The DNA content in nuclei of germ cells: spermatogonia B and spermatides in the maturation phase of male mice receiving commercial insecticide preparations commonly used in this country was cytophotometrically measured. The following insecticides were tested: 1% Metox-30 solution, containing 30% of methoxychlorine (p,p-dimethoxydiphenyltrichlorethane), 0,3% solution of Sadofos-30 containing 30% of malathione (1,2-dicarboethoxyethyl-0,0-dimethyldithiophosphate) and 0,3% solution of Foschlor-50 containing about 50% trichlorfon (2,2,2-trichloro-1-hydroxyethyl-0,0-dimethyl-diphosphonate). It was found that a statistically significant number of spermatogonium B and spermatide nuclei in the phase of maturation of the animals treated with the insecticides exhibited an abnormal DNA content as compared with the nuclei of control animals. The chromatin of these nuclei is also more sensitive to acid hydrolysis in Feulgen reaction.

Observations of the cytoskeleton visualized with Coomassie brilliant blue R250 were carried out on chick embryo fibroblasts emigrating from cell aggregates onto clean or siliconized cover glasses. Conditions for persistent isometric contraction were created in cells attached strongly to glass and to the aggregate. These cells showed a presence of abundant stress fibres in their cytoplasm. Upon induction of lamellipodia retraction, the stress fibres transformed into typical retraction fibres. In cells spreading onto siliconized glass, the fibres were rare. Upon cell contraction their lamellipodia detached, folded and fibres, if originally present, disintegrated. These observations extend the conclusions of Wohlfarth-Bottermann to tissue culture cells, formulated on the basis of extensive study on the structure and behaviour of cytoplasmic fibrils in slime mold plasmodia, that bundles of parallely oriented actomyosin filaments arrange as a result of the persistent isometric contraction of cytoplasm. The present result point out the significance of cell surface adhesive properties for the organization of cell cytoskeleton.

The rapid, two steps method of purification of an elastase-like proteinase from cytoplasmic granules of horse leucocytes is described. This enzyme called the proteinase 1 is released easily from isolated granules in the low ionic strength solutions in opposite to the other two molecular forms of which one differs slightly in isoelectric point from the other. The enzyme is a typical neutral proteinase of a broad substrate specificity and wide pH optimum. In a physiological conditions the enzyme is built of two polypeptide chains of molecular weight about 30000 and 20000, respectively.

On the basis of measurements taken, the antibiotic was shown to cause a statistically significant fall of the alkaline phosphatase activity and the enzyme activity was found to decrease with consecutive doses of the drug under investigation.

The binding affinity of 125I-TAA(tumor associated antigen) to in vitro restimulated TAA-primed lymph node cells (LNC) from CBA mice was studied by using saturation analysis. Differences between values of equilibrium constant K of antigen-lymphocyte reactions were found for in vitro TAA-stimulated TAA-primed and unprimed lymphocytes. On the other hand it has been established that TAA binding affinity increased not only as a result of TAA-specific lymphocytes restimulation but as a result of nonspecific Con A stimulation as well. The overall conclusion from the study is that saturation analysis might have been a useful tool in a preliminary monitoring of in vitro lymphocytes-antigen interactions but for the explanation of these phenomena others methods would be also used.

Histochemical observations were made on the activities of 17 beta-hydroxysteroid dehydrogenase (HSDH), 3 beta-hydroxysteroid dehydrogenase (3 beta-HSDH) and 3 oC-hydroxysteroid dehydrogenase (3 oC-HSDH) in the preputial glands of normal and isoproterenol treated animals. Observed decrease in the activities of all the three HSDH's are correlated with the possibility of decreased biological effectiveness of circulating steroids after isoproterenol treatment.

The following examinations were made: radiological examinations of the jaws, a teeth analysis in the microanalyser for the distribution of fundamental elements, and ultrastructure observations in the scanning electron microscope. The jaws of the experimental rats were smaller in the radiological picture. The disturbed arrangement of the enamel prisms was found in the enamel substructure of the experimental rats. Phosphorus values in the enamel and dentine were lower in the experimental animals. Calcium to phosphorus ratio was higher in the rachitic rats. Disturbances of calcium-phosphorus balance were observed.

It is commonly accepted that hormonal disturbances in experimental animals produce pathological changes in several organs namely in mammary glands and kidney. Taking into consideration the close relationship between acid phosphatase and Golgi apparatus, the present study was undertaken, to study the relationship in kidney and mammary gland cells under the influence of DES. The observation indicate, that acid phosphatase and Golgi apparatus were localized in the cytoplasm of mammary gland and kidney cells. It has also been found a parallel change in the activity of acid phosphatase and the degree of development of Golgi apparatus after DES administration.
