To understand the genetic diversity and genetic differentiation status of Craspedacusta sowerbyi Lankester, 1880 in Guizhou Province, genetic diversity and genetic structure analyses of three geographical populations of C. sowerbyi in Guizhou Province were performed using the results of PCR amplification and direct sequencing based on the mitochondrial DNA COI gene. Sixty-two individuals displayed four haplotypes, and the overall haplotype diversity (Hd) and nucleotide diversity (Pi) of the three C. sowerbyi groups were 0.531 and 0.00067, respectively. Analysis of molecular variance (AMOVA) showed that inter- and intrapopulation variation accounted for 41.63039% and 58.36961%, respectively, of the total variation, and the genetic variation coefficient (F st ) was 0.05268. The gene flow (Nm) values between the three C. sowerbyi populations were <1. The results of neutrality, Fu’s Fs and Tajima’s D tests did not show significant negative values. The overall nucleotide diversity of the three C. sowerbyi populations in Guizhou Province was low, especially in the Xiuwen County population, the genetic variation mainly occurred within the populations.

The Musca domestica is a serious hygienic pest of poultry, humans, and livestock facilities with the immense ability for resistance development against chemical insecticides. The current study aimed to evaluate the susceptibility and resistance status of M. domestica against different classes of insecticides. For this purpose, adult M. domestica populations were collected from five different localities of Sargodha division (Sargodha, Khushab, Jauharabad, Mianwali, and Bhakkar), Punjab, Pakistan, and tested against selected insecticides. The resistance ratios (RR) at LC 50 ranged from 10.32-35.37 folds for deltamethrin, 17.49-38.13 folds for fipronil, and 10.70-18.81 folds for chlorpyrifos. The RR values at LC 50 for imidacloprid and pyriproxyfen ranged from 4.35-28.0 and 10.56-21.45 folds, respectively. The study showed varying levels of resistance in M . domestica populations from area to area and from insecticide to insecticide. Therefore, to control resistance development in M . domestica from livestock facilities, inappropriate and excessive use of insecticides must be controlled through proper mechanisms and strategies.

Multiple sclerosis (MS) is a complex disorder, characterized by demyelination and loss of axonal parts of neurons in the central nervous system involving multiple genetic and environmental factors. Although, the demyelinated lesions develop throughout the brain, but more frequently are extensive in white matter. Currently, three different approaches are being utilized to treat MS, where synthetic drugs are the most frequently used but they do not cure the disease. Secondly, the stem cell therapy but this too has limited success in treating MS in humans. The thirds technique involving administering hormones has been found to be most effective method but this too have some significant side effects. Alternatively, natural products can potentially serve as an affordable and effective substitute for the treatment of MS with minimum or no side effects. Blackseeds ( Nigella sativa) and honey possessing potent neuroprotective, antioxidant, and anti-inflammatory properties with no reported side effects can be a prospective candidate for an alternate remedial treatment of MS in animal model as well as in humans. In this study we established a new animal model (quail), to assess the synergistic efficacy of honey and black seed against demyelination within brain. A total of 35 male quails were used, among 10 were non treated and 25 were treated with 200 mg/kg/day cuprizone (CPZ) demyelination for six months to induce demyelination. After that they were divided into seven groups of five animal each where 3 CPZ treated groups received either honey, black seed oil or mixture of both for 6 weeks after demyelination. Behavioral tests were performed at the end of treatment. Afterwards, oligodendrocyte population was estimated in cerebellar white matter after histology. It was found that all three treatments efficiently induce remyelination. Interestingly, the mixture of honey and black seed was significantly more efficient than honey and black seed alone. Our data support the need of clinical trials for administration of N. sativa and honey in MS patients.
