In order to determine the pathogen of the skin rot disease of Rana spinosa and provide theoretical basis for the prevention and control of the disease in production, this study isolated the pathogenic bacteria from the diseased frog focus, carried out artificial infection and the morphological characteristics, physical and chemical characteristics and molecular biological analysis of the pathogenic bacteria, and carried out drug sensitivity test and vaccine immunity and control test. The results showed that the dominant bacteria QS01 isolated from the diseased lesions of Rana spino-sa was confirmed to be the pathogen of skin rot disease of Rana spinosa by artificial regression infection test. Combined with its phenotypic characteristics and 16S rRNA, gyrB gene sequence analysis, it was determined to be Citrobacter freundii . The medicine sensitive test result showed that the bacteria is sensitive to eight kinds of antibiotics including Enrofloxacin, Chloramphenicol, Ceftazi-dimeetc, but lowly sensitive or resistant to other tested antimicrobial agents. Adopting these ways included injection, immersion and atomizing to inoculate frog with vaccine named QS01 inactivated by formaldehyde, average titer of three immunity groups reached the peak in the 20 d. The highest serum antibody titers were 1:64~128 (101.6), 1:16~32 (20.2) and 1:16~32 (16), which were significantly higher than those of the control group. The immune protection rate for Q. spi-nosa after injection, immersion and spray of QS01 inactivated vaccine can reach 100%, 85.71% and 71.43%. Our research shows that Citrobacter frederii is the pathogen of skin rot disease of Rana spinosa , and it is effective and feasible to use vaccine to control ulcerative skin disease.

Objective: To detect the expression levels of cytokines secreted by immune cells in patients with chronic hepatitis B (CHB), liver cirrhosis (LC) and hepatocellular carcinoma (HCC), and to investigate their correlation with clinical indicators, and in order to provide new for clinical diagnosis and treatment. Methods: 106 cases of CHB, 96 cases of LC, 21 cases of HCC from March 1, 2020 to May 31, 2020, in the third people’s Hospital of Kunming. The changes of liver function indexes such as ALT, AST, TBIL, IBIL and DBIL were detected by automatic biochemical analyzer. The expression of cytokines IL-1 β , IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL-12P70, IL-17, TNF-α , IFN-γ and IFN-α in peripheral blood was detected by multiple microsphere flow immunofluorescence assay. And the correlation between the above liver function indexes and cytokines was analyzed. As comparison, 100 healthy people served as normal control group. Results: The expressions of AST, ALT, TBIL, DBIL and IBIL in CHB, LC and HCC groups were higher than normal control group. Excepted IL-5, IFN-α and IL-6, the expression of IL-2, IL-1 β , IFN-γ , IL-8, IL-17, IL-12P70 and TNF-α in CHB, LC and HCC group was significantly higher than normal control group ( P < 0.05), while the expression of IL-4 and IL-10 was significantly lower than normal control group ( P < 0.05). There were significant differences in TNF-α and IL-4 among CHB, LC and HCC groups ( P < 0.05). Cytokines IL-17 and TNF-α were significantly correlated with AST, ALT, TBIL, DBIL and IBIL ( P < 0.05). Conclusion: The secretion of pro-inflammatory cytokines (IL-1 β , IL-2, IL-5, IL-6, IL-8, IL-12P70, IL-17, TNF-α , IFN-γ , IFN-α ) increased, while the secretion of anti-inflammatory cytokines (IL-4, IL-10) decreased in CHB, LC and HCC. Enterobacter sakazakii and Staphylococcus aureus are common detection items with high pathogenicity in dairy products. In this study, we first achieved effective enrichment of target bacteria through immunomagnetic separation, realized signal amplification and sensitive detection of Enterobacter sakazakii and Staphylococcus aureus through quantum dots and biotin streptavidin system combined with immunochromatography. By optimizing the overall research, both bacterial detection sensitivities can reach 2 × 10 3 CFU/mL, which increases the detection sensitivity by nearly 2000 times compared to traditional immunochromatography. It can achieve simultaneous sensitive detection of foodborne pathogens, providing a new approach for food safety to quickly, sensitively, and accurately detect large molecular targets.
