Objective: The zinc finger E-box binding homeobox (ZEB2), which can accelerate the nuclear DNA replication by inducing the activation of upstream transcription promoters, was widely considered as an oncogene. Recent study has found that the overexpression of ZEB2 is associated with a better prognosis in hepatocellular carcinoma. However, its roles in tumor growth, metastasis, and immunology are yet to be elucidated in COAD. Methods: The pan-cancer sequencing data was acquired from The Cancer Genome Atlas (TCGA)-Pan cancer cohort, normal human tissue data was acquired from the Genotype-tissue expression (GTEx) database, and Broad Institute Cancer Cell Line Encyclopedia (CCLE) were downloaded from UCSC Xena. We used the cBioPortal webtool to analyze and visualize the ZEB2 pan-cancer genomic alteration rate. GEO Expression Datasets were used to explore ZEB2 expression levels in COAD patients. UCSC Xena database was used to download prognostic information of COAD patients. The Cox regression and Kaplan–Meier analyses were used to assess the prognostic role of ZEB2 in COAD. Kyoto Encyclopedia of Genes and Genomes (KEGG) was performed to determine the biological pathways. Gene Ontology (GO) enrichment analysis was performed to determine the biological processes, molecular functions, and cellular components that were altered in a ZEB2-dependent manner in COAD. The module analysis of PPI interaction network was performed using the MCODE tool of Cytoscape software, and the characteristic molecules were selected by cytohHubba tool. CIBERSORTx database was used to analyze the ZEB2 expression in the presence of 22 types of immune infiltrating cells. Results: This study found that ZEB2 was aberrantly expressed in most cancer types, and it was significantly downregulated in COAD compared with normal tissue. In addition, our findings also show that overexpression of ZEB2 was associated with a better prognosis in COAD. Mechanistic analysis revealed that overexpression of ZEB2 was associated with the neutrophil extracellular trap formation in COAD. And the results show that ZEB2 expression was significantly correlated with several kinds of immune cell infiltration. Conclusion: This study demonstrates that overexpression of ZEB2 was associated with better prognoses in patients with COAD. ZEB2 has close relationship with ACTB, which was highly related to NETs. These findings suggest a dual role of ZEB2 in COAD growth, metastasis, and immunology.
方法:泛癌测序数据来源于The Cancer Genome Atlas (TCGA)-Pan Cancer cohort,正常人组织数据来源于gene - type-tissue expression (GTEx)数据库,Broad Institute Cancer Cell Line Encyclopedia (CCLE)来源于UCSC Xena。

Objective: To evaluate the effects of lactulose for bowel preparation prior to colonoscopy in adults. Methods: By searching for literature pertaining to RCT (randomized controlled trial) of bowel preparation with lactulose in adults up to 10th of July, 2021. The experimental group utilizes lactulose oral solution as the solution of choice for bowel preparation while the control group uses the standard bowel preparation solution. Literature selection and data extraction were completed independently by 2 researchers, including literature quality control. Data analysis was completed using the software RevMan5.2. Results: 11 RCT was included for data analysis. Meta-analysis showed that the lactulose group had a lower incidence of adverse effects [OR=13.58, 95CI% (6.10, 30.24), p<0.0001] and better tolerability [OR=13.58, 95CI% (6.10,30.24), p<0.0001] when compared with the control group. However, the bowel cleanliness was statistically insignificant between the two groups [WMD=0.26,95%CI (-0.31,0.82), p=0.38]. Conclusion: according to current evidences, as a solution for bower preparation, lactulose has a lower incidence of adverse effects such as emesis or diarrhea, while effectively increasing tolerability, however it is not a superior solution for bowel preparation in terms of bowel cleanliness when compared with other standard choices. More high quality researches will be required to support its use.

Prostate cancer is a type of malignancy that is defined by abnormal development of cells in the prostate tissue. Prostate cancer needs early intervention since its incidence and prevalence is high across the world leading to high morbidity and mortality. Prostatic specific antigen test which is the commonly used screening test in Kenya and across the world is nonspecific, expensive and inaccessible to many people in rural setting who are in need. The definitive histological test is invasive and requires specialized facilities and personnel. This study sought to investigate sarcosine in urine as a predictor of prostate cancer to supplement prostatic specific antigen test in the diagnosis of prostate carcinoma. Cross sectional study design was employed in this study for all suspected prostate cancer identified according to clinical assessment during the study period. Midstream urine samples of about 30mls was collected in plastic tubes, centrifuged and supernatant collected and analyzed using ELISA method for sarcosine. Raw data obtained was tabulated in excel and transferred to statistical package for social science. Differences in means and standard deviation from various age groups was analyzed using one-way Anova and Independent t test. The Bonferroni was used as post Hoc to test the means that were significant from others. Significance level was set at 95%. The concentration of sarcosine (4.30±0.11nmol/ml) in prostate cancer participants was significantly higher than the concentration (0.47±0.06nmol/ml) of control participants using ELISA (p<0.001;). Hence Sarcosine in urine needs to be analyzed for the testing of prostate tumor since it is raised in confirmed prostate carcinoma participants as compared to negative control units. The age groups of the prostate tumor participants had no significant variation in sarcosine concentration using ELISA method (p=0.57). Similarly, the age groups of the control individuals were not significantly different in sarcosine concentration (p=0.17). Future studies need to dwell in incorporating sarcosine metabolite in urine.

Due to the uncontrolled increase in the administration of antibiotics, the prevalence of multidrug-resistant organisms in patients admitted to intensive care units has also increased. Thus, this study aimed to investigate the pattern of sensitivity and compliance of treatment with the results of culture in gram-negative MDR bacteria in patients admitted to the intensive care unit of Shahid Sadoughi Hospital in Yazd. This cross-sectional study was performed over a period of one year on the results of culture of patients admitted to the intensive care unit of Shahid Sadoughi Hospital in Yazd (2020). The samples included blood, urine, wounds, bronchoalveolar lavage, shunt, and throat. All patient information and antibiotics prescribed were obtained using their records. Finally, the data was entered into SPSS (v. 26) and analyzed. In total, out of 298 patients for whom initial antibiotics were started experimentally, 226 cases had the culture results consistent with one of the antibiotics that were started initially. Among the antibiotics that were used more, clindamycin showed the least agreement with antibiotic sensitivity results. Among antibiotics, clindamycin had significantly the most compatibility in blood and urine and the least compatibility in wound. The present study showed a relatively good agreement in the initial experimental administration of antibiotics.
