Effect of Newly Characterized Probiotic Lactobacilli on Weight Gain, Immunomodulation and Gut Microbiota of Campylobacter jejuni Challenged Broiler Chicken 新型益生菌乳酸杆菌对空肠弯曲杆菌挑战肉鸡增重、免疫调节和肠道微生物群的影响

Received: Revised: Accepted: Published online: January 08, 2019 February 12, 2019 February 13, 2019 April 06, 2019 The aim of the present study was to determine the competitive exclusion of Campylobacter jejuni in poultry gut by three potential probiotic lactobacilli strains. A total of 135 birds of one day age were randomly divided into nine groups. The groups of prevention model received respective strains from day 1-35 and treatment model received lactobacilli from day 15-35 (after challenging with C. jejuni). These groups were given lactobacilli (~10 CFUs) while challenged with C. jejuni on day 14 by oral gavage (10 CFUs/bird). There were three control groups including A, B and I. Negative control (A) did not receive any treatment, C. jejuni was given to group B and group I was given Enrofloxacin formulation. Cloacal swabs were collected from birds of each group before and after challenge while the ceca were collected from birds after slaughtering (on day 35) for enumeration of aerobic bacteria, coliform, C. jejuni, lactobacilli and Bifidobacterium on selective agar plates. Effect of lactobacilli on weight gain and New Castle Disease vaccine (NDV) titer were also evaluated. Probiotic strain Lactobacillus gallinarum PL 53 considerably decreased log10 values of aerobic plate count (3.19±0.66), coliform count (2.83±0.22) and C. jejuni (3.98±0.77) in poultry. The probiotics also enhanced Lactobacillus and Bifidobacterium spp counts (~2-3 log increase). Treatment group C had maximum weight gain (1994±188.32g) and geometric mean titer (274.4) on day 28. The results of in-vivo experiments concluded that probiotic administration may be effective for targeted mitigation of C. jejuni in broiler birds. ©2019 PVJ. All rights reserved

In rats, spontaneous testicular tumours, particularly Leydig cell tumours, are very rare. The aim of the study was to carry out a histological and immunohistochemical analysis of a Leydig cell tumour in a rat as well as to present the similarities and differences in the expression of the proteins used to diagnose this type of tumour in dogs and men. Following the histopathological and immunohistochemical analysis (including antibodies against vimentin, calretinin, inhibin-α, β catenin and E-cadherin), significant similarities were found in the level of expression of the studied cell markers of the rat, dog and male leydigoma This indicates their usefulness as diagnostic markers of testicle tumours both in humans and animals. ©2019 PVJ. Antidiabetic Activity of Date Seed Methanolic Extracts in Alloxan-Induced Diabetic Rats 椰枣籽甲醇提取物对四氧嘧啶糖尿病大鼠的抗糖尿病作用

L. )date palm ( is one of the main fruits in North of Africa and Middle East. The date seeds are considered as wastes despite of their medicinal properties. Different seed preparations are used as expectorant, anti-diarrheic, hypoglycemic, tonic, and aphrodisiac agents. We established the anti-diabetic and antilipidemic activities of methanolic extracts of date seed in alloxan-induced diabetic rats. Animals were divided in to 6 groups (each group contained six animals), comprising of control group and diabetic rats. Animals were treated with different amounts of extract. To assess the hypolipidemic, and anti-hyperglycemic activity, cholesterol serum levels, low density lipoprotein (LDL), high density lipoprotein (HDL), creatinine, urea and alkaline phosphatase were measured in serum of the treated animals. The anti-diabetic potential was investigated through the levels of glucose and body weight. Moreover, to assess the extract’s safety, acute toxicity test was performed. In comparison with control group, significant reductions were observed in LDL, cholesterol, as well as blood glucose levels in diabetic rats that received date seed extract. Extract supplemented diabetic rats also showed a better tolerance to glucose, as well as reduced amounts of creatinine, urea, and alkaline phosphatase in their serum samples. In addition, no toxicity (acute toxicity), was detected even after high dosage of extract administration. Our results confirmed the anti-lipidemic and anti-diabetic potentials of date seed methanolic extract in diabetic rats. ©2019 PVJ. All rights reserved

Effect of Recombinant Bovine Somatotropin (rbST) and Oxytocin on Health Biomarkers, Reproductive Performance and Milk Composition of Nili-Ravi Buffaloes (Bubalus bubalis) 重组牛生长激素(rbST)和催产素对尼利-拉维水牛健康生物指标、繁殖性能和乳成分的影响

That is why this study was conducted to check its effect on health and production in buffaloes. Selected buffaloes were divided into groups (n=10) on the basis of body condition scores (BCS) i.e. (Low) BCS 1-3, (Medium) BCS 4-6 and (High) BCS 7-9, while ten animals served as control. All the groups received rbST (Boostin-250) 35 mg on alternate days and oxytocin 1 ml (10 I.U), starting on day 70 postpartum till the end of lactation. Blood samples were collected from each animal and serum was separated. Milk samples were used for milk composition parameters. The data on reproductive performance parameters was obtained from the farm record. The results indicated that mean serum Alanine aminotransferase (ALT), Aspartate aminotransferase (AST) and total oxidant status (TOS) values were higher (P≤0.01) during summer, while total antioxidant status showed opposite trend. Mean serum AST and TOS values were significantly (P≤0.01) higher in medium and high BCS buffaloes. Percentage of pregnant buffaloes was high in the high BCS group during summer. Mean milk fat contents were higher (P≤0.01) in buffaloes of high BCS while mean milk protein and lactose contents were significantly (P≤0.01) higher in medium BCS buffaloes. Fat, protein, lactose and solid not-fat (SNF) contents were higher during spring than summer. Mean milk lactose and SNF contents were higher (P≤0.01) in high than low and medium milk production groups. In conclusion, milk production was increased during summer and with rbST and decreased with oxytocin during spring, rbST and oxytocin treatments showed adverse effects on general health of Nili-Ravi buffaloes. ©2019 PVJ. Persistent survival inside macrophages is a major cause of chronic infections with Brucella. Leucine-rich repeat kinase 2 (LRRK2), a large multidomain protein kinase, is associated with Parkinson’s disease, chronic inflammation and intracellular infections. Hence, we monitored the activation of the LRRK2 kinase in Brucella-infected cells and evaluated the function of LRRK2 kinase in the intracellular survival of Brucella. Our results show that Brucella abortus 2308 activated the LRRK2 kinase and that the kinase activity was inhibited by a LRRK2-specific inhibitor in a dose-dependent manner. LRRK2 silencing significantly increased the Th1 immune response and reduced the replication of Brucella abortus 2308, both in vitro and in vivo. LRRK2 also enhanced the phosphorylation level of Akt, thereby inhibiting Akt-mediated humoral immune responses during Brucella infection. Collectively, these findings confirm that LRRK2 acts to reduce innate immune responses by activation of PI3K-Akt pathway, thereby contributing to the intracellular survival of Brucella abortus 2308 in murine macrophages and in a mouse infection model. Therefore, LRRK2 kinase play a key role in infections with Brucella abortus 2308, which will provide new insights into the pathogenic mechanisms used by Brucella. ©2019 PVJ. All rights reserved

Pharmacokinetics of Cefquinome in Layer Birds Following Intramuscular and Intravenous Administration 肌肉和静脉给药后头孢醌在蛋鸡体内的药代动力学

This study was conducted to evaluate the pharmacokinetics of Cefquinome after intramuscular (IM) and intravenous (IV) administration in layer birds for its safety evaluation. Twelve healthy layer birds were randomly allocated in two equal groups. Each group was administered with 5mg/kg b.w. of Cefquinome by intramuscular and intravenous routes. Highly sensitive high performance liquid chromatography with ultraviolet detection (HPLC-UV) method was developed for quantification of Cefquinome in layer bird’s plasma with >80% recovery. The Limit of detection and quantification were 0.02 and 0.05μg/ml, respectively. The pharmacokinetic data showed that mean Area under the curve (AUC) after IM and IV administration were 7.838±0.165 and 11.729±0.346h×μg/ml, respectively. The maximum concentration (Cmax) following IM administration (4.525±0.129 μg/ml) was almost same the mean Cmax after IV administration (4.635±0.270μg/ml). Time to reach maximum concentration (Tmax) after IV and IM administration were 0.1h and 0.5h, respectively. Cefquinome sulphate had a relatively shorter half-life (1.19±0.14h) after IV dose administration. The shorter half-life depicts a rapid elimination. Total recovery after each administration was greater than 75%. The mean resident time after IM and IV administration was found to be 1.528±0.09h and 1.988±0.0311h, respectively. The bioavailability after IM administration was 66.84±2.05%. This study indicated that Cefquinome sulphate has favorable pharmacokinetics following both administrations in healthy layer birds which can help to form optimum dosage regimes thus ultimately leading to its use for eradication of various systemic and local infections. ©2019 PVJ. Antibacterial Activity of Recombinant Porcine β-Defensin 2 重组猪β-防御素2的抗菌活性

To assess its antimicrobial activity against Escherichia coli, Salmonella paratyphi A, Pseudomonas aeruginosa, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus pumilus, and Bacillus subtilis, liquid growth inhibition assay was used. The antibacterial activity was further kinetically evaluated against representative bacterial strains E. coli and S. aureus. The hemolysis activity of rpBD-2 was also checked against porcine erythrocytes. rpBD2 was expressed and purified successfully using yeast Pichia Pastoris expression system. The yield of final product was 9 mg per liter of the harvested supernatant. rpBD-2 possessed antimicrobial activity against E. coli, S. aureus, S. epidermidis, B. subtilis, and B. pumilus at 33.3-66.6 μM and killed all E. coli and S. aureus within 30 min. Negligible hemolytic activity was observed against porcine erythrocytes. We have successfully developed rpBD-2 as an effective antibacterial peptide against both gram-positive and negative bacteria. rpBD-2 will be a potential novel therapeutic agent against bacterial infection. ©2018 PVJ. Protective Effect of Probiotics in Combination with Vaccination on Antibody Response, Biochemical and Hematological Indices in Broilers 益生菌联合疫苗接种对肉仔鸡抗体应答、生化和血液学指标的保护作用

A total of sixty day-old broiler chicks were equally divided into four groups: Control, Probiotic, Vaccine and Probiotic + Vaccine. Probiotics were offered at 2g/500ml of water for 5 days/week up to 6 weeks. NDV vaccine was given by eye drops at 0.025ml/bird at day 7. At 21 day NDV Lasota vaccine was offered in drinking distil water at 0.50ml /bird. IBDV vaccine was administered by drinking water at 10, 22 and 30 day of experiment. Blood samples were collected at 15, 30 and 45 days to measure antibody response, blood chemistry and hematological parameters. Antibody titer against NDV vaccine and IBD vaccine showed decline in control and probiotic groups. In case of NDV vaccine, there was no significant difference in Vaccine group (GMT=log2 ) and Probiotic + Vaccine group (GMT=log2 ) at day 45 of experiment. For IBDV vaccine, there was significant difference at 45 day in ELISA titer of vaccine group (2233) and Probiotic+Vaccine group (4206). Cholesterol and ALT showed significant decrease in PV group at 45 day. Significant increase in WBC count was observed in PV group. Level of significance was P≤0.05. Probiotics when administered along with vaccine showed improvement in antibody titer but no harmful increase or decrease in blood chemistry and hematological parameters was observed. ©2018 PVJ. Received: Revised: Accepted: Published online: July 04, 2017 November 06, 2017 January 24, 2018 June 19, 2019 Fish culture in ponds with heated water offers better fish growth compared to farms in natural sea water temperature, but the increase in temperature favors the development of bacteria and fish disease. In this study, in European sea bass (Dicentrarchus labrax) specimens reared in ponds with heated water from a power plant showing symptoms of a vibriosis-like disease we aimed to characterize the bacterial community as well as some serum metabolic and immune parameters. Bacteriological analysis revealed an important contamination with fecal coliforms, Pseudomonas aeruginosa, Vibrio alginolyticus, yeasts and molds whilst fish tissues from both healthy and diseased specimens were positive for total aerobic flora and several potential pathogenic bacteria such as V. alginolyticus, Aeromonas hydrophila, Pasteurella multocida, P. aeruginosa, Proteus vulgaris and P. mirabilis. Additionally, Grimontia hollisae was only present in diseased fish tissues. Serum metabolic parameters such as aspartate aminotransferase, bile acids, creatine kinase, uric acid, glucose, globulin and potassium were generally increased in diseased fish though only the levels of phosphorous reached a significant level compared to the healthy specimens. Regarding fish innate immunity, our results showed no statistical differences between healthy and diseased fish on peroxidase, protease or antiprotease activity while the bactericidal activity against V. anguillarum was significantly decreased in diseased fish. In this study, in European sea bass (Dicentrarchus labrax) specimens reared in ponds with heated water from a power plant showing symptoms of a vibriosis-like disease we aimed to characterize the bacterial community as well as some serum metabolic and immune parameters. Bacteriological analysis revealed an important contamination with fecal coliforms, Pseudomonas aeruginosa, Vibrio alginolyticus, yeasts and molds whilst fish tissues from both healthy and diseased specimens were positive for total aerobic flora and several potential pathogenic bacteria such as V. alginolyticus, Aeromonas hydrophila, Pasteurella multocida, P. aeruginosa, Proteus vulgaris and P. mirabilis. Additionally, Grimontia hollisae was only present in diseased fish tissues. Serum metabolic parameters such as aspartate aminotransferase, bile acids, creatine kinase, uric acid, glucose, globulin and potassium were generally increased in diseased fish though only the levels of phosphorous reached a significant level compared to the healthy specimens. Regarding fish innate immunity, our results showed no statistical differences between healthy and diseased fish on peroxidase, protease or antiprotease activity while the bactericidal activity against V. anguillarum was significantly decreased in diseased fish.
