Abstrak. Validitas reaksi qPCR untuk aplikasi teknologi diagnostik ataupun deteksi sangat penting. Reaksi qPCR yang valid, akan memberikan hasil dengan tingkat kepercayaan yang tinggi. Untuk mengukur validitas reaksi dapat dilakukan dengan mengukur nilai Koefisien korelasi (R2), Efisiensi (E), dan Coefficient of variability (CV) yang terbentuk. Her-2 merupakan protein proto-onkogen sebagi biomarker prognosis dan factor prediktif pengobatan terhadap trastuzumab. Penentuan skoring status HER-2 dapat dilakukan menggunakan teknik Immunohistochemistry (IHC), Fluorosence In Situ Hibriditation (FISH) dan qPCR. Dari ketiga teknik diatas, pendekatan menggunakan qPCR merupakan pendekatan yang dapat dikuantifikasi. Untuk mengkuantifikasi status skoring HER-2 dibutuhkan pengukuran amplifikasi HER-2 dan gen kalibrator. Gen WHN merupakan salah satu gen yang dapat digunakan sebagai kalibrator skoring HER-2. Efisiensi qPCR dilaporkan dipengaruhi oleh banyak faktor, diantaranya adalah pH. pH diketahui berpengaruh terhadap kerja dari polymerase dan integritas DNA. Pada penelitian ini akan dibandingkan dua pelarut berbeda, H2O pH 5,2 dan Buffer TE pH 7,8 terhadap efisiensi reaksi qPCR skoring HER-2. Preparasi pGEM-T easy whn juga akan dilaporkan pada penelitian ini menggunakan teknik yang standar, yang kemudian akan digunakan sebagai bahan DNA template untuk pengenceran secara serial dengan dua kali factor pengenceran untuk mengukur nilai R2, E, dan CV yang menjadi parameter validitas reaksi qPCR. Dari hasil penelitian ini nilai Ct pGEM-T whn yang dilarutkan dengan H2O pH 5,2 memiliki nilai Ct lebih rendah dibandingkan dengan TE buffer pH 7,8, yang secara signifikan mempengaruhi efisiensi qPCR. Nilai R2 dan E reaksi qPCR untuk H2O pH 5,2 dan TE buffer pH 7,8 adalah: 0,98 dan 115%; 0,99 dan 99% untuk masing-masingnya . Selanjutnya nilai CV yang terbentuk berada dibawah 10% pada kedua pelarut tersebut. Hasil konfirmasi dengan elektroforesis gel dan analisis melt peak untuk kedua standar menunjukkan satu pita tunggal dengan ukuran yang ditargetkan (93 bp) dan peak tunggal dengan Tm ±81 oC. Berdasarkan hasil penelitian ini, TE buffer pH 7,8 merupakan pelarut ideal untuk mendapatkan efisiensi uji qPCR yang ideal untuk aplikasi penentuan skoring status HER-2 pada pasien kanker payudara berdasarkan metode qPCR. AbstractThis study aims to describe the process of developing spiritual and social attitudes competencies, in a holistic learning of mathematics sourced in the Qur'an. Qualitative research is applied in the preparation of teaching materials based on the integration of core competencies of revised Curriculum-13, and the development of attitude competencies in mathematics learning based on the Qur'anic. The subjects of the study were SMP / MTs students in the lessons of Straight Line Equation. The research data includes documents, learning recordings, and interviews, and it’s analyzed using the Miles & Huberman Method. The study resulted in: (1) mathematics teaching materials on the topic of the Straight Line Equations sourced of the Qur'anic (al Anfal) verses 65-66, to teach holistically, (2) The process of developing competency attitudes is carried out as long as the learning takes place from introduction to closing the lesson. In developing a spiritual attitude, two students are assigned to read the verses of the Qur’an and their translations, then discuss the content of mathematics. Students are aroused by curiosity with questions about verse, working collaboratively with friends, choosing the best ideas (tolerant), and being responsible for completing tasks. In developing skills and knowledge competencies, students are guided to achieve the process of modeling problems related to the Qur'anic verses, so that they find problem solving procedures, or mathematical concepts from the topics studied.Keywords: attitude competency development, holistic mathematics learning.

AbstractThe group discussion is the activity of a group of students, speaking exchanging information and opinions on a topic, where each student wants to find answers to all the possibilities. This study aims to find out and analyze the improvement of understanding the concept of mathematics students of class IX A SMP Negeri 21 Pontianak after the learning of mathematics through discussion groups assisted props. This study is a classroom action research consisting of two cycles, with the help of teacher and student activity observation sheets, student activity sheets for group discussion and test result of learning. Based on the results of data analysis, there is an increased understanding of mathematical concepts of students Through the implementation of teaching-assisted group discussion of learning aids occur increased student activity in learning. An increase in the percentage of student activity from cycles I and cycle II by 18%. Through the implementation of learning group-assisted discussion of visual aids, there was an increase in the understanding of mathematical concepts of students from cycle I and cycle II seen from the results of concept comprehension tests provided, there was an average score increase of 9.45 and a 22.85% increase in the percentage of mastery. AbstractThe people in Rasau Jaya Umum village Kubu Raya regency have used plants as ingredient food that has been inherited from generation to generation for daily cooking or to make the typical food. The typical food in Rasau Jaya Umum village like Rengginang, crackers, and salted fish. The purpose of research to collect data of plants used as food additives by people in Rasau Jaya Umum village. The respondents were chosen through snowball sampling technique and then given a series of questions using observation sheet. The head in Rasau Jaya Umum village is chosen as the main respondent then proceeded to the traders and home industry owners who produce the typical food. The results showed that there are 18 species of 15 plant families used as natural additives, namely Liliaceae, Zingiberaceae, Amaranthaceae, Oxalidaceae, Rutaceae, Arecaceae, Poaceae, Gnetaceae, Malvaceae, Cactaceae, Pandanaceae, Piperaceae, Polygonaceae, Anacardiaceae, and Myrtaceae.Keywords: plants, food additives, Rasau Jaya Umum village

AbstractThis study aimed to determine the composition and structure of Semayang forest and the Importance Value Index (IVI) of riparian forest in its role of reducing the wave rate. The method used is multilevel plot method of 20x20 m for sampling of tree level, 5x5 m for sapling level and 2x2 m for seedling level. The result showed that the highest Importance Value Index at tree level was dominated by 3 species, they were L. javanicum (101,663), M. speciosa (76,201) and G. renghas (65,962). At sapling level dominated by P. indica (102,990). At seedling level dominated by M. pigra (95,004)andP. brevistipitata (70,109). In addition, Semayang riparian forest has an ecological function as a wave retaining. It can be seen based on the vegetation density and horizontal structure of species that make up the forest. AbstractThe Sundanese have been long used the home garden as a source of food, medicines, and construction materials. This study aims to document the plants diversity and its utilization in the Sindang Jaya Village, Disstict Cianjur, West Java. The research method was conducted by ethnobotany approach through surveys and interview. The data obtained were analyzed qualitatively by using descriptive statistics. This study found 100 species of the useful plants belonging 88 genera and 46 families in the yard in the Sindang Jaya home garden. The plants in the home garden uses as an ornamental (46 species), fruit (18 species), vegetable (14 species), and traditional medicine (species). The mangkokan (Polyschias scutellaria) and kedondong pagar (Polyscias fruticosa) are potential to be developed as the living fence as well as function as a source of food or vegetables. The Dioscorea hispida in home garden is one of the efforts of local communities to conserve wild plants.Keywords: Disocorea hispida, Etnis Sunda, Polyschias scutellaria, Polyscias fruticosa.

This study aims to investigate students’ views toward physics representations while solving physics problems by involving preservice physics teachers of two universities in Kalimantan Barat. A physics representation survey was developed to measure students’ perception about physics representations. The results show that the majority of the students (above 75%) agree that representations are beneficial for them to understand physics problems and physics concepts and also to find out the best solution of a problem. Some of the students say that they remain provide representations while solving a problem even though there is no extra credit given by instructors. Some of the students feel that they face impediment to grasp representations provided in some physcis text book.Keywords: physics representations, views

AbstractThe aim of the research was to find out the effect of learning models on mathematics achievement. The learning model compared were NHT(Numbered Head Together), snowball throwing and convensional. The type of the research was a quasi-experimental by 3×1 factorial design. The population was the eight grade student of junior high school in Cibinong district on academic year 2017/2018. The sampling tecnique was cluster random sampling. The size of the sampel was 103 students consisted of 36 students in the first experimental group, 34 students in the second experimental group and 33 students in third experimental group. The instruments used were mathematics achievement test. The data was analyzed using unbalanced one-way analysis of variance. The conclusions of the research was NHT learning model gives better mathematics achievement than Snowball Throwing and convensional learning model, and snowball throwing learning model give better mathematics achievement than convensional learning model.Keywords: NHT, Snowball throwing, mathematics achievement
摘要本研究旨在探讨学习模式对数学成绩的影响。比较的学习模式有NHT(number Head Together)、雪球投掷和常规模式。研究类型为准实验,采用3×1析因设计。研究对象为2017/2018学年慈滨农区初中八年级学生。抽样方法为整群随机抽样。样本规模为103人,其中第一实验组36人,第二实验组34人,第三实验组33人。使用的工具是数学成绩测试。数据分析采用非平衡单因素方差分析。研究结果表明:NHT学习模式的数学成绩优于抛雪球学习模式和常规学习模式,抛雪球学习模式的数学成绩优于常规学习模式。关键词:NHT,雪球投掷,数学成绩

AbstractThis study aims to determine the improvement of students' thinking skills through the Earth and Space Science (IPBA) lectures that integrate science activities with interactive multimedia and local science. The research method used was experimental design (quasy experiment) with comparative group of pretest-posttest group design. Data collection techniques that will be done are measurement techniques in the form of students' thinking skills tests and surveys in the form of student and lecturer responses to the lectures are held. Physics education student semester 4 academic year 2016/2017 will participate as research subject. The results showed that the increase (N-gain) of students' thinking skill in experimental class was 0,70 and control class was 0,40 (both medium category). By using Paired Sample Test obtained tcount = 8,338 at significance level P = 0,000. The significance level is less than 0.05 (P <α), so it can be concluded that the application of lectures that integrate local science and technology in IPBA lectures can improve students' thinking skills significantly compared with conventional lectures.Keywords: high order thinking skills, earth and space science, multimedia interaktif.

AbstractThis study aims to 1) Produce instructional textbook of physics education based on constructivistic approach, and 2) Knowing the quality of textbooks in terms of feasibility aspects of content, feasibility of presentation, language feasibility and feasibility grafika according to experts. This research is an R & D research with procedural model that adapted the procedure of ADDIE development which consists of three stages: analysis, design, and development. The research instrument is in the form of scale of textbook quality assessment using Likert scale made in the form of checlist. The subjects of textbook quality assessment are three material experts and three media experts. Textbook quality data obtained in the form of quantitative data is converted by product quality percentage scale. The result of this research are: (1) Developed instructional product in the form of textbook based on constructivistic approach as learning material of physics education evaluation in accordance with KKNI curriculum, and (2) The quality of textbook developed has excellent quality (SB) based on assessment of material experts with a percentage of 84.05% of ideal high score 148, and excellent quality (SB) based on media expert's judgment with a percentage of 82.56% of the ideal high score of 80.Keywords: textbooks, physical education evaluations, constructivistically
