EFFECT OF ALA-GLN FEEDING ON GROWTH, ANTIOXIDANT LEVEL,AND IMMUNITY OF CYPRINUS CARPIO VAR. JIAN ala-gln饲喂对鲤生长、抗氧化水平及免疫的影响

This study was carried out using healthy Cyprinus with the initial body weight of(33.52±0.17) g in a single recirculating system for 8 weeks.Diets with equal levels of protein(35 g/kg) and energy(17 kJ/g) were administered to the Cyprinus at concentrations of 0.0 %(control diet) and 0.5%(trial diet) Ala-Gln of dietary dry matter in five different feeding modes: continuous 8 weeks of control diet(group I or control),trial diet at an interval of 2 weeks(group II),trial diet at 4 weeks and then control diet at 4 weeks(group III),the contrary to group Ⅲ(group IV) and continuous 8-week trial diet(group V).The trail was conducted to investigate the effects of different feeding mode of Ala-Gln on growth,immunity and antioxidant in Cyprinus.Our results indicated that growth rates under all different feeding modes of Ala-Gln were significantly higher than that of the control group(P0.05).Growth rate of Ⅱ was higher than that of Ⅲ,Ⅳ,Ⅴgroups(P0.05),and growth rate of Ⅲ was higher than that of Ⅴgroup(P0.05).Serum SOD of the Ⅳ and Ⅴgroups were significantly higher than the control group(P0.05),and serum SOD of the Ⅴgroup was significantly higher than those of Ⅱ and Ⅲ groups(P0.05).GSH-Px in hepatopancreas of different feeding modes were significantly higher than that of the control group(P0.05),Serum GSH-Px of Ⅳ and Ⅴgroups were higher than that of the control group(P0.05).LZM in the head kidneys of the Ala-Gln groups were higher than the control group,only group Ⅱ showed no significant difference(P0.05),LZM in the spleen of the groups Ⅳ and Ⅴ were significantly higher than that of the control group(P0.05).In the current experiment,we concluded that adding Ala-Gln could improve the growth,immunity and antioxidant levels of Cyprinus.There were significant differences of different feeding modes.Given economical efficiency and practicability,feeding mode with 2-week intervals were suggested in the practical aquaculture of Cyprinus.
本研究选用初始体重为(33.52±0.17)g的健康鲤,在单一循环系统中饲养8周。以饲粮干物质Ala-Gln浓度为0.0%(对照饲粮)和0.5%(试验饲粮)的5种不同饲养模式,分别饲喂蛋白质(35 g/kg)和能量(17 kJ/g)水平相等的饲粮。连续饲喂8周的对照饲粮(ⅰ组或对照组),每隔2周饲喂一次试验饲粮(ⅱ组),每隔4周饲喂一次试验饲粮,然后每隔4周饲喂一次对照饲粮(Ⅲ组),与Ⅲ组(ⅳ组)和连续饲喂8周试验饲粮(ⅴ组)相反。本试验旨在探讨不同饲喂方式Ala-Gln对鲤生长、免疫和抗氧化能力的影响。结果表明,不同饲养方式下Ala-Gln的生长率均显著高于对照组(P0.05)。Ⅱ组的生长率高于Ⅲ、Ⅳ、Ⅴ组(P0.05),Ⅲ组的生长率高于Ⅴ组(P0.05)。Ⅳ组和Ⅴ组血清SOD显著高于对照组(P0.05),Ⅴ组血清SOD显著高于Ⅱ组和Ⅲ组(P0.05)。不同饲养方式肝胰脏GSH-Px均显著高于对照组(P0.05),Ⅳ和Ⅴ组血清GSH-Px均高于对照组(P0.05)。Ala-Gln组大鼠头肾LZM均高于对照组,只有Ⅱ组差异无统计学意义(P0.05),Ⅳ、Ⅴ组大鼠脾脏LZM均显著高于对照组(P0.05)。在本试验中,我们认为添加Ala-Gln可以提高鲤的生长、免疫和抗氧化水平。不同饲养方式之间存在显著差异。考虑到经济效益和实用性,建议在实际养殖中采用每2周饲喂一次的模式。

GROWTH AND POPULATION STRUCTURE VARIATIONS IN CRUCIAN CARP (CARASSIUS AURATUS) BETWEEN A MACROPHYTIC LAKE AND AN ALGAL LAKE AT THE MIDDLE PART OF YANGTZE RIVER BASIN 长江中游大型植物湖与藻类湖间鲫鱼生长及种群结构的变化

Human activities have profoundly affected the environment and population structures of fishes in Yangtze lakes.In order to evaluate the effects of environmental degradation on fish population structure,a comparative study on population characteristics of crucian carp Carassius auratus between a eutrophic urban lake(Yuehu Lake) and a macrophytic suburban lake(Niushan Lake),both in Wuhan,was carried out during March to May 2006.Fishes were collected and compared from the two different lakes,and the water quality was measured at the same time.Results showed that:(1) pH,conductivity,TN-N,TP-P and Chl.a of Yuehu Lake were higher than those of Niushan Lake.The numbers of total fish species and carnivorous fish species in the Yuehu Lake were fewer than those in the Niushan Lake.(2) The relative biomass of crucian carp in Yuehu Lake were higher than that in Niushan Lake,as per the multimesh gillnet catch per unit effort(CPUE) in Yuehu lake was higher.(3) Length distributions of crucian carp in both Niushan and Yuehu Lakes showed typical distributions when tested by kolmogorov-Smirnov,with a length range 63—271 mm(peak at 150 mm) and 72—191 mm(peak at 120 mm),respectively.(4) The crucian carp in Niushan Lake had a more complex age structure with six age groups,while Yuehu Lake had only three,with age 1 representing the largest proportion of 89%.(5) The relationship between weight(W) and total length(L) has been represented by the equation W= aL b,where W=1*10 5 L 3.06(R 2 =0.9893) in the Niushan Lake and W=6×10 6 L 3.22(R 2 =0.9649) in the Yuehu Lake.Analysis of covariance demonstrated that the length-weight relationship of crucian carp are significantly different between the two lakes(P0.01).Meanwhile,the crucian carp in Yuehu Lake showed a growth advantage compared to those in Niushan Lake for fish condition factors of males and females from all of the three length groups in Yuehu Lake were all significantly greater than those in Niushan Lake.(6) As lengths of crucian carps in both lakes were increasing,the relative proportion of males declined;this change for male crucian carp in Yuehu Lake was more acute than in Niushan Lake.

RESEARCH ON THE DEGRADATION OF IVERMECTIN AND ITS ACUTE TOXICITY TO SEVEN AQUATIC ORGANISMS 伊维菌素的降解及其对7种水生生物的急性毒性研究

Ivermectin(IVM) is a widely applied veterinary pharmaceutical which is used in large amount to treat livestock.It has been a potential environmental problem,that the drug often appears intact in the metabolite of livestock,and its transport from soil to surface water via runoff.In natural environment,dissipation is relatively slow,and the residues can cause mortality in a diverse range of invertebrates.So ivermectin may destroy the aquatic ecosystems.To evaluate the potential toxicity of ivermectin to aquatic organisms,we simulated a natural riverway environment when monitoring the degradation speed of it,and then chose seven aquatic organisms which take up different ecological riche to test acute toxicity by national standard metords.The results displayed that the degradation of Ivermectin(IVM) in water environment was slow,and the degradation rate of IVM in sediment was only 28.3% of 70 days’ experiment under 25℃.The results showed no obviously toxicity to Photobacterium phosphoreum and the 96h EC50 to Chlorella vulgaris was 19.80 mg/L,it did not exhibit high toxicity.But the toxicity of IVM to Brachydanio rerio,Gambusia affinis,Carassius auratus,Macrobrachium rosenbergii and Daphnia magna were relatively higher,and date was 40.48 μg/L(96h LC50),34.81 μg/L(96h LC50),13.79 μg/L(96h LC50),7.87 μg/L(96h LC50) and 4.81 ng/L(24h LC50) respectively.IVM would influence the aqua-biology obviously when the drug residue appeared in water environment,so it is necessary to learn more on ivermectin.

MORPHOLOGICAL STUDIES OF OCCLUDED PROCESSES IN THALASSIOSIRA SPECIES FROM CHINA 中国海藻种闭塞过程的形态学研究

Thalassiosira Cleve is a common diatom genus with high species diversity in coastal waters around the world.Till now,nearly one hundred and fifty taxa belonging to this genus have been reported.Thalassiosira is the main contributor to nano-phytoplankton assemblage because cell sizes of most species are below 20 μm.Previously,some Thalassiosira species were considered as good model organisms of centric diatoms and Bacillariophyta.Many studies concerning genetics,sexual reproduction,feeding pressure,impact on egg producing,hatching of zooplankton,and environmental indicator have been performed.On the other hand,some Thalassiosira species are also important bloom-causing organisms.Algal blooms induced by T.curviseriata and T.diporocyclus have been reported in the Changjiang River Estuary and coast of Hainan Province,respectively.Due to very small cell sizes,electron microscopy(EM) is needed for accurate identification.In order to characterize species diversity and distribution of the genus Thalassiosira along Chinese coastal waters,water samples were collected from different locations.Thalassiosira specimens within these samples as well as several clonal strains isolated from natural samples were studied using scanning electron microscopy.All samples were fixed with Lugo’s solution and were concentrated to a final volume about 2—10 mL by settling overnight for several times.For observation under scanning electron microscope,sub-samples were taken and treated in several steps.Firstly,after the same volume of concentrated H2SO4 was added,these samples were boiled in the water bath for about 10—15 minutes to remove organic materials.Secondly,these samples were rinsed with distilled water several times to rid the acid.Then,these samples were coated by gold and thus could be observed,identified,and photographed under scanning electron microscope.The morphology and pattern of processes,including three types,fultoportula,rimoportula and occluded process,are key taxonomic characteristics for Thalassiosira species,even for some pennatae diatoms.Fultoportula and rimoportula are very common structures in all Thalassiosira taxa,while occluded process is just found in no more than 20 taxa from presently known 150 reported Thalassiosira species.The different features between occluded process and other two types of process is that occluded process does not penetrate the valve,so there is no structure of occluded process on the internal valve face.Previous reports about Thalassiosira species with occluded processes from China coast were rare,and almost no EM photos of occluded processes have been provided.In present study,five Thalassiosira species with occluded processes were observed and they were T.cedarkeyensis Prasad,T.eccentrica(Ehrenberg) Cleve,T.lundiana Fryxell,T.punctigera(Castracane) Hasle and T.tubifera Fryxell.Three of them are newly recorded species for China,T.cedarkeyensis,T.punctigera and T.tubifera.The detailed morphological description and habits

ANALYSIS ON THE SPECIES DIVERSITY OF FISHERY ANIMALS IN DAIQUYANG 代曲阳渔业动物物种多样性分析

The Daiquyang locates between the Daishan and Qushan Islands in the Zhoushan Archipelago.Due to its optimal location and suitable environmental characteristics,it is the spawning and feeding ground of Larimichthys crocea of the Daiqu Race,and also of other fishery animals(fish species such as Larimichthys polyactis,Trichiurus japonicus,Pampus argenteus and Engraulis japonicus,crustacean species such as Portunus trituberculatus,Charybdis japonica,Exopalaemon carinicauda,Acetes chinensis and Oratosquilla oratoria,cephalopods such as Sepiella maindroni,Octopus variabilis,Octopus ocellatus and Loligo duvaucelii,and other animal species such as Nassarius variciferus,Sliqua minima,Cantharus cecillei and Moerella irideseens).For a long time,although the Daiquyang plays an important role in marine fisheries in the East China Sea,there have been no studies focused on the species diversity of its fishery animals.In recent years,both the environment and fishery resources in the Daiquyang have been facing new threats arisen from rapid development of harbor industries and a mass of shore reclamation in the Zhoushan Archipelago District,Zhejiang.Therefore it is necessary to study the species diversity of fishery animals in this area.We analyzed fishery animal diversity and the status of fishery resources based on four surveys conducted by bottom trawl,setting gillnet and crab pot from April to November 2007 in the Daiquyang.Three analytical methods including sweep-area method,biodiversity analysis and analysis of variance were used.The results showed that: 1) a total of 98 fishery animals were sampled,including fishes(58 species),crustaceans(32 species),cephalopods(3 species) and other species(6 species),belonging to 53 families of 20 orders,7 classes and 4 phylums.2) In our trawl collections,a total of 68 fishery animals were obtained,including fishes(38 species),crustaceans(23 species),cephalopods(2 species) and other species(5 species),belonging to 42 families of 18 orders,7 classes and 4 phylums.The richness index(D) of fishery animals in April was significantly different from those in August and November(P 0.05).Both the biomass and the number abundance density indices of fishery animals were the highest in August,significantly different from those in May and November(P 0.05).The dominant species were Harpodon nehereus,Coilia mystus,Palaemon gravieri and Alpheus japonicus in April,Harpodon nehereus,Coilia mystus,Exopalaemon annandalei and Charybdis japonica in May,Harpodon nehereu and Portunus trituberculatus in August,Harpodon nehereus,Charybdis japonica and Oratosquilla oratoria in December,and among them,Harpodon nehereus was the dominant species during all study months.Average body weight of Harpodon nehereus in August was significantly different from those in other three months(P 0.05) because of the spawning of the juveniles.For trawl collections,the biological community was mainly composed of warm temperature fish species,eurythermal and euryhaline
