Phytochemical analysis, cytotoxic, antioxidant and anti-diabetic activities of the aerial parts of Sorghum halepense 高粱地上部分的植物化学分析、细胞毒、抗氧化和抗糖尿病活性

Phytochemical screening, cytotoxic activity, total phenolic content, antioxidant and anti-diabetic activities of the Sorghum halepense methanolic extract and its different fractions were evaluated. Methanolic extract and its various fractions revealed the presence of reducing sugars, tannins, steroids,  glycosides, flavonoids in the methanolic extract where they were absent in n-hexane fraction except flavonoids. Gums and saponins were absent in all the samples. Methanolic extract indicated the highest cytotoxic (80.7 ± 1.3%) and anti-diabetic (62.5%) activities. The maximum total phenolic contents (28.7 ± 1.4 mg/mL) were found in the chloroform fraction. An aqueous fraction expressed the highest antioxidant activity 74.1 and 97.1% free radical scavenging properties in DPPH and ABTS assays respectively whereas, in the case of H2O2, methanolic extract indicated maximum (36.9%) activity. In conclusion, the extract of aerial parts of S. halepense is a source of compounds against cancer, diabetic and free radical associated disorders.

Antidepressant-like activity of hydroalcoholic extract of Agaricus blazei in stressed and unstressed mice 姬松茸水酒精提取物对应激和非应激小鼠的抗抑郁活性

The aim of this study was to investigate the antidepressant activity of hydroalcoholic extract Agaricus blazei (273 and 819 mg/kg; orally) in stressed and unstressed Swiss albino mice. Mice were immobilized to induce stress. Fluoxetine 20 mg/kg orally was given to stressed and unstressed animals and immobility time was noted by using forced swim test and tail suspension test. The concentration of plasma nitrite was also evaluated in stressed and unstressed mice. The effect of prazosin (α1-adrenoceptor antagonist), p-CPA (parachlorophenylalanine–tryptophan hydroxylase inhibitor) and 7-nitro-indazole (nNOS inhibitor) on the antidepressant activity of A. blazei was also evaluated. A. blazei and fluoxetine significantly decreased the duration of immobility time in stressed and unstressed mice, showing significant antidepressant activity. No substantial change was found in the locomotor activity. However, a significant reduction in the level of plasma nitrite was also noted in stressed mice. Hydroalcoholic extract showed prominent antidepressant activity in mice.

Antimicrobial assay of endophytic fungi from Rumex madaio and chemical study of strain R1 马马菌内生真菌抑菌活性测定及菌株R1的化学性质研究

In order to search more antimicrobial natural products for new drug discovery, bioassay and chemical investigation of endophytic strains from Rumex madaio were carried out. Six fungal strains (numbered as L1, L6, L9, L10, R1, R2) were successfully isolated and their antimicrobial effects on three human pathogens, Escherichia coli, Staphyloccocus aureus and Candida albicans, were evaluated. The results indicated that ethyl acetate extracts of fermentation broth of these strains had no potent antimicorbial activity against E. coli and C. albicans while strains L1 and R1 had stronger inhibitory effects on S. aureus. On basis of morphological characteristics and 18S rRNA sequence analysis, strains L1 and R1 were respectively classified as Colletotrichum and Fusarium geni. The chemical study of strain R1 led to isolation of two known antimicrobial agents, dibutyl phthalate (1) and beauvericin (2).

Antibacterial activity of Weissella confusa by disc diffusion method 圆盘扩散法抑菌活性研究

A visual experiment was conducted to investigate the antimicrobial potential of cell-free culture supernatant of Weissella confusa (DD_A7) against the foodborne pathogenic bacteria Escherichia coli 0157: H7 by disc diffusion method. The result revealed that DD_A7 exerted effective antibacterial potential to control the growth of E. coli upon treatment with 100 µg/mL of the culture supernatant. This study demonstrated that DD_A7 is a potent probiotic candidate to reduce the risk factor associated with Shiga toxin-producing E. coli. Hydrogen sulfide has been shown to protect  myocardium against ischemia-reperfusion injury by preserving interfibrillar mitochondria functional activi-ties than subsarcolemmal mitochondria. In this study, the role of the KATP channel in modulating the mitochondrial subpopulations during the cardioprotection mediated by NaSH (H2S donor) was investigated. Isolated rat hearts were treated with mitochondrial KATP channel closer glibenclamide (10 μM)/opener diazoxide (0.8 mM) via Langendorff perfusion apparatus before ischemia-reperfusion. The results showed that NaSH pre-conditioning in presence of glibenclamide significantly improved cardiac recovery without any significant difference between interfibrillar mitochondria and subsarcolemmal mitochondria.  In conclusion, targeting KATP channel may not be good option to target interfibrillar mitochondria/subsarcolemmal mitochondria against ischemia-reperfusion injury.

This study was conducted to evaluate the topical efficacy of Solanum incanum for the treatment of partial-thickness burn in mice model. Mice were treated with topical ointment of S. incanum three times daily for 14 days. The wound healing was observed through wound contraction and histological parameters. The group treated with S. incanum ointment showed 81% reduction in wound area as compared to negative control where wound area reduced to 22%. The histological analysis further confirmed that ointment favors the tissue regeneration and reepithelization thus heal wound rapidly as com-pared to other groups. In conclusion, S. incanum extract enhances wound healing and tissue regeneration.

Nephroprotective effect of Costus pictus extract against doxorubicin-induced toxicity on Wistar rat 木香提取物抗阿霉素致Wistar大鼠肾保护作用

The present study was conducted to evaluate the nephroprotective effect of a medicinal herb Costus pictus against doxorubicin-induced toxicity. Rats were divided into six groups and treated with doxorubicin and ethanol extract of the C. pictus. Doxorubicin was administered intraperitoneally with a single dose (4 mg/kg) per week for three weeks. The extract (200 or 400 mg/kg) was administered orally for 4 weeks to two doxorubicin groups. Significant changes of the serum kidney markers, albumin, urea, uric acid and creatinine, and glutathione peroxidase, glutathione–S-transferase, catalase, superoxide dismutase, reduced glutathione and lipid peroxides in the kidney of doxorubicin-treated rat were observed. Histological features were also severely affected. However, biochemical and histological changes in the extract-treated rat were non-significant, showing that the herb is nephroprotective. The effects were comparable to the anti-oxidant vitamin E.
