l-threo-β-Hydroxyleucine has been isolated from Deutzia gracilis by ion-exchange resin chromatography, and by preparative paper chromatography. The substance is reduced to leucine by hydroiodic acid and offers the characteristic reaction of β-hydroxyamino acids with periodic acid. Alkaline decomposition confirms the β position of the alcohol function. The natural substance and the dl-threo-β-hydroxyleucine are inseparable by paper chromatography, and the study of their nuclear-magnetic-resonance spectra brings supplementary arguments in favour of the threo form. The natural substance has the same infrared spectrum as synthetic threo-β-hydroxyleucine.

The formation of testosterone in vivo has been investigated in anesthetized dogs by infusing radioactive steroids via the left spermatic artery and isolating radioactive testosterone in venous blood obtained from the left spermatic vein. The data from such experiments indicate that the formation of 17α-hydroxypregnenolone and the metabolism of this steroid to testosterone is a major route in the biosynthesis in vivo of testosterone from pregnenolone by the canine testis.

1. When an equal amount of [4−14C]progesterone and 17α-hydroxy[7α-3H]pregnenolone were infused at a constant rate by the spermatic artery, the concentration of 3H in isolated spermatic vein testosterone was from 3–8 times that of 14C.

2. When [7α-3H]prenenolone was infused by the left spermatic artery for 5 min, more [3H]17α-hydroxypregnenolone than [3H]progesterone was found in the left testis at the end of this infusion.

Under incubation conditions including deprivation of nitrogen sources, deprivation of lysine, inhibition by oxamycin and inhibition by penicillin, the accumulation of UDP-N-acetylamino sugar derivatives in Staphylococcus aureus cells, with time, has been studied. UDP-N-acetylglucosamine and UDP-N-acetylglucosamine-3-O-lactic ether accumulated in cells which were incubated in amino acid-free medium. During the initial phase of incubation there was a rapid rise in the appearance of UDP-N-acetylglucosamine followed later by a decrease of this nucleotide; concomitant accumulation of UDP-N-acetylglucosamine-3-O-lactic ether and UDP-N-acetylglucosamine-3-O-lactyl-peptides was observed under every condition which induced UDP-N-acetylglycosamine-3-O-lactyl-peptide accumulation. The relationship of the observed accumulation to the pathway of cell-wall mucopeptide synthesis is discussed.
