Tannic Acid Regulates Autophagy in the Ovary of Female Brandt’s Voles Affecting their Reproduction 单宁酸调节雌性布氏田鼠卵巢自噬,影响其繁殖

Tannic acid, as a polyphenol, is widely present in dicotyledonous plants and angiosperms. The study found that tannins could affect the ovarian development of Brandt’s voles. However, the specific effects of tannic acid on reproduction and its underlying mechanisms are still poorly understood. Therefore, this study explored the effects of tannic acid on the reproduction of Brandt’s voles. We treated female Brandt’s voles from four weeks postpartum with varying tannic acid doses (0 (control), 0.3% (low dose) or 0.6% (high dose) tannic acids) until 8 weeks (puberty) and 13 weeks (sexual maturity)). Ovarian specimens were collected for immunohistochemical analysis and RT-PCR. The treated Brandt’s voles were mated with male Brandt’s voles of the same age and we recorded the number of embryos, litters and the male to female ratio of the offspring. We found that the female Brandt’s voles in the low-dose tannic acid group had enhanced reproductive abilities, characterized by reduced follicular atresia and increased numbers of embryos and litters of offspring. The different doses of tannic acid altered the female-male ratio of the Brandt’s voles’ offspring. While the low-dose of tannic acid enhanced the level of ovarian autophagy by significantly increasing the expression of Beclin1 and LC3 proteins in the ovaries of the Brandt’s voles during sexual maturity. In short, tannic acid can promote the reproduction of Brandt’s voles by affecting the level of ovarian autophagy and improving the development of follicles. Tannins can boost the reproduction of Brandt’s voles.

Expression, Characterization and Purification of Latcripin-5 from Lentinula edodes Strain C91-3 and its In Vitro Anticancer Activities 香菇C91-3中Latcripin-5的表达、纯化及体外抗癌活性研究

Lentinula edodes C91-3 is an edible mushroom with demonstrated medicinal activity against various types of cancer in vitro and in vivo . The gene for Latcripin-5 (LP-5) was obtained from Lentinula edodes strain C 91-3 and inserted into a pET32a (+) vector, which was then, expressed in the Rosetta gami (DE3) strain. The purified LP-5 protein was analyzed using SDS-PAGE and western blot. The optimal solubilization parameters were found to be 0.6 mM IPGT for 6 h incubation at 37 ℃ . The solubilized protein was refolded using a refolding buffer and then dialyzed and concentrated using a dialysis buffer and PEG 20000, respectively. Phyre2 bioinformatics tool was used for protein modelling. The concentrated LP-5 protein was tested for its effect on the viability and cytotoxicity of various cancerous and non-cancerous cell lines using a cell counting kit-8 (CCK-8) assay. The LP-5 protein had the lowest IC50 value against the liver cancer line HepG2, at 58.15 μg/ml. Dose-and time-dependent morphological changes, such as cell shrinkage, blebbing formation, and cell fragmentation, were observed in treated cells. Apoptosis markers were evaluated using qPCR, flow cytometry, and western blot, and it was found that LP-5 protein increased the expression of Bax, Caspass-3, -8, -9, Cytochrome-C, and PARP, while decreasing the expression of Bcl2. Cell cycle arrest was also analyzed through qPCR, flow cytometry. Western blot results showed upregulated p21 and p27while CDK2, CDK4, CDK6, Cycline D1, and Cycline E1 were downregulated. These results suggest that LP-5 protein has potential as an anticancer agent against liver cancer cells.

Optimization of Size Exclusion Chromatography for the Purification and Quantification of Foot and Mouth Disease Virus Serotype “O” 口蹄疫病毒“O”型分离纯化及定量的粒径排除色谱优化

Foot and mouth disease (FMD) is economically devastating disease of livestock overwhelming the international trading of animal based products. The unpurified inactivated vaccine containing Foot and Mouth Disease Virus a (FMDV) and non-structural proteins (NSPs) used to control the disease stimulated the production of anti-NSP antibodies. Countries striving to establish FMD free status must ensure the absence of circulating FMDV and anti-NSP antibodies in vaccinated animals, which are produced in natural infection. Production of purified FMD vaccines contrary to other animal’s vaccines required extra step of virus purification from its NSPs. Present project was designed to optimize different parameters of size exclusion chromatography (SEC) for the purification of local isolate of FMDV from NSPs. For this purpose Bio-rad Econo-Column 15/50 was packed with Sephacryl TM S-300 resin and fitted with BioRad BioLogic LP Chromatographic system. Mixture of blue dextran and bovine serum albumin used as biomarkers for FMDV and NSPs respectively was employed to optimize resin bed height in column based on resolution between curves, while that FMDV was used to optimize flow rate of mobile phase and sample volume based on column efficiency. The chromatographic fraction of FMDV was ensured for purity through protein analysis by SDS-PAGE and serologically by 3ABC-NSP ELISA. It was found that maximum resolution of 1.86 was achieved at 48cm of resin bed height followed by 1.5 and 1.27 at 40cm and 32cm respectively. FMDV was eluted earlier with retention time 138.02+2.02min at flow of 0.7ml/min than 165.76+1.28min at 0.5ml/min along with better column efficiency with sample volume at 4% of column volume. SEC could detect and quantify from 603.75µg/ml in PEG concentrated FMDV up to 3.33µg/ml in 1:64 dilution as lower limit of detection and quantification. Presence of two bands of structural protein with absence of NSPs bands on SDS-PAGE and negative NSP% in hyperimmune sera raised in goats revealed the successful elimination of NSPs contents from FMDV suspension by SEC.

Sulfate Reducing Bacterial Corrosion of Mild Steel in Liquid and Solid Media 硫酸盐降低低碳钢在液体和固体介质中的细菌腐蚀

This study was carried out to check the corrosive impact of sulfate-reducing bacteria (SRB) on mild steel coupons in solid and liquid media under two variable nutritional conditions. The bacterial species for the experimental trials was isolated from buried corroded metallic installment and found motile, Gram- negative, non-spore former and identified by 16S rRNA gene sequencing as Desulfovibrio desulfuricans . The corrosive impact of SRB on steel coupons was performed in water as liquid medium and the processed clay as the solid medium without and with the provision of Postgate B medium. After 60 days of anaerobic incubation, corrosion rate (CR) and average percent weight loss (APWL) of the steel pieces were calculated. Higher CR and APWL were observed in the liquid medium (water) than in the solid medium (clay). The CR and APWL in water were 249 ± 2 µg dm −2 d −1 and 4.16 ± 0.25 %, respectively, while the corresponding figures for CR and APWL in clay were 108 ± 2 and 1.88 ± 0.29, respectively. In addition, the CR and APWL were higher in the presence of Postgate B medium compared to the medium without it. X-ray diffraction (XRD) and Fourier transform infrared (FTIR) spectroscopic analyses reveals the presence of FeS in the corrosion product. Our findings of this study will be helpful for devising corrosion-protecting strategies.
