IMPACT OF STRATEGIC FLEXIBILITY ON CRISIS MANAGEMENT: EXAMINING THE MEDIATING ROLE OF COGNITIVE ABILITY 战略灵活性对危机管理的影响:认知能力的中介作用检验

This study explored impact of Strategic flexibility on crisis management and also examined the mediating role of cognitive ability of decision makers in telecommunication sector of Amman, Jordan. This study was quantitative & cross-sectional by using the deductive approach and positivist philosophy. Primary data acquired through instrument based on close-ended questions. There were five relevant range of choices for agreement and disagreement. Data acquired by simple random sampling that is component of probability sampling from the target population. A total of 352 Managers and Assistant Managers were identified as the sample size out of 3500 total population at margin of error 5% and confidence level 95%. (Krejcie & Morgan, 1970). Data analyzed and interpreted by SPSS and PLS-SEM. Descriptive analysis performed by SPSS and model measurement and verification performed by PLS-SEM. There were 5-objectives that were accomplished by developing & testing 5-hypotheses. All direct relationships tested positive and significant amid independent & dependent variables. The study approved mediating role between CA, SF and CM.

EVALUATION OF IMMUNOGENICITY AND DOSE DEPENDENT IMMUNE RESPONSE AGAINST COMMON BOVINE ORIGINATED MASTITOGENS IN RABBIT MODEL 兔模型对普通牛源乳腺炎原免疫原性及剂量依赖性免疫反应的评价

This study evaluated the immunogenicity and dose-dependent immune response of toxinotypes of bovine origin viz Staphylococcus aureus (tst), Streptococcus uberis ( Cpn-60 targeted STUB ), and Escherichia coli (aggR) in a rabbit model – a step forward towards an effective polyvalent mastitis vaccine. To evaluate the primary and secondary immune response (immunogenicity) against the concentration of 10 6 cells/mL of each preparation containing subject vaccinal isolates, 24 rabbits were divided randomly into 4 equal groups viz A, B, C and D. Each antigenic preparation was inoculated to the rabbits of group A to C @ 0.2 mL SC while group D served as control (placebo) at day 0 as priming dose and a booster dose at day 7 respectively. The serum antibodies titers were recorded in terms of Optical Density (OD) values at day 0 (pre-inoculation), 7, 14, 21 and 28 (post-inoculation) by iELISA Mastitis Kit (Abbexa, UK). The antibody response was significantly higher (p<0.05) in the post-booster samples at day-14 and day-21 for all the selected isolates. The highest primary antibody response (1.74 ± 0.14) as well as secondary immune response (2.04 ± 0.13) was shown by Strept. uberis (Cpn-60 STUB) in group B on day-14 followed in order by group C and A respectively. Likewise, dose-dependent immune response to composite antigens of 3 different antigenic concentrations of vaccinal isolates: 10 8 cells/mL; 10 10 cells/mL; 10 12 cells/mL of each of vaccinal isolates in 18 adult rabbits divided randomly into 3 groups of 6 (A, B and C) respectively for dose standardization was evaluated. Serum samples were collected at weekly intervals following completion of 2 nd shot of inocula of each dose for 3 consecutive weeks. The rabbits of group B injected with 10 10 cells mL -1 of S. aureus (tst) showed a significantly higher (p<0.05) serum iELISA O.D value (2.97 A, a ± 0.10) followed in order by E. coli (aggR) (2.84 a ± 0.08) and Str. uberis (cpn60 STUB) (2.78 a ± 0.07) at day-14 post-inoculation. This study revealed the concentration of 10 10 cells/mL of each subject mastitogens as a standard bacterial load for polyvalent mastitis vaccines with higher and sustained antigenicity in rabbits.

EFFECTS OF CIPROFLOXACIN-OXYTETRACYCLINE COMPOUNDS ON TOBACCO SEEDLING GROWTH IN HYDROPONICES 环丙沙星-土霉素复合物对水培烟苗生长的影响

Tobacco leaf quality matter its value but is easily influenced by environmental factors, such as antibiotics, which reside in soil by utilization of manure. Experiments were conducted by adding ciprofloxacin (CIP) and oxytetracycline (OTC) compounds to hydroponic tobacco seedlings with three varieties, Honghua Dajinyuan (HD), Yunyan87 (Y87) and K326, and their growth was observed. Results showed that concentration of chlorophyll a decreased by 83.33%, 88.36% and 87.59% in HD, Y87 and K326 at 60 mg L -1 CIP+60 mg L -1 OTC treatment. Aboveground fresh weight decreased from 1.24 g to 0.2 g in HD, from 0.53 g to 0.25 g in Y87 and from 0.83 g to 0.38 g in K326 at 60 mg L -1 CIP+60 mg L -1 OTC treatment. With increasing antibiotic concentrations, CIP was accumulated from 1.33 to 1.56 µg g -1 in Y87 and from 4.66 to 7.98 µg g -1 in K326, and OTC was accumulated from 0.54 to 0.84 µg g -1 in Y87 and from 3.30 to 5.53 µg g -1 in HD. With increasing antibiotic concentrations, chlorophyll and carotenoid contents, root activity, enzyme activity and fresh weight decreased significantly, and more antibiotics accumulated from 0.54-7.98 µg g -1 differently in all three varieties of tobacco. Comparing experiment results it suggests that chlorophyll and carotenoid contents and aboveground fresh weight decreased less in Y87, and it accumulated less antibiotics as well. Y87 could grow better and accumulate less antibiotic compared with the other two varieties.

IDENTIFICATION OF REFERENCE GENES FOR GENE EXPRESSION ANALYSIS USING RT-QPCR IN PINELLIA TERNATA 半夏rt-qpcr基因表达分析的内参鉴定

Pinellia ternata (Thunb.) Briet. is a well-known traditional Chinese herbal medicine that has many beneficial effects such as anti-tumor, anti-fertility, blood fat-reducing, and liver-protective. The comprehensive utilization value of P. ternata is high and the development prospect is broad. However, low tuber production resulting from “sprout tumble” (ST) due to high temperature and light intensity has largely reduced its use. Therefore, it is necessary to explore the functional gene responses to high temperature and light. Real-time quantitative PCR (RT-qPCR) is commonly used for accurately gene expression detection during gene function exploration. However, because the RT-qPCR results may vary depending on the conditions and environment, an internal reference gene (IRG) is needed to normalize the RT-qPCR data. Analyzing IRGs in P. ternata can help with subsequent functional characterization and verification of the genes regulating P. ternata growth. In this study, we screened 8 suitable IRGs, and determined the most suitable IRGs using the Ct value, GeNorm, NormFinder, and BestKeeper under shade and high-temperature conditions, and in different tissues. The results showed that 18S was a suitable IRG for P. ternata studies. This discovery has laid the foundation for further research on P. ternata , and is also beneficial for the exploration of more IRGs in medicinal plants.

The comprehensive utilization value of P. ternata is high and the development prospect is broad. However, low tuber production resulting from “sprout tumble” (ST) due to high temperature and light intensity has largely reduced its use. Therefore, it is necessary to explore the functional gene responses to high temperature and light. Real-time quantitative PCR (RT-qPCR) is commonly used for accurately gene expression detection during gene function exploration. However, because the RT-qPCR results may vary depending on the conditions and environment, an internal reference gene (IRG) is needed to normalize the RT-qPCR data. Analyzing IRGs in P. ternata can help with subsequent functional characterization and verification of the genes regulating P. ternata growth. In this study, we screened 8 suitable IRGs, and determined the most suitable IRGs using the Ct value, GeNorm, NormFinder, and BestKeeper under shade and high-temperature conditions, and in different tissues. The results showed that 18S was a suitable IRG for P. ternata studies. MOLECULAR CHARACTERIZATION OF COLD TOLERANT GERMPLASM OF PHASEOLUS BEANS WITH SEQUENCE RELATED AMPLIFIED POLYMORPHISM (SRAP) AND RETROTRANSPOSON-BASED INTERPRIMER BINDING SITES (iPBS) MARKERS 利用序列相关扩增多态性(SRAP)和基于反转录转座子的引物间结合位点(iPBS)标记对菜豆耐寒种质进行分子鉴定

In this study genetic diversity of 55 Phaseolus sp. beans selected for cold tolerance from different regions of Türkiye was investigated by using sequence related amplified polymorphism (SRAP) and retrotransposon-based interprimer binding sites (iPBS)markers. Four commercially registered cultivars, one accession of Phaseolus coccineus and a tepary bean Phaseaolus acutifolius species were included for comparison. Genomic DNA was isolated from young fresh leaves and PCR reaction was carried out using 30 SRAP ( sequence related amplified polymorphism) and 12 iPBS (retrotransposon-based interprimer binding sites) primers. Similarity analyses were performed and dendrograms were produced according to the Unweighted Pair-Group Mean Arithmetic method (UPGMA). In PCR reactions, 331 total and 146 polymorphic bands were produced with 30 SRAP primer combinations. The number of polymorphic bands ranged between 1 and 12 with an average 4.86 polymorphic marker for each primer pair. Twelve iPBS primers produced 156 total bands and 72 of them were polymorphic. The highest polymorphism was obtained with SRAP primer combinations of Me8Em3 and Me7Em14 and iPBS primers 2270, 2394 and 2252. Characterization of germplasm with SRAP and iPBS primers was discussed in relation to cold tolerance, species, source, seed size, seed color and growth type. In conclusion, genetic variability of germplasm of 55 Phaseolus bean species, genotypes and cultivars selected for cold tolerance were effectively assessed by PCR based molecular techniques, SRAP and iPBS. High levels of polymorphism determined in the core collection may be used in breeding programs for the development of cold tolerant superior cultivars
