The technique of the power ultrasound was tested in vitro and in vivo in 20 porcine pancreas. Simultaneous temperature measurements and laboratory tests do not produce any restriction. Enzymatic disorders were provable adequate to the clinical course. The ultrasonic effect guarantees a nearly riskless dissection in the layer. Joining processes, by adding Ligament-FIMOMED, produced a waterproof occlusion of parenchymal wounds. The cross-section of the pancreas with the pancreatic duct was sealed sufficiently. Also the jointed pancreato-jejunostomy remained sufficient. The reaction of the pancreatic parenchyma to the power ultrasound was small. A scarred metaplasia evolved immediately at the adhesive as a chronic atrophic interstitial pancreatitis. Only in case of occlusion of the pancreatic duct this alteration comprehended the whole pancreas.

We studied the respiratory (R) and cardiovascular (CV) effects of 1.25 MAC halothane (H), isoflurane (I) and enflurane (E) in oxygen, delivered by the Jackson-Rees breathing system in 5 normothermic rats. Mean arterial pressure (MAP), heart rate (HR) were depressed after 30 minutes than stabilized after 2 hours anaesthesia regardless of the inhalational agent. Respiratory rate (RR) slowed down markedly unter I and E anaesthesia. Significant R depression was developed (PCO2 53.5 +/- 2.44 Torr) under E. Stabil metabolic condition was maintained. The R and CV changes caused by H and I were far less than changes produced by E. H or I is preferable to E for small animal anaesthesia with spontaneous breathing. Maintenance of constant temperature minimizes the CV and R disturbances.

Measurements of the ultrasound attenuation were carried out in 10 normal testes and 20 pathologically changed testicular tissue biopsies in vitro after formalin fixation. The ultrasound attenuation as function of the frequency was recorded ultrasound-spectroscopically about the complete frequency spectrum of the ultrasound-broad range transformer (3-8 MHz). The frequency dependence of the attenuation behaviour differed in the 10 normal testes from the 20 pathologically changed biopsies by smaller values of the ultrasound attenuation coefficient a (linear regression analysis). The ultrasound attenuation in a characteristic measuring point allows the differentiation between normal and pathologically changed testicular tissue. The following comparison with the histologic evidences showed the correspondence of the results.

In vitro tests of new biomaterials require internationally and nationally eluates increasingly. Only the extracted substances become effective in the cells in that way. The material surface has the greatest influence on the quantity of the released substances by elution. The surface of diverse undefined mixture quantities is definable exactly by means of a gas adsorption method (BET method). The influence of the secondary technologies was tested by the atom adsorption spectrometry. The Jena bioglass ceramics could be estimated successfully by this method.

A standard method for bovine islet isolation has been developed. The mean islet yield per gram pancreas obtained from the last twelve consecutive glands was 2,098 +/- 292 representing an insulin recovery of 36.1 +/- 6.5%. The static incubation of freshly isolated bovine islets in the presence of glucose resulted in a less distinct increase of insulin secretion than in the presence of free fatty acids. Nevertheless, xenotransplantation of bovine islets to immunosuppressed C57BL6J mice did reverse streptozotocin-induced diabetes for 2 to 6 days.

The influence of hydrocortisone and dopamine to the course of the organic functional changes and the activity of same lysosomal hepatic enzymes were tested in 30 dogs in endotoxic shock. A continuous lowering of the blood pressure and the central venous pressure were observed in the non-treated group. The hemodynamic impairments turned out in the treated groups in a less acute course. The coupled treatment with hydrocortisone and dopamine in the experimental endotoxic shock seems to reduce the activity of the lysosomal enzymes in the liver and in the peripheral blood.

The effect of the specific alpha1-blocker prazosin (Adversuten) on the microcirculation in the region of the terminal ileum was examined on 10 rabbits. Before and after injection of prazosin into the superior mesenteric artery an isotope-clearance of a 133Xe-solution, applied into the intestinal wall, was performed. Because of the systemic blood pressure decreasing effect of prazosin the circulation of the examined part of the intestine was reduced in spite of the intended vasodilation of the intestinal vessels. The therapeutic applicability of alpha-blockers for intestinal diseases of vascular genesis remains questionable.

The videodensitometry is a suitable method for recording the gastric wall motility. It provides biosignals amenable by computer. There are no special demands to any organic form or kind of contraction. The asymmetry of the stomach makes no difference, therefore. The hitherto usual manometry will not be superseded. Both methods complete themselves in an ingenious manner. The radiation exposure by videodensitometry can be diminished by modifying the system for image causation.

In the pathogenesis of acute pancreatitis the events and mechanisms increasing the digestibility of the acinar cells are widely unknown. To study these processes at cellular level, isolated exocrine cells from rat pancreas were utilized as experimental tool. The investigations revealed that extracellularly acting noxae seem to be of less importance to triggering autodigestion. The plasma membrane of the acinar cells was relatively resistant to extracellular trypsin, while anoxia or uncoupling of oxidative phosphorylation obviously reduced the survival of these cells. Combined action of small amounts of trypsin and partial uncoupling resulted in a potentiated cell destruction. The findings presented stress the importance of a well-functioning energy metabolism for the protective potential of acinar cells to further harmful conditions.

The behaviour of the small bowel absorption in the immediate postoperative phase after cholecystectomy was tested in 20 patients by means of the modified D-xylose-test. Either the conventional insufflation anesthesia with halothane or the neuroleptic analgesia was used in 10 patients in each case. Characteristic changes of the small bowel absorption were provable after the operation. The postoperative restriction of the absorption was not changed statistically significantly in dependence on the different anesthesia methods. An absorption without restriction exists in both groups at the beginning of the 3rd post-operative day.
