Ewing's Sarcoma is a rare malignant tumor that grows in bones or nearby soft tissues. Current treatment focuses on chemotherapy, radiation therapy, and surgery, yet chemoresistance is common and remains the leading cause of treatment failure. PC002 is recently found effective in treating Ewing's Sarcoma in vitro and in vivo. However, the mechanism of action of PC002 is still under dispute, and no experiments have been conducted to explore its combination therapy in Ewing's Sarcoma cells. Thus, this study was performed to elucidate the mechanism of action of PC002 and facilitates the selection of future combination therapies for Ewing's Sarcoma. Vincristine, ABT-199, ABT-263, and Olaparib were combined with PC002. High-throughput screening and Fluorescence-activated Cell Sorting tests were performed in the A673 Ewing cell line to examine the drugs’ effects on inhibiting cell proliferation and inducing apoptosis. The results suggest that there was no antagonism between PC002 and these drugs. We discovered that the combination of PC002 and ABT-263 is highly synergistic (p<0.001), which should be further explored as a promising combination drug. The combinations of PC002 with Vincristine and with Olaparib are also potential therapeutic tools in combating chemoresistance and increasing the efficacy of the drugs in Ewing's Sarcoma.

Objective: To analyze the prescription rules of Chinese medicine health foods for improving sleep, and to provide references for the research and development of related health foods. Methods: Retrieve 581 types of sleep-improving Chinese medicine health food data approved before December 2020 from the official website of the State Administration of Market Supervision and Administration (SAMR), establish a health food database and conduct an overall analysis, including dosage form, function, frequency, etc., for raw materials The 109 formula, which is all Chinese herbal medicine, carries out frequency statistics, association rule analysis and visual analysis. Results: There were only 418 kinds of sleep improving effects, including 163 kinds of two or more effects. All of them were Chinese herbal medicines with high frequency of use, such as SEMEN ZIZIPHI SPINOSAE, PORIA and FRUCTUS SCHISANDRAE CHINENSIS. According to the efficacy classification, the top three traditional Chinese medicines with the highest frequency of use were Deficience-tonifying Drugs, Anxiety Drugs and Liver-soothing and Wind Resuscitating Drugs. The combination rule high-frequency drug pair includes RADIX SALVIAE MILTIORRHIZAE → SEMEN ZIZIPHI SPINOSAE; FRUCTUS SCHISANDRAE CHINENSIS, PORIA → SEMEN ZIZIPHI SPINOSAE; RADIX ET CAULIS ACANTHOPANACIS SENTICOSI → SEMEN ZIZIPHI SPINOSAE, etc.; The visualization results of association rule drug pairs show that the core compatibility is SEMEN ZIZIPHI SPINOSAE - PORIA - GINSENG - FRUCTUS SCHISANDRAE CHINENSIS - RHIZOMA GASTRODIAE. It is basically consistent with the principle of improving sleep in traditional Chinese medicine. Conclusion: Data mining and basic theoretical analysis of traditional Chinese medicine on the prescription rules of existing sleep-improving health foods can provide new ideas for the research and development of traditional Chinese medicine health products, so as to give full play to the advantages of traditional Chinese medicine and provide a basis for the public service of Chinese medicine health foods.

The focus of this research project is to predict the location of phosphorylation on human proteins by their amino acid sequences. Signals are sent into cells through receptors during a process called signaling cascade, which may alter the state of cells. One essential element of signaling cascade is protein kinases, which are enzymes that give the target protein a phosphate group. They can activate, deactivate proteins or act as signals to be passed onto other proteins. Through the Convolutional Neural Network (CNN), our model aims to provide a plausible prediction of the phosphorylation pattern of a given human protein sequence, on which future experiments can be based.

Muscular dystrophy and neuromuscular diseases have been persisting and its causes range from genetic inheritance to injuries, resulting in muscle weakening and paralysis. In consideration of muscle contraction and its relevance to the mitochondria, an investigation into primary skeletal muscle cells was carried out to observe the amino acid taurine, focusing on its effect on the mitochondria from a cellular level. It was concluded that taurine may slow down the progressive muscle weakness and aging through enhancing calcium homeostasis and suppressing the production of ROS. Taurine deficiency could also be associated with aging animals, indicating that taurine may have anti-aging actions. Thus, it was assumed that taurine deficiency is associated with the increase in aging-associated tissue damage, reducing the life span of the mouse model by knocking out the taurine transporter. In addition, it was concluded that a reduce in taurine level is associated with the rise of unfolded protein response (UPR), meaning that the protein folding by taurine can be improved.

In this paper, by searching the relevant literature on the study of dual targeting therapy for HER-2 positive breast cancer in recent years, we summarized and analyzed the long-term follow-up results and new research on neoadjuvant, adjuvant and late rescue therapy of HER-2 positive breast cancer with anti-HER-2 dual targeting regimen, so as to further improve the clinical understanding of anti-HER-2 dual targeting therapy. Many research results show that anti-HER-2 double targeted therapy can achieve higher pathological complete remission rate and better prognosis in neoadjuvant therapy for HER-2 positive breast cancer, and also can obtain better survival benefit in adjuvant therapy and late rescue therapy. However, more large-scale prospective clinical studies and time validation are still needed to find the best targeted drug combination with more benefits, fewer complications and more economy.

Acute promyelocytic leukemia, a hematologic malignancy that predominantly affects young adults, has a relatively high mortality rate. Although the application of all-trans retinoic acid (ATRA) therapy over the last decades has significantly improved the long-term survival rate of APL patients, this method of treatment is often accompanied by severe complications such as RAS. This study investigates the effect of using milder traditional Chinese herbal medicine Salvia Miltiorrhiza Bunge, containing tanshinone IIA as an active compound, to treat APL with minor adverse reactions. This experiment will use a known human acute promyelocytic leukemia cell line (NB-4). After treatment with tanshinone, measure phospho-AKT and AKT by western blot, indicating activity of apoptosis. The positive control is taxol, and the negative control is PBS/DMSO. Differentiation of NB4 will be measured by FACS for CD11b and CD11c. The positive control is ATRA, and negative control is no treatment of tanshinone IIA.

With the increasingly diverse needs of the elderly for elderly care services in the new era and the continuous deepening of informatization construction in the field of elderly care, smart healthcare, as an important component of smart elderly care, is being built in full swing. Based on the data from 317 valid questionnaires of elderly citizens over the age of 60 in the five main urban areas of Harbin, taking Andersen's Behavioral Model as the theoretical framework, this paper discusses the factors that affect the use of smart healthcare by the elderly, including propensity factors, enabling factors, and demand factors. The analysis results show that propensity factors (residence mode, understanding of smart elderly care), enabling factors (monthly income), and demand factors (health condition, loneliness) have different degrees of influence on the elderly's demand for smart healthcare. Relevant government departments should continue to promote smart healthcare, such as increasing the publicity of smart elderly care, focusing on the needs of low-income elderly living alone for smart healthcare and improving the awareness of disease prevention among the elderly.

Correct classification and diagnosis of vascular anomalies is an important prerequisite for further clinical treatment. In general, clinical diagnosis of vascular anomalies is conducted according to different characteristics of ultrasonography. However, the clinical presentation of infantile hemangioma (IH) and venous mal- formation (VM) is similar and not easy to be separated. The difficulties result from the special diagnostic ultrasonography mode, indistinguishable grayscale images, rich texture, extensive involvement area, blurred boundary, and the unique blood flow ultrasonography acquisition method of VM. Here we propose a deep learning algorithm for detection and recognition of IH and VM lesions from ultrasonography. The experimental results have shown a good recognition performance on IH and VM ultrasonic images.
