Identification by in situ hybridization of IFN-β-producing murine macrophages obtained from high and low interferon producers 高干扰素和低干扰素产生者产生IFN-β的小鼠巨噬细胞的原位杂交鉴定

Resident peritoneal macrophages obtained from If-1h and If-11 mice were induced in vitro with Newcastle disease virus. Then, 3, 5 and 7 h after induction, the cells were fixed and analysed for the presence of Mu IFN-β mRNA by in situ hybridization, using a 35S-labelled Mu IFN-β cDNA as a probe. The number of cells that were scored positive was the same in cultures derived from high responder If-1h and from low responder If-11 mice, and reached 100% of the cells present in the cultures. This result highly suggests that low responder mice have the same number of IFN-producing cells as high responders, and that the difference in production is not due to an increased number of producer cells in high responders.

La production d'interféron a été induite in vitro par le virus de la maladie de Newcastle dans des macrophages prélevés dans la cavité péritonéale de souris If-1h et If-11. La mise en culture d'adénovirus détectés dans les selles de patients par un examen en microscopie électronique, a permis de mettre en évidence l'effet favorisant, voire nécessaire, de la trypsine dans le milieu de culture pour le développement de certains de ces adénovirus; outre le fait surprenant de noter ces effets pour des sérotypes croissant en général aisément sur cellules KB, on a observé l'amélioration ou la possibilité de la culture d'un nouveau sérotype (candidat adénovirus 42) et de 4 adénovirus non identifiés, grâce à la présence de trypsine dans le milieu.

Evaluation of herpes simplex virus susceptibility to acyclovir using an enzyme-linked immunosorbent assay 用酶联免疫吸附试验评价单纯疱疹病毒对阿昔洛韦的易感性

An in situ ELISA was performed directly on the adherent cell monolayer in order to determine the susceptibility of herpes simplex virus isolates to acyclovir. Various fixation procedures and antisera conjugated to different enzymes were tested. The use of glutaraldehyde for fixation and betagalactosidase as a labelling enzyme was shown to give the best results. As with other currently used assays, 50% inhibitory doses were subject to an inoculum effect. The data obtained indicate that this assay is suitable for routine determination of herpes simplex virus susceptibility to antiviral drugs.

Pour tester la sensibilité du virus de l'herpes simplex à l'acyclovir, nous avons réalisé un test ELISA directement sur monocouche cellulaire. Différentes méthodes de fixation et différents enzymes conjugués aux immunsérums ont été essayés. Les meilleurs résultats ont été obtenus avec le glutaraldéhyde comme fixateur et avec la beta-galactosidase comme enzyme. Comme pour les autres tests, les doses inhibitrices 50% varient en fonction de l'inoculum. Les résultats obtenus montrent que ce test est simple et rapide et qu'il est adapté à une détermination de routine de la sensibilité des virus herpes simplex aux antiviraux.
