and highly effective,but also noninvasive,making it suitable for large-scale epidemiological studies.

Biological functions of zinc finger protein ZBTB2:recent advance 锌指蛋白ZBTB2的生物学功能研究进展

Zinc finger protein ZBTB20 is a novel member of the ZBTB zinc finger protein family and shares closest homology with BCL-6 and PLZF.Recent studies on genetically modified mice and patient cohort have revealed that ZBTB20 plays indispensable roles in multiple organs and systems,including the nervous system,liver,pancreatic islets,and immune system.ZBTB20 is an essential regulatory factor in a variety of physiological processes,such as ontogenesis,pain sensation,learning and memory,as well as glucose and lipid metabolism.Meanwhile,recent researches have also showed a close relationship between the abnormality or dysfunction of ZBTB20 and pathophysiological procedures of diseases,such as maldevelopment,mental retardation,tumors and metabolic disorders.As a key repressor governing AFP gene transcription,ZBTB20 exerts a great influence on the gene inactivation of liver AFP after birth and is closely related to the prognosis of hepatocellular carcinogenesis.In humans,the deletion of ZBTB20 is detected in diverse tumors and its point mutation leads to Primrose syndrome.This review summarizes the current knowledge concerning the biological functions of ZBTB20.

Red blood cell distribution width is elevated in patients with Graves' ophthalmopathy Graves眼病患者红细胞分布宽度增高

Objective To study the relationship between red blood cell distribution width(RDW)and thyroid associated ophthalmopathy(TAO).Methods Fifty-two patients with Graves' disease(GD,18 males and 34 females,39cases of GD patients with Graves' ophthalmopathy[GO])and 22 healthy controls(8 males and 14females)were recruited in the present study.The RDW and thyroid functions were measured in all participants.Ophthalmological examinations including Hertel's exophthalmometer readings(HER)and clinical activity score(CAS)evaluation were obtained in GD patients,and the results were compared between different groups.Results Thyroid functions were significantly different between GD patients and healthy controls(P0.05),while they were not between GD patients with or without GO;the HER level was significantly higher in those with GO compared with those without GO(P0.05).The RDW value in patients with GD was significantly higher than that in the healthy controls(P0.05).For GD patients,RDW increased with the increase of CAS,and those with GO had significantly higher RDW than those without GO(P0.05).Conclusion RDW is associated with the state of the GD patients,indicating that RDW may provide evidence for assessing the severity of GO.

Psychological experience of Chinese nurses during China's anti-Ebola aid in Liberia: a qualitative interview 中国护士在利比里亚抗埃博拉援助中的心理体验:质性访谈

Objective To understand the psychological experience of Chinese nurses working in Liberia during the aid to fight against Ebola virus disease,so as to lay a foundation for future similar missions.Methods We conducted a qualitative interview study with 11 Chinese nurses who helped to fight against Ebola virus disease in Liberia by semi-structured interview,and then the data collected were analyzed.Results Analysis showed that the psychological experience of Chinese nurses in Liberia could be divided into 4stages.Stage one was from being informed of the aid mission to going aboard to Liberia,with the psychological experience being panic and having anxiety.Stage two was the first 2weeks of working in Chinese Ebola Treatment Unit(ETU)in Liberia,with the psychological experience being lost,sad and having anxiety.Stage three was from the third week of working in ETU to before going back to China,with the psychological experience being attached and emotional.And stage four was the quarantine period in China,with the psychological experience being prevailing loneliness.Conclusion Efforts should be made to support,to train and to guide the nurses who work in situations like Ebola according to their psychology in different stages.And the nurses should have an active attitude,improve understanding of Ebola disease,and ensure safe selfprotection during work.

Nursing experience of patients with Ebola virus disease in Chinese Ebola Treatment Unit in Liberia 中国驻利比里亚埃博拉治疗中心埃博拉患者护理体会

Objective To discuss the nursing interventions of patients with confirmed Ebola virus disease.Methods and Results We retrospectively analyzed the nursing method and experience of 4confirmed cases with Ebola virus disease in Chinese Ebola Treatment Unit,which was established by Chinese People's Liberation Army medical team in Liberia.Following the strict personal protective process,we provided timely symptomatic care,seamless psychological care,carefully basic care and comprehensive health education for the patients.We also timely summarized our experience of nursing interventions.Through dedicated treatment and nursing,despite 1case died,3cases were cured and discharged.Conclusion Since there have been no specific and effective medications for Ebola virus disease by now,timely and correct symptomatic treatment and dedicated nursing with high efficiency are of great significance for patients to fight against with Ebola virus disease and regain their health.
目的探讨埃博拉确诊患者的护理干预措施。方法与结果回顾性分析中国人民解放军驻利比里亚医疗队埃博拉治疗室收治的4例埃博拉病毒病确诊病例的护理方法和经验。我们遵循严格的个人防护流程,为患者提供及时的症状护理、无缝的心理护理、细致的基础护理和全面的健康教育。及时总结护理干预经验。经过精心的治疗和护理,1例死亡,3例治愈出院。结论由于目前尚无针对埃博拉病毒病的特异性有效药物,及时、正确的对症治疗和专门、高效的护理对患者抗击埃博拉病毒病、恢复健康具有重要意义。

Objective To explore the mutations of hepatocellular carcinoma(HCC)-related hepatitis B virus(HBV)during mother-to-child transmission,so as to provide theoretic evidence for prophylaxis of HCC from the very beginning.Methods A total of 413 HBsAg-positive mothers and their newborns were enrolled in this study.Serum HBV DNA levels in maternal peripheral blood and cord blood of the newborns were measured using real-time quantitative PCR.Nested PCR together with cloning and sequencing methods were applied to examine the HCC-related HBV mutations in the preS and basal core promoter regions of HBV genome.All the newborns received standard HBV vaccination.Of the 413 newborns,104were successfully followed-up 7 months after birth,and the HBV mutations were examined if their circulating HBV DNA was detectable.Results Of the 413 newborns,41(9.9%)had HBV DNA level103 copies/mL in their cord blood.Four(3.8%)of the 104 newborns who were successfully followed up had circulating HBV DNA level103 copies/mL 7months after birth.Compared to mothers without HBV trans-placental transmission,those with HBV trans-placental transmission had no increase in HBV mutations in the basal core promoter region.However,the viral mutations containing T2898G/C,C3000 T,C3116T,T31 C,and T52 Cin the preS region of HBV subgenotype C2 significantly increased the risk of HBV trans-placental transmission(P0.05).The frequencies of the HCC-related mutations in the preS and basal core promoter regions of HBV genome were not significantly different between maternal peripheral blood and the cord blood of the newborns.Importantly,the HCC-related mutations were rarely found in the HBV-positive infants at 7months after birth.Conclusion The HBV mutations in the preS region of HBV subgenotype C2 may affect the trans-placental transmission of HBV.However,the quasispecies of HCC-related HBV mutants have no advantage in causing chronic HBV infection in infants.The HBV mutants which can promote HCC are selected during the long term chronic infection.
