Composition of phenolic compounds and antioxidant activity of different varieties of Mirabelle plum (P. domestica L. ssp syrica var cerea). 不同品种紫梅酚类化合物组成及抗氧化活性研究

The aim of this study was to evaluate the impact of Mirabelle plum variety, the part of the fruit (peel or pulp) and soil quality on composition of phenolic compounds and antioxidant activity of these fruits. Two varieties of Mirabelle plum fruits (yellow and red) were harvested in 2 places in the North of Poland - Lubawa and Debiniec - that varied in soil quality. Fruit peel and flesh were separated and tested individually. The composition of phenolic compounds was characterized using HPLC-DAD-MS, total antioxidant activity was assessed by spectrophotometric tests (ABTS, DPPH, FC). Antioxidant profiles were obtained using TLC and HPLC with post-column derivatization. All considered factors turned out to have strong impact on composition of phenolic compounds as well as on antioxidant activity. The highest content of phenolic compounds was observed in red Mirabelle plum peel from Debiniec (29.8 mg/g d.w.). Peels showed three to ten times higher content of phenolic compounds than pulp from corresponding fruits. Red fruits showed higher antioxidant activity than yellow fruits, because of their anthocyanins content. Both red and yellow varieties harvested in Debiniec turned out to be richer source of phenolic compounds than the same varieties harvested from Lubawa.

Natural nutraceuticals - do we know what we are using? 天然营养保健品-我们知道我们在使用什么吗?

In the current „industrial“ world, the „natural“ nutraceuticals of plant origin are increasingly popular as dietary supplements, herbal remedies or fitness enhancers, preferred by many in comparison with the „chemical drugs“, produced by the pharmaceutical industry. Alas, one well-known disadvantage of the drugs and extracts based on natural products is their potential variability, which may make any of their effects irreproducible or unreliable. We demonstrate this variability on the preparations based on the sea-buckthorn (Hippopae rhamnoides), which is a rather popular source of unsatorated fatty acids, rich in vitamins, containing also a plethora of polyphenolics and other potentially biologically active coumpounds. Analytical data from literature and from various commercially available preparates of „oil from berries“ were compared. Both the macroscopic parameters of the preparations, and the details of their composition vary rather widely, reflecting not only biological (and geographic) variablity of the species, but possibly also the influence of the used extraction method and of the final adjustment in the commercial preparate. This variablity is reflected also in the differing stability during storage and it may quite strongly influence the potential biological effects and benefits connected with the use ot the sea-buckthorn-based nutraceuticals.

IMPACT OF A BLACKBERRY EXTRACT AND SINGLE ANTHOCYANINS ON THE INFLAMMATORY RESPONSE OF HUMAN MACROPHAGE THP-1 CELLS 黑莓提取物和单一花青素对人巨噬细胞thp-1细胞炎症反应的影响

A diet rich in anthocyanins is considered to be beneficial for health. Thus, investigations on anthocyanin-rich extracts and single constituents are of great interest. We investigated a blackberry extract, containing a spectrum of anthocyanins, in comparison to two sub-fractions comprising cyanidin-3-(6''-dioxalylglucoside and cyanidin-3-(malonylglucoside), respectively, regarding their impact on inflammation-related cytokine expression and secretion in human macrophages. Relative gene transcription and secretion of IL-6 and TNF-alpha were measured using quantitative real time PCR and human cytokine bead-based multiplex immunoassay, respectively. Human THP-1 monocytes were differentiated with phorbol-12-myristate-13-acetate into macrophages which were pre-incubated with physiologically relevant concentrations of blackberry extract or single compounds. Subsequent stimulation with lipopolysaccharides from E.coli induced gene transcription and cytokine release mimicking the inflammatory response. Cyanidin-3-(malonylglucoside) had no impact on LPS-induced IL6 and TNF-alpha expression. In contrast, the blackberry extract and cyanidin-3-(6''-dioxalylglucoside) caused further stimulatory effects for the transcription and secretion of IL-6 at higher concentrations ≥0.05 µg/ml. The secretion of TNF-alpha was increased as well by both, yet with no impact on the transcript level after 3 h of incubation. The results clearly show that single as well as combinatory effects might be of relevance for the immune-stimulatory impact of the blackberry extract.

Antioxidant activity and polyphenols stability in ice cream enriched with cocoa bean shell during simulated in vitro gastrointestinal digestion 可可豆壳冰淇淋在模拟体外胃肠消化过程中的抗氧化活性和多酚稳定性

Cocoa bean shells (CBS) is one of the main by-products of the cocoa roasting process, which represents 12% of total weight after the husking and grinding1. CBS is a potential source of dietary fibre and polyphenols recognized as dietary factors responsible for potential beneficial effects on human health. The aim of this study was to evaluate the stability and bioaccessibility of phenolic compounds and the changes on the antioxidant activity of enriched ice cream during gastrointestinal digestion. CBS flour was added to ice cream at different concentration (2%, 4%, 6% and 8%) and the total phenolic content (TPC), total flavonoids, tannins and radical scavenging activity  (DPPH) were evaluated during in vitro digestion at different steps of digestion (oral, gastric and duodenal). After in vitro digestion total flavonoids and tannins increase slightly and TPC increase up to 50% compared to control before digestion. The radical scavenging activity of ice cream containing cocoa bean shell increases up to 25% compared to ice cream before digestion. The results suggested that CBS may be a promising functional ingredient to producing highly nutritive value ice cream minimizing the volume of agro-industrial wastes.

IMPACT OF ELECTROTECHNOLOGIES AND ULTRASOUND TREATMENT IN THE FLAVONOIDS PROFILE OF A PAPAYA, MANGO AND STEVIA REBAUDIANA BEVERAGE 电子技术和超声波处理对木瓜、芒果和甜菊饮料中黄酮类化合物的影响

A high content of flavonoids in beverages is desirable not only due to their beneficial health effects, but also due to their ability to protect fruit juices from oxidation. However, food processing has a profound effect on these phytochemicals, with the further loss of most of their beneficial properties. For this reason, five flavonoids (catechin, rutin, hesperidin, quercetin and apigenin) were analyzed by HPLC in accordance to Kelebek et al. (2015) in a fruit juice-stevia beverage before and after pulsed electric fields (PEF), high voltage electical discharges (HVED) and ultrasounds (USN) treatments at two equivalent energy inputs. Catechin and hesperidin were the mayor phenolic compounds. For this two flavonoids, along with quercetin, PEF and USN treatments resulted in a significant increase compared to untreated beverage. By contrast, rutin decreased after PEF, HVED and USN treatment. The highest decrease was obtained when the beverage was treated by HVED at 256 kJ/kg. Apigenin content was far much lower in the USN treated beverage, as this compound may be severely affected by the cavitation phenomena. Results obtained highlight that individual flavonoids undergo different changes immediately after HVED, PEF and USN treatments, whereas others remain unchanged or are even enhanced.

Catechin and hesperidin were the mayor phenolic compounds. For this two flavonoids, along with quercetin, PEF and USN treatments resulted in a significant increase compared to untreated beverage. By contrast, rutin decreased after PEF, HVED and USN treatment. The highest decrease was obtained when the beverage was treated by HVED at 256 kJ/kg. Apigenin content was far much lower in the USN treated beverage, as this compound may be severely affected by the cavitation phenomena. FORMULATION OF GLUTEN FREE BREAD FRIENDLY FOR DIABETICS 对糖尿病患者友好的无麸质面包配方

To reduce the glycaemic index of gluten free foods maintaining their nutritional and sensory characteristics is nowadays a scientific challenge and the aim of the present investigation. To achieve the goal the formulation of a commercial gluten-free pre-mix was improved by adding as novel ingredients concentrated rice protein, inulin and  robusta coffee silverskin extract (RCSE) (del Castillo M. D. et al., 2013) enriched in phytochemicals presenting antioxidant and antidiabetic properties (del Castillo M. D. et al., 2014; Fernandez-Gomez B. et al., 2016; Mesias M. et al., 2014). Breads were prepared following the instructions of the manufacture of the pre-mix in presence and absence of RCSE (controls) in an automatic bread maker. The bread characterization was performed by analysis of protein, total carbohydrates, sugars, total phenolic compounds, overall antioxidant capacity, activity of alfa-glucosidase and sensory analysis. A novel bread source of protein, high content of dietary fibre, enriched in antioxidants (2.23 mg eq. CGA/g of bread) and inhibitors of alfa-glucosidase (IC50 27 mg/mL) as well as with acceptable sensory quality was obtained. In conclusion, a novel food friendly for celiac and diabetics has been developed.

In vitro protective effect of the association of wine polyphenols, vitamin E and zinc on muscle fatigue and damages induced by physical exercise 葡萄酒多酚、维生素E和锌联合对肌肉疲劳和运动损伤的体外保护作用

Period of intense or prolonged muscle activity can cause a decline in performance as well as muscle weakness. If the weakness is largely and quickly reversible, it is named fatigue. If the weakness is slowly or poorly reversible and associated with structural changes, it is described as muscle damages and can lead to immediate and/or delayed soreness. The study aimed to investigate the protective effect of the association of wine polyphenols, vitamin E and zinc on muscular fatigue and damages on human skeletal muscle cells (HSMC) challenged with A23187 calcium ionophore, mimicking a physical exercise (patent application filed). Fatigue and damages were monitored by quantifying release of lactate and IL-6, and activity of creatine kinase. A23187 significantly increased the release of lactate and IL-6 and decreased the intracellular activity of creatine kinase in HSMC. The association of wine polyphenols, vitamin E and zinc was able to significantly protect the cells against the effects of A23187, in a higher way than the ingredients alone. The study demonstrated the ability of the association of wine polyphenols, vitamin E and zinc to protect muscles against fatigue and damages. Consequently it could contribute to the optimization of performance and recovery during physical exercise.

CONTENT AND PHYSIOLOGICAL PROPERTIUES OF POLYPHENOLS IN HIGH- AND LOW-TANNINS FABA BEAN SEED APPLIED IN POULTRY DIET 家禽饲粮中高、低单宁蚕豆籽中多酚的含量及生理特性

Composition and physiological properties of polyphenol fractions from faba bean with low tannin content (LT), compared with high tannin FB (HT) and soybean meal (SBM) was to investigated. In comparison with HT seeds, in LT ones two times less polyphenols content, higher ABTS and DPPH and but lower FRAP values was determined. Irrespective of the FB type seeds, SBM was characterized by a lower polyphenol content and lower value of ABTS and FRAP. Both types of FB seeds were used as a substitute for SBM at three dietary levels: 10, 20 and 30% of a diet for for growing turkeys. After 8 weeks of experimental feeding, biochemical serum parameters and liver TBA were determined. Differences in the content of polyphenols and antioxidant properties of faba beans seeds had no effect on the analyzed biomarkers, except for lowering effect on blood glucose level caused by low-tannin seeds. Most observed effects, irrespective to the applied dietary dosages, were connected to the dietary substitution of SBM by Vicia faba beans, and as the whole should be regarded as positive changes.

Influence of epigallocatechin-3-gallate (EGCG) and ascorbic acid (AsA) on heavy metal tolerance in plants 表没食子儿茶素-没食子酸酯(EGCG)和抗坏血酸(AsA)对植物重金属耐受性的影响

Heavy metals induce reactive oxygen species (ROS) and damage of DNA or protein resulting in cell death and plant growth inhibition. It is important to investigate mechanism of detoxification and tolerance of heavy metals to improve plant growth under heavy metal stress. Here, we elucidated that the effect of epigallocatechin-3-gallate (EGCG), the major polyphenol in green tea, and ascorbic acid (AsA) on heavy metal tolerance in Arabidopsis and tobacco. Germination and seedling growth in Arabidopsis and tobacco were inhibited by Cd, Cu, Zn, Ni or Hg treatment with their concentration. Exogenous EGCG did not affect Cu tolerance but ameliorated the inhibition of seedling growth by Cd, Ni, Hg but Cu in Arabidopsis and tobacco. H2O2 and lipid peroxidation greatly increased by these metals but EGCG ameliorated them suggesting that EGCG plays a role as an antioxidant. On the other hand, AsA application remarkably enhanced the Hg-inhibited seedling growth of Arabidopsis and tobacco. When Arabidopsis seedlings were treated with AsA and Hg, higher chlorophyll and lower H2O2 were determined in comparison to Hg alone treatment. Our results suggest that EGCG and AsA could protect plant cells from heavy metal induced ROS mediated toxicity.
