Simple formulas (單方) of muscle section in Donguibogam (東醫寶鑑) have long been prescribed for strengthening muscle and/or prevention of age-related muscle loss. However, biological activity and mechanisms by which they influence myoblast differentiation have not been studied. Therefore, in this study, we evaluated the effects of 14 simple formulas on myoblast differentiation in C2C12 myoblast cells under non-cytotoxic (0.5 ㎎/㎖) conditions. C2C12 cells were treated with water extracts of simple formulas for 72 h, and RT-PCR was performed to determine the gene expression levels of myogenic regulatory factors (MRFs), including myoD, myogenin, MRF4, myf5, and insulin like growth factor-1 (IGF-1). Treatment with Colocasiae Rhizoma (CR), Pini Semen (PS), and Sesami Semen (SS) resulted in a significant increase in expression of myogenin in C2C12 cells. Treatment with Allii Macrostemi Bulbus (AM), Colocasiae Rhizoma (CR), and Pini Semen (PS) also resulted in increased expression of MRF4 in C2C12 cells. In addition, enhanced expression of IGF-1 was observed in treatment with Eucommiae cortex (EC), Dioscoreae Rhizoma (DR), Colocasiae Rhizoma (CR), Pini Semen (PS), and Sesami Semen (SS) in C2C12 cells. These results indicate that simple formulas of muscle section in Donguibogam could potentially enhance myoblast differentiation at least in part via increasing expression of myogenin, and/or MRF4 and/or IGF-1.

The purpose of this study is to investigate the modulatory effect of oroxylin A on hydrogen peroxide production in TM4 mouse testis sertoli cells induced by the synthetic analog of double-stranded RNA [polyinosinic-polycytidylic acid]. Hydrogen peroxide production was measured by dihydrorhodamine 123 (DHR) assay. Oroxylin A significantly inhibited the polyinosinic-polycytidylic acid (PIC)-induced production of hydrogen peroxide for 0.5, 2, 12, 18, and 24 hr incubation at the concentrations of 5, 10, 25, and 50 μM in TM4 (P < 0.05) in dose dependent manner. These results suggest that oroxylin A has a protective effect against PIC-induced cellular toxicity with its inhibition of hydrogen peroxide production in PIC-induced sertoli cells.

The purpose of this study was to validate the Sasang Digestive function Inventory (SDFI) and Sasang Personality Questionnaire (SPQ) which measures the pathophysiolgoical symptoms and temperament of each Sasang types. The SDFI, SPQ along with Questionnaire for Sasang Constitution Classification II (QSCCII) were measured with 191 College students. The correlation coefficient was measured with Pearson correlation between SDFI and SPQ. The SDFI and SPQ subscale score of each Sasang constitution groups based on QSCCII were compared using analysis of variance and profile analysis. The SDFI and SPQ total score of So-Eum Sasang type were significantly (p 0.05) between SDFI and SPQ total score, however the SDFI-Digestion scale showed significant correlation with SPQ subscales. We found that the SDFI and SPQ can measure the Sasang type-specific pathophysiological symptoms and temperaments, and it would be useful for the clinical application and interdisciplinary research when combined together.

Amyloid-β protein (Aβ) is a pathological component of Alzheimer’s disease (AD) by participating in the senile plaque formation in the patient’s brain. Although the exact mechanism of Aβ toxicity is not fully elucidated, it is considered to be closely related to its fibrillation process. For prevention of AD, recent studies have suggested various small molecules which inhibit Aβ fibrillation. In this report, β-asarone found in acorus plant has been investigated as an anti-amyloid molecule. β-Asarone was demonstrated to prevent in vitro fibrillation of Aβ by inducing the oligomer formation that obviously decreased cytotoxicity. Therefore, β-asarone could be suggested as an inhibitory agent of Aβ fibrillation and toxicity, which would help us not only to understand underlying principle of amyloidogenesis mechanism but also to develop a controlling strategy toward AD.

This study aimed at examining the anti-inflammatory effects of Scutellariae Radix & Lonicerae Caulis water extract(SC). RAW 264.7 mouse macrophage cells were treated with 25∼200μg/ml SC for 24 hours. Cell viability was then measured using MTT assays. The nitric oxide(NO) production and the creation of several cytokines in LPS-stimulated RAW 264.7 cells were investigated. SC inhibited significantly increasing the production of NO in LPS-induced RAW 264.7 cell at the density of 25, 50 and 200 μg/ml. SC inhibited significantly the TNF-α of the RAW 264.7 cell induced by LPS at the density of 50μg/ml. SC inhibited significantly the MIP-1α of the RAW 264.7 cell induced by LPS at the density of 25, 50 and 100μg/ml. SC inhibited significantly the MIP-1β, MIP-2 at the density of 50, 100μg/ml in the RAW 264.7 cell increased by LPS, respectively. SC did not affect the production levels of VEGF in RAW 264.7 cell. As a result, SC significantly inhibited the inductions of MIP-1α, MIP-1β, MIP-2 and NO in LPS-induced RAW 264.7 cell without causing the toxicity. These results signify that SC has anti-inflammatory effects on controlling the over inflammatory reaction on the RAW 264.7 cell.
