The main purpose of this present study is to evaluate the ability of near infrared technology as an alternative method in determining and assessing quality parameters of meat product where in this case is frozen beef. At first, beef samples from chest and legs parts were sliced and taken at the amount of 100g per sample to be frozen. Then spectra data of beef samples were obtained using near infrared spectrophotometer (PSD IR i16) in wavelength range from 1000 to 2500 nm with optical gain 4x. Actual protein contents were obtained by Kjeldahl method and measured in triplicate. The near infrared spectra data were enhanced and improved by means of mean centering (MC) and baseline shift correction (BSC) methods. The results showed that protein content of frozen beef samples can be predicted rapidly with maximum correlation coefficient is 0.91. Heat properties of beef samples changes exponentially during freezing and thus, optimum freezing temperature and time can be predicted as well. Based on those obtained results, it may conclude that near infrared technology can assess frozen beef qualities rapidly and effectively.
本研究的主要目的是评估近红外技术作为确定和评估肉制品质量参数的替代方法的能力,在这种情况下是冷冻牛肉。首先,从胸部和腿部部分的牛肉样品切片,每个样品取100g冷冻。利用近红外分光光度计(PSD IR i16)在1000 ~ 2500 nm波长范围内获得牛肉样品的光谱数据,光学增益为4倍。用凯氏定氮法测定实际蛋白质含量,并分三次测定。采用平均定心(MC)和基线偏移校正(BSC)方法对近红外光谱数据进行了增强和改进。结果表明,该方法可以快速预测冷冻牛肉样品的蛋白质含量,相关系数最大可达0.91。牛肉样品的热特性在冷冻过程中呈指数变化,因此可以预测最佳冷冻温度和时间。结果表明,近红外技术可以快速有效地评价冷冻牛肉的品质。

The purpose of this research was to determine the effect of decoction of pulutan leaves simplicia( Urenalobata L.) on the development of the uterus of Balb C mice ( Musmusculus ). Female mice aged 10-12 weeks were given decoction of pulutan leaves simplicia with concentration of 5%, 7.5%, 10%, 12.5%, 15%, and control. The administration of pulutan leaves was performed by gavage for 11 consecutive days. On day 12, mice were dissected, then, the uterus was taken for histological preparations using paraffin method. Data such as uterine diameter and myometrium and epimetrium thickness were analyzed using one way analysis of variance followed by least significant different. The results showed that decoction of pulutan leaves simplicia decrease the uterine diameter and epimetrium and myometrium thickness compared to control. The decrease of uterine diameter and uterine myometrium and epimetrium thickness was started at the concentration of 7.5%. Decoction of pulutan leaves simplicia could reduce uterine diameter and thickness of uterine myometrium and epimetrium, thus, it might decrease the uterine function as the site of implantation. The decline in the function of the uterus might decrease the number of born children, thus decoction of pulutan leaves simplicia is potential as antifertility agent.

Abstract The use of vaginal sponge in estrous synchronization has been a popular method to uniform estrous cycle of small ruminants. In order to increase the litter size, the use of the sponge is combined with PMSG. Therefore the objective of this study was to evaluate the effect of PMSG in combination with vaginal sponge on estrous response and litter size of Javanese local sheep. The study was conducted in Faculty of Animal Husbandry, Padjadjaran University, Indonesia with 32 non-pregnant local sheep. All these sheep was divided into four groups of treatment, i.e: 1) group of sheep was subjected to vaginal sponge (VS) treatment containing 30 mg Medroxyprogesterone acetat (MAP) inserted in the vagina for 14 days; 2) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection -1 d before VS removal; 3) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection at the day of VS removal; 4) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection +1 d after VS removal. The result showed that all treatment groups had 100% of estrous response. However, the occurrence of estrous time varied among group after VS removal. Group 1 (G1) and G4 tend to have estrous occurrence at day 3 after VS removal by 55% and 75% response, respectively. Whilst, G2 and G3 tend to have estrous occurrence respectively at day 1 (62,5%) and day 2 (50%) after VS removal. A 100% of pregnancy rate occurs in G2 and G4, while G1 and G3 have a 88,9% and 87,5%, respectively. No significant effects of the treatments on the ewes litter size. In conclusion, the use of PMSG administration 24 h prior, after, or at sponge withdrawal increase a tighter synchrony of oestrous and pregnancy rate, but has no effect on the increasing of litter size in Javanese local sheep. 摘要阴道海绵同步发情是目前小反刍动物发情周期的一种常用方法。为了增加垃圾大小,海绵的使用与PMSG相结合。因此,本研究的目的是评估PMSG与阴道海绵联合使用对爪哇当地绵羊发情反应和产仔数的影响。这项研究是在印度尼西亚帕贾贾兰大学畜牧学院对32只未怀孕的当地绵羊进行的。将这些绵羊分为四组,即:1)绵羊组接受阴道海绵(VS)治疗,阴道内插入30mg甲羟孕酮(MAP),为期14天;2) 绵羊组在VS去除前1天接受30 mg MAP VS加300 IU PMSG注射;3) 绵羊组在VS去除当天接受30mg MAP VS加300IU PMSG注射;4) 绵羊组在VS去除后+1天接受30 mg MAP VS加300 IU PMSG注射。结果显示,所有治疗组的发情反应均为100%。然而,VS切除后各组发情时间不同。第1组(G1)和G4倾向于在VS去除后第3天出现发情,分别有55%和75%的应答。而G2和G3倾向于分别在VS去除后的第1天(62.5%)和第2天(50%)出现发情。G2和G4的妊娠率为100%,而G1和G3的妊娠率分别为88.9%和87.5%。处理对母羊产仔数无显著影响。总之,在拔出海绵前、拔出海绵后或拔出海绵时24小时使用PMSG可提高发情率和妊娠率的同步性,但对爪哇当地绵羊产仔数的增加没有影响。关键词:阴道海绵,PMSG,发情反应,妊娠率,产仔数,绵羊

virus was isolated from suspected Newcastle disease (ND) chicken in backyard farm at Sibang Village, Badung Regency, Bali Province. The isolates were then propagated and confirmed for NDV serologically. RNA isolation was performed by standard Trizol method. Phylogenetic tree analysis of NP gene (nt1020-1561) and HN gene (nt7019-7754) were performed using sequence of Badung-02/AK/14 and selected NDV strains from GenBank. Based on the NP gene sequence, the newly isolate closely related with other NDV strains belong to genotype VII that previously isolated such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 with genetic distance 0.2%, 12.6%, and 18.0% respectively. The genetic distance with LaSota/46 virus (genotype II) is 19.6%. Based on HN gene sequence, genetic distance of the Badung-02/AK/14 with other viruses belong to genotype VII such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 are 0.4%, 3.7%, and 4.2% respectively. The genetic distance with LaSota/46 virus (genotype II) is 9.0%. There were no difference between the result of nucleotide and amino acid sequence analysis, both in NP and HN gene (P>0.05).

This research purpose was to study the phenotype characters of body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1) derived from crossbreed between parent stock broiler strain Cobb-500 and Pelung (Gallus gallus gallus). The chicks were divided into 3 groups and each group consists of 5 days-old chicks (DOC). Group 1 (T1) was used as control involving DOC derived from crossbreed between female and male of broiler Cobb-500 strain. Group 2 (T2) involving DOC derived from crossbreed between female and male Pelung, while Group 3 (T3) was DOC derived from crossbreed between female broiler strain Cobb-500 and male Pelung. The chicken was grown up to 49 days to evaluate the body weight, the pectoralis thoracicus weight, the pectoralis thoracicus area, and myofiber area. Data were statistically analyzed using one way Anova. The results showed that the average of body weight in T1; T2; and T3 were 2,170±86.02; 506.2±20.02; and 1,238±68.25 g, respectively (P<0.05). The average of pectoralis thoracicus muscle weight in T1; T2; and T3 were 165.04±11.97; 21.26±2.44; and 68.74±1.94 g, respectively (P<0.05). The average of pectoralis thoracicus muscle area in T1; T2; and T3 were 97.0±9,7; 61±6,4; and 63,4±1,8 cm2 , respectively (P<0,05). The average of pectoralis thoracicus myofiber area in T1; T2; and T3 were 14.72±1.04; 4.66±2.1; and 13.13±1.3 µm2 , respectively (P<0.05). It was concluded that crossbreed between broiler strain Cobb-500 and Pelung improved the body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1).

The aim of this research was to analyze the anti-oxidant activities of ethanol extract of mahogany seeds on the liver tissues of diabetic experimental rats to overcome oxidative stress condition. This study used male Rattus norvegicus strain Sprague Dawley which were divided into 5 groups: (i) negative control group (K-), (ii) positive control group/diabetes mellitus (DM) (K+), (iii) DM group treated with 500 mg/kg BW ethanol extract of mahogany seeds (EM), (iv) DM group treated with acarbose (KO), and (v) non-DM group treated with 500 mg/kg BW ethanol extract of mahogany seed (KE). Alloxan with dose of 110 mg/kg BW was used to induce diabetes in rats. The treatments were conducted for 28 days. At the end of treatment, the liver tissues were collected and analyzed for malondialdehyde (MDA) content using thiobarbituric acid reactive substance (TBARS) method and anti-oxidant cooper zinc superoxide dismutase (Cu,Zn-SOD) content using immunohistochemical technique. The result showed that level of MDA were significantly different (P<0.05) in K-; K+; EM; KO; and KE with values were 0.81±0.17; 1.19±0.26; 1.10±0.04; 0.95±0.13; and 0.92±0.0 µmol/gram, respectively. The anti-oxidant content (Cu, Zn-SOD) of rat liver tissue in K+ lower than in K-, while anti-oxidant content in EM and KE were higher compared to K+. The study concluded that ethanol mahogany seeds extract decreased blood glucose levels and increased the anti-oxidant status in the liver tissues of diabetic rats.

At the end of treatment, the liver tissues were collected and analyzed for malondialdehyde (MDA) content using thiobarbituric acid reactive substance (TBARS) method and anti-oxidant cooper zinc superoxide dismutase (Cu,Zn-SOD) content using immunohistochemical technique. The result showed that level of MDA were significantly different (P<0.05) in K-; K+; EM; KO; and KE with values were 0.81±0.17; 1.19±0.26; 1.10±0.04; 0.95±0.13; and 0.92±0.0 µmol/gram, respectively. The anti-oxidant content (Cu, Zn-SOD) of rat liver tissue in K+ lower than in K-, while anti-oxidant content in EM and KE were higher compared to K+. This study was conducted to evaluate fertility grade of Friesian Holstein cows based on milk urea levels. A number of eight cows which fulfill the criteria of not pregnant and in second to sixth lactation, produced milk of 2.5 liters/day, and body weight ranging from 400-450 kg were used in this research. The cows were given fodder that consisted of 30 kg of forage and 10 kg of a mixture of concentrate (pollard, pulp out, and molasses), in the morning and afternoon. Milk and blood samples were taken once a week for five weeks in the morning before fed. Milk urea levels were measured using the urea FS (DiaSys) and blood samples were tested using enzyme linked immuno-sorbent assay (ELISA) with the progesterone hormone kit (DRG, Germany). The variables measured were pregnancy rate, service per conception (S/C), days open, and progesterone levels. Reproduction and milk urea levels data were analyzed by linear regression to determine the relationship between two variables. Progesterone data were analyzed descriptively. The results showed that the average calving interval, days open, and S/C were 607.5±170.27 days; 341.25±187.65 days; and 2.41±2.32 days, respectively. The average of milk urea levels was 14.16±2.55 mg/dL. Regression analysis showed a significant effect (P<0.05) and negative correlation between milk urea level and S/C. The average of non-pregnant progesterone hormone level was 12.04±7.13 ng/mL. In conclusion, the fertility grade of Friesian Holstein cows decreased when the milk urea level at 14.16 mg/dL and non-pregnant progesterone at 12.04±7.13 ng/mL.

Three intestinal bacteria isolates (namely C1, C2, and C3) from common carp (Cyprinus carpio) were isolated and identified as new potential probiotics. This research aimed to study the effect of three isolates potential probiotics addition on growth performance, survival rate and feed efficiency of common carp. Completely randomized design was assigned to fourth diet treatments with five replicates. As a control, we used the feed without any supplemention of potential probiotic. The result showed that the addition of potential probiotic bacteria (C1, C2, and C3) gave significant effect on daily growth rate, survival rate and feed efficiency and could be used in aquaculture industry to reduce feeding cost.

This study was aimed to determine the influence of the washing method under running water on nitrite levels of edible bird’s nest (EBN). Total of 40 samples of EBN were divided into four groups with different washing frequency, control group without washing treatmet (P0), once, twice, and three times washing treatment (P1, P2, and P3) respectively. Each washing was performed for 30 seconds under running water. Nitrite levels assessment was carried out by spectrophotometry at 540 nm of wavelength. The results showed that the average nitrite levels of EBN in P0, P1, P2, and P3 were 93.12±4.40 ppm, 65.24±3.38 ppm, 63.60±3.81 ppm, and 30.87±2.11 ppm, respectively. The nitrit level in edible bird’s nest decreased significantly (P<0.05) by using three times washing.
