Inhibitory Effects of Radix Pulsatillae Extract on Insoluble Glucan Synthesis and Adhesion of Streptococcus mutans 白头翁提取物对变形链球菌不溶性葡聚糖合成及黏附的抑制作用

Streptococcus mutans plays a virtal role in trigering dental caries establishment due to its ability to synthesize two significant factors. The two factors are organic acids and glucans. The former demineralized dental enamel and the latter mediates the attachment of bacteria to tooth surface. It is believed that demineralization of dental enamel and attachment of bacteria are the crucial events that indicate and develop dental caries. For this reason, we studied the effect of the ethanol extracts of Radix Pulsatillae on the growth and acid production of S. mutans. Ethanol extracts of the Radix Pulsatillae showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition compared to the control groups (p<0.05). The extracts inhibited S. mutans adherence to hydroxyapatite treated with saliva, and cell adherence was repressed by Radix Pulsatillae. the ethanol extract of Radix Pulsatillae showed remarkable inhibition of glucosyltransferase, which synthesizes water insoluble glucan form sucrose. Phytochemical analysis showed Radix Pulsatillae contained major components such as phenolic compounds, glycosides, steroids, terpenoid, and saponin. These results suggest that Radix Pulsatillae may have anti-cariogenic properties, which may be related with major components such as phenolic compounds, glycosides, steroids, terpenoid, and saponin.

용담초 추출물의 화장품 소재로서 약리활성에 관한 연구 龙潭草提取物作为化妆品材料的药理活性研究

In this study, we investigated the effect of ethanol extracts from Gentianae scabrae bunge (GSB) on the activities of antioxidant, whitening and anti-inflammation. Viability of cells was measured by neutral red (NR) assay, and inhibitory effects of GSB on melanin synthesis was determined the melanin production in B16F10 cells. The expression level of matrix metalloproteinase-1 (MMP-1) in media was analyzed by ELISA kit, and nitric oxide (NO) production in RAW264.7 cells was monitored by measuring the nitrite content in culture medium. GSB showed highly efficacy in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and significantly reduced melanin synthesis in B16F10 cells. MMP-1 production in UVB-stimulated human dermal fibroblast (HDF) cells was inhibited by GSB treatments. NO production was suppressed by the treatment of GSB in LPS-stimulated RAW264.7 cells. From this results, it was indicated that GSB could be utilized as anti-aging and whitening cosmetic ingredients.

Study of Developmet of External Medicine Based on the Result of Prior Study and Folk Remedy Using Scolopendra subspinipes mutilans as a Single Medicine in Korea and China 韩中两国以残棘棘棘为单药的外用药发展及民间偏方研究

Scolpendra subspinipes (centipede) is pungent in flavor, activates the channels to stop pain, and, removes toxins and resolves masses. So it has been used as a folk remedy. We researched folk remedies which used centipede as a single medicine and results of its prior clinical and experimental study, and figured out that centipedes are effective on ulcerative disease, herpes zoster, clavus, insect bite, inflammatory disease, furunculus, pain disease by external therapy, and oral administration is effective on anti-cancer such as hepatoma and uterine cancer, cerebrovascular disease, inhibition of atherosclerosis, sedation of central nervous system, antipyretic action, and anti-inflammation, and centipede Herb-Acupuncture is effective on inflammatory disease and pain disease. Centipede preparation materials for external therapy were sesame oil, perilla oil, vinegar, salt, and, burnt alum. Fumigation of centipede treated paronychia, pertussis, and hemorrhoids. For the external therapy, the way of centipede powder mixing with other materials has been used. It is needed that we use dusted powder to make skin absorb its ingredients more easily, and we study intensely minor ingredients to make centipede components absorbed more effectively and do pharmacological action. Also, fumigation, a kind of another external therapy, is considered to be effective treatment of upper respiratory infection like pertussis, so it needs an in-depth study on centipede fumigation.

Anti-cancer Effects of Scutellaria barbata in AGS Human Gastric Adenocarcinoma Cells 枸杞子对AGS人胃腺癌细胞的抗癌作用

The aim of the study is to investigate the anti-cancer effects of Scutellaria barbata in AGS human gastric adenocarcinoma cells. MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay and caspase 3 or 9 activity assay were carried out to examine cell death with Scutellaria barbata. To elucidate the inhibitory effects of Scutellaria barbata, cell cycle (sub-G1) analysis and mitochondrial membrane potential were performed in AGS cells after 24 h incubation with Scutellaria barbata. Scutellaria barbata induced apoptosis in AGS cells by using the MTT assay, the sub-G1 analysis and mitochondrial membrane potential assay. The stronger inhibition effects of AGS cell growth was observed by application of Scutellaria barbata combined with several anti-cancer drugs (paclitaxel, 5-fluorouracil, cisplatin, ectoposide, doxorubicin and docetaxel) in comparison to the application of Scutellaria barbata or anti-cancer drugs. Our findings provide insight into unraveling the effects of Scutellaria barbata in human gastric cancer cells and developing therapeutic agents against gastric cancer.

MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay and caspase 3 or 9 activity assay were carried out to examine cell death with Scutellaria barbata. To elucidate the inhibitory effects of Scutellaria barbata, cell cycle (sub-G1) analysis and mitochondrial membrane potential were performed in AGS cells after 24 h incubation with Scutellaria barbata. Scutellaria barbata induced apoptosis in AGS cells by using the MTT assay, the sub-G1 analysis and mitochondrial membrane potential assay. The stronger inhibition effects of AGS cell growth was observed by application of Scutellaria barbata combined with several anti-cancer drugs (paclitaxel, 5-fluorouracil, cisplatin, ectoposide, doxorubicin and docetaxel) in comparison to the application of Scutellaria barbata or anti-cancer drugs.
