Mortality of farmed coho salmon Oncorhynchus kisutch due to erythrocytic inclusion body syndrome (EIBS) has been posing severe economic losses in Japan since 1986. The etiologic agent of EIBS was reported to be piscine orthoreovirus 2 (PRV-2). We detected PRV-2 in the ovarian fluid samples of 10% of EIBS-experienced spawners (n = 30) by RT-PCR. When each of 30 individual ovarian fluid samples was inoculated into a naïve coho salmon, 37% of the blood samples from the fish tested positive for PRV-2 at 20 days post injection. These observations suggest that the ovarian fluid of EIBS-experienced fish can be contaminated with the infective viral particles of PRV-2.

― Outbreaks of suspected viral nervous necrosis emerged in cultured groupers in Guangxi, China in August 2018. Histopathology showed severe vacuolation in the brain tissue of the diseased juvenile hybrid grouper ( Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂ ). A nervous necrosis virus (NNV) was isolated from pooled sample of the brain and eyes using a grouper cell line EAGB. Cytopathic effect with multiple cytoplasmic vacuolation cells developed in EAGB cells. The coat protein gene sequence of the isolate (GNNV-Gx) showed that it belonged to RGNNV genotype and was most closely related to that of a tiger grouper NNV registered in GenBank. The infectivity of the isolate decreased remarkably at pH 3 and pH 10 and was susceptible to 60 ° C.

Streptococcus Infections in Cultured Pond Loach Paramisgurnus dabryanus in China 中国池塘泥鳅链球菌感染的研究

In 2017, high mortalities were observed in cultured pond loach Paramisgurnus dabryanus in Tianjin, China. The symptoms observed in the moribund loach included hemorrhage on the body surface and head, exophthalmos, and erratic swimming around the water surface. Two different Gram-positive bacteria were isolated from the diseased loach in two different fish farms. They were identified as Streptococcus dysgalactiae and Streptococcus agalactiae, using 16S rRNA gene sequencing and biochemical characterization. The median lethal doses of the bacterial strains in the loach were 3.2 × 10 and 5.0 × 10 CFU/fish, respectively.

Worm Cataract of Hatchery-Reared Japanese Dace Tribolodon hakonensis Caused by Diplostomum sp. (Digenea: Diplostomidae) 孵化饲养的日本海参双孔虫引起的白内障(didi亚纲:双孔虫科)

In 2015, cataract was observed in juvenile Japanese dace Tribolodon hakonensis, reared in a hatchery in Nagano Prefecture, Japan. Metacercariae were found in the lens, with the prevalence and mean intensity of infection of dace (0.5–2.4 g in body weight) reaching 100% and 39.7, respectively, in the most heavily infected fish group. Histologically, the lens became partially fragmented around the metacercariae, which occupied a considerable space, surrounded by their own excreta. Among snails sampled from outdoor ponds and waterways, cercariae with a bifurcated tail were found in the lymnaeid, Radix auricularia japonica. ITS1-5.8S-ITS2 and cytochrome oxidase subunit I (COI) sequences of metacercariae from the lens of dace and cercariae from the lymnaeid were nearly identical to each other, indicating that the lymnaeid is the first and dace is the second intermediate host. However, since the obtained COI gene sequences were close but not identical to known Diplostomum spp., this species was not identified to the species level. The morphology of metacercaria was also described. This is the second reported case of worm cataract in fish caused by Diplostomum metacercariae in Japan.

In Vitro Biofilm Formation of Flavobacterium psychrophilum 嗜冷黄杆菌体外生物膜的形成

The fish pathogen Flavobacterium psychrophilum has been shown to occur in two colony phenotypes, rough and smooth. The aim of this study was to examine the effect of environmental and nutritional factors on the biofilm formation of both phenotypes under in vitro static conditions. The present study revealed that smooth cells displayed higher abilities of biofilm formation compared to rough cells in nutrient-rich media (TYES broth and fish feed) under static conditions, while biofilms formed by cells of both phenotypes were mainly Ca2+ dependent than Mg互 Neither of the colony phenotypes was able to form biofilms in the low-nutrient medium (lake water) within 5 days.

We developed a PCR-RFLP method to distinguish Benedenia seriolae and Neobenedenia girellae, both infecting amberjacks Seriola spp. more precisely than a morphological method. PCR products of 28S rRNA and COX1 were digested by Cla I and EcoR I, respectively. For all individuals of B. seriolae, the digestions split each of the PCR products into two fragments. Great cormorants Phalacrocorax carbo were found dead in a goldfish farm in Aichi Prefecture. They were infected with trematodes in the oral cavity. Based on morphological observation and genetic analysis, the trematodes were identified as Clinostomum complanatum. This is the first case reporting C. complanatum infection of great cormorants in Japan. Heavy metacercarial infection of goldfish was found in the same fish farm. The metacercariae were shown to be the same species as the adult worms from the bird by molecular analysis. These results suggest that great cormorant is involved in the life cycle of this trematode in

Inheritance of the Resistant Trait in Goldfish against Herpesviral Hematopoietic Necrosis 金鱼抗疱疹病毒造血坏死抗性性状的遗传

― It was reported that progeny from the parents of goldfish Carassius auratus which had survived in herpesviral hematopoietic necrosis (HVHN) increased resistance against the disease, and consequently a resistant strain of Azumanishiki variety has been established in Saitama Fisheries Research Institute. In this study, we started breeding between the resistant Azumanishiki variety and a susceptible Kurodemekin variety and investigated the heredity of the resistant trait in the offspring in order to establish a new resistant strain of Kurodemekin. F 1 bred between resistant Azumanishiki and Kurodemekin showed resistance in the challenge test with the causative virus (cyprinid herpesvirus 2), but F 1 had no pop-eyed character. Crosses between F 1 survivors in virus challenge produced F 2 , and about 25% of F 2 showed pop eyes. We finally obtained the resistant pop-eyed Kurodemekin of F 3 crossed between pop-eyed F 2 survivors in virus challenge. Backcrosses between F 1 survivors and Kurodemekin were also tested for the resistance in virus challenge. The mortality rates of F 1 , F 2 , F 3 and the backcrosses in virus challenge suggested that the resistant trait against HVHN in the goldfish linage exhibited a Mendelian dominant inheritance pattern.

First Report of Thelohanellus testudineus (Myxosporea) from the Skin of Goldfish Carassius auratus in Japan 日本金鱼Carassius auratus皮肤上的粘孢子目testudineus首次报道

― In 2017, one individual of goldfish Carassius auratus which had tumor-like massive lesions in the skin was found in a goldfish wholesale market in Aichi Prefecture, Japan. The lesions were 2–4 mm in diameter and hemorrhage was observed in some of them. Microscopic observation revealed numerous myxospores with a single polar capsule. The spores were 17.1 ± 1.0 μ m (mean ± SD) long, and 6.1 ± 0.7 μ m wide. The partial sequence of 18S rRNA gene was 100% identical to that of Thelohanellus testudineus . This is the first report of T. testudineus in goldfish.
