Potential Interference of Maternally Derived Antibodies (MDA) of Commercial Broiler Chicks on the Efficacy of Different Vectored –ND Vaccines 商品肉鸡母源性抗体(MDA)对不同载体新城疫疫苗效力的潜在干扰

Maternally derived antibodies (MDA) are a major problem for the application of NDV-vectored vaccines in commercial chickens with high levels of MDA, which could impair its efficacy. In this study, we assessed the effectiveness of different commercial vectored ND in broilers chicks with MDA against local heterologous NDV genotype VII. For this purpose, Two hundred and forty one day old chicks were divided into eight groups (30 chicks/group) (G1, G2, G3, G4, G5, G6 and G7) vaccinated with four different recombinant ND vaccines using different vaccination scheme. In addition, Group 8 (30 chicks) kept as a control non-vaccinated group. Efficacy of this type of vaccine is evaluated routinely by vaccination-challenge test. ELISA test were used for detection antibodies from collected sera of all chickens groups. Challenge test were applied at 31th day's old age of chickens. To calculate viral Shedding, cloacal swabs were collected from all groups and examined by real time RT-PCR. Obtained results for G8 (control) was recorded high antibody titer at one day but decreased gradually till become negative at 24 day old. While vaccinated groups exhibited antibody titers at 38 days old which were 3.6, 3.7, 4, 3.7, 3.66, 3.77, and 3.3 for G1, G2, G3, G4, G5, G6 and G7, respectively. Protection% of vectored ND vaccines groups ranged from 80 to 95 % despite inactivated ND vaccine recorded 60%. It was concluded that the recombinant ND vectored vaccines more potent than inactivated one in broiler chickens with MDA. Administration of rFowl-Pox+ND vaccine at 10 day exhibited higher immune response and conferred more protection and reduction of viral shedding in comparison to vaccinated at hatchery. Using rHVT+ND vaccine in one day old chicks confer optimum immune response and protection% in presence of MDA. Prime-boost vaccination strategy can overcome the problem of MDA in broiler chickens vaccination. So, it was recommended to measure the level of MDA in offspring to determine the ideal time for vaccination

Protective Effect of L-carnitine against Cefquinome Sulfate Induced Testicular Damage and Oxidative Stress in Male Albino Rats 左旋肉碱对硫酸头孢喹肟致雄性白化大鼠睾丸损伤和氧化应激的保护作用

The present study was conducted to evaluate the protective effect of L-carnitine (LC) against possible deleterious effects of cefquinome sulfate (CS) on antioxidant capacity, fertility parameters and histopathological findings of reproductive organs in male rats. Therefore, seventy two mature male rats were equally divided into four equal groups as following: The control group of rats was administered two s.c. doses of physiological saline (vehicle of other drugs) (2ml/kg b.wt. once daily for 5 successive days and 2ml/kg b.wt. once daily for 8 successive weeks), the second group was s.c. administered LC (200 mg/kg b.wt. once daily for 8 successive weeks), the third group was s.c. administered CS (50 mg/kg b.wt. once daily for 5 successive days) and the fourth group was s.c. administered CS concomitantly with LC by the same doses. The obtained results showed that administration of CS induced a significant (P≤ 0.05) increase in serum level of Malondialdehyde (MDA) and total sperm abnormalities with significant (P≤ 0.05) decrease in serum level of total antioxidant capacity (TAC), sperm motility and total sperm count. Moreover, CS induced histopathological changes in tissues of testes and epididymis. Conversely, administration of LC concomitantly with CS ameliorates all the previous antioxidant, fertility and histopathological alterations induced by CS. It could be concluded that CS induced some undesired effects on antioxidant activity, fertility parameters and histopathological findings which could be ameliorated by LC in male albino rats.

Phenotypic and Genotypic Characterization of Klebsiella pneumoniae isolated from Clinical Mastitis in Buffaloes 水牛临床乳腺炎肺炎克雷伯菌的表型和基因型分析

Mastitis is the most common serious and economical disease affecting the dairy industry. The current study aimed to determine the molecular characterization of K. pneumoniae isolated from buffaloes with clinical mastitis as well as determine the antimicrobial pattern and virulence associated genes. Out of examination one hundred and fifty dairy buffaloes at Beheria governorate, 57 (38%) showed clinical mastitis signs. All samples are submitted to bacteriological examination and confirmed by biochemical tests. The result showed that K.pneumoniae was identified in only five (8.7%). Regarding to the antibiogram profile, K. pneumoniae isolates exhibited high resistance pattern against Carbenicillin, Cefotaxime, Flumequine, Gentamicin, Kanamycin (100%) followed by Neomycin, Nitrofurantoin, Penicillin (80%) and Oxytetracycline, Sulfamethoxazole / trimethoprim (60%). Meanwhile, higher susceptibility to Ampicillin (100%), followed by Chloramphenicol, Colistin Sulphate, Erythromycin, and Streptomycin (80%). Molecular identification using the mPCR approach applied efficiently to detect the K. pneumoniae virulence genes (mrkA, mrkD, iutA) at 475, 226, 300bp respectively. In addition, the successfully detection of blaTEM and blaSHV for β lactams resistance genes at 516 and 392 bp. In conclusion, the phenotypic and genotypic resistance pattern of K. pneumoniae isolates indicates the portability of transmission of resistance genes through milk and food chain with public health concern.

Impact of Hazard Analysis Critical Control Point System (HACCP) Application for Production of High Quality and Safe Table Eggs at Alexandria Governorate 危害分析关键控制点系统(HACCP)应用对亚历山大省优质安全食用蛋生产的影响

HACCP is an important system for producing safe eggs from poultry farms. Also, HACCP helps to reduce the risk of foodborne illnesses associated with eggs by ensuring that they are produced in a hygienic environment with proper sanitation practices. Therefore, 120 eggs representing 24 groups (5 eggs group) were collected from one poultry farm before and after the application of the HACCP system (12 groups before and 12 groups after application of HACCP system) located in Alexandria governorate. The obtained results revealed that the poultry farm after application of HACCP system produced safe eggs with lower aerobic bacterial count and yeast & mould counts than the results of the same farm before application of HACCP system. In addition, egg content from the farm after application of HACCP system is free from pathogenic microorganisms (Salmonella species and Escherichia coli), free from toxic elements (Lead and Cadmium) and free from veterinary drug residues. While the results from the same farm before application of HACCP system produced eggs with a higher incidence of total bacterial, yeast & mould count. Besides, two groups of egg from the same farm before application HACCP system were contaminated with Salmonella species which serological identified into Salmonella enteritidis; also, three groups contaminated with Escherichia coli which serologically identified into O91: H21 (EHEC) (2 isolates) and O2: H6 (EPEC) (1 isolate); In addition, egg content from this farm before application HACCP is contaminated with toxic elements such as lead and cadmium with incidence level 75 % and 66.7%, respectively. Finally, examination of veterinary drug residues in the farm before application of HACCP system indicated presence of Tylosin and Oxytetracycline residues in 7 egg groups. Therefore, the application of the HACCP system in poultry farms is essential for ensuring the safety of the eggs produced.

Effect of Bioactive Compounds of Lemon, Onion and Garlic Extract on Buffalo Semen Quality, Antioxidant Status and Ultrastructure Changes 柠檬、洋葱和大蒜提取物生物活性成分对水牛精液品质、抗氧化状态和超微结构的影响

The present study aimed to compare semen and fertility parameters of frozen-thawed buffalo semen extended in egg yolk based extender and the same extender supplemented with lemon, onion and garlic (LOG) extract as a natural antioxidant agents. The semen samples were re-suspended with basic diluents containing LOG extracts (10, 20, 30, 50, 100, 150; and 200 µl/ml). The control diluents comprised of only extender and lacked any antioxidant agent. Semen was collected from 10 mature buffalo bulls twice a week using artificial vagina. The supplementation of LOG at a concentration of 30 and 50 µl/ml significantly improved post-thawing motility and sperm viability. The status of acrosome and membrane integrity significantly improved in spermatozoa when supplemented with 30 and 50 µl/mlLOG extract. Also, 30 and 50 µl/ml LOG extract significantly reduced lipid peroxidation and increased total antioxidant activity (TAC). The highest rate of conception was observed in spermatozoa treated with 30 µl/ml LOG extract. The application of LOG extract resulted in improvement in the quality and fertility of frozen-thawed spermatozoa. Nevertheless, future studies are wanted to elucidate how the bioactive components in the LOG extract (especially in the low concentration) prevent cryo-damages in spermatozoa.

This study assessed the microbiological evaluation of environmental contamination of mostly consumed fruits and vegetables by intestinal parasites and enteric bacteria in three major markets (Waaso, Aarada and Akande) Ogbomoso, Oyo State Nigeria. A total of 5 fruits and 5 vegetables commonly consumed by dwellers of these communities were randomly purchased from the three markets. They were washed, and examined for microbial and parasitic organisms. Of the 150 samples of fruits and vegetables that were collected, processed and examined, 83 (55.3%) of them were contaminated with intestinal parasites, results was generated by determining the frequency distribution in which values was obtained by the Mean±SD and the significance of differences was also obtained using analysis of variance (ANOVA) and p-value of <0.05 was considered significant. The intestinal parasites observed included: Ascaris lumbricoides, Balantidium coli, Entamoeba coli, Entamoeba histolytica, Strongyloides stercoralis, Taenia saginata, Trichuris trichuira and Ancyclostoma duodenale. The predominant bacteria isolated from both fruits and vegetables included: Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Escherichia coli, Enterococcus species, Klebsiella oxytoca, Staphylococcus saprophyticus and Klebsiella species. The parasites recovered from fruits and vegetables sold in Waaso market were more than those recovered from Aarada and Akande market which are 34(41.0%), 27(32.5%) and 22(26.5%) parasites respectively. Results show that 83(55.3%) out of the 150 fruits and vegetable samples in total examined had intestinal parasites. The prevalence of intestinal parasites in these fruits and vegetables was found to be 4.0% in Banana, 8.7% in Cabbage, 2.7% in Carrot, 3.3% in Cucumber, 6.7% in Jute mallow, 4.0% in Orange, 6.0% in Pineapple, 5.3% in Spinach, 8.6% in Tomato and 6.0% in Water melon. Findings from this study revealed that raw fruits and vegetables could be potential sources of microbial and parasitic infections, it is therefore essential to educate the populace on the need to inculcate the habit of thorough washing of these products with clean water before consumption to avoid potential health risk.

Probiotic Effects on Behavior, Stress Indicators and Antioxidant Activity of Transported Broiler Chicken 益生菌对运输肉鸡行为、应激指标及抗氧化活性的影响

Transportation before slaughtering causes bad effects on chicken welfare and health, but little is known about the effect of probiotic dietary fed in broiler chickens having transport stress. So, this study was designed to detect the effects of two doses of a probiotic (Bacillus subtilis) on some stress indicators, antioxidant activity and fear response of broiler chickens exposed to transportation stress. Two hundred and ten broiler chicks were distributed among 21 floor pens (10 chicks in each pen), each pen was exposed to one of three diet treatments containing a probiotic at 0 (control), 0.25 (0.25X), and 0.5 (0.5X) g/kg. At 35 days old, birds were exposed to transportation and then blood samples were collected for measuring stress and antioxidant markers. The serum corticosterone and cholesterol levels were decreased in probiotic fed birds in comparison with controls (P < 0.05). Compared to control broiler chickens, 0.5X birds had higher levels of catalase and serum superoxide dismutase (P < 0.05). In addition, the 0.5X birds spent less time in the tonic immobility test (P = 0.0427). The results suggest that the probiotic supplement may prove to be an important management tool for the broiler industry to diminish the negative effects of transport stress, potentially safeguarding the welfare and health of broiler chickens.

The Effect of Antibiotics Used Throughout Chicken Cycle on the Bacterial Count of Their Meat 鸡全周期使用抗生素对肉中细菌数量的影响

There is no doubt that poultry industry is a very useful to our society Therefore poultry meat has its own value even economic value or nutrional value. The feature of poultry meat helps the consumer to obtain the more nutritional value such as Protein, Low fat, Carbs and vitamins. The contamination of poultry meat effects on the quality of meat and its content. Therefore it effects the population and can cuase infectious diseases .The present experiment was conducted on the broilers that obtained from private farm located at Egypt-Alexandria road. The broiler chicks were fed on well-balanced diet. Starter diet was given till the 20th day of age after that chicks were fed on finisher diet which was given from the 21st day till the end of the experiment. Water and food were offered adlibitium. The chicks were housed in clean well ventilated separate experimental rooms, previously fumigated with formalin and potassium permanganate. The floor was bedded by fresh clean chopped wheat straw forming litter of 3.5 cm depth. Each room was provided by suitable numbers of feeders and water utensils. Each group of bird was provided by suitable feeder and water. This study aimed to investigate the effect of antibiotics administrated during broilers chicken cycle on the bacteriological count of their meat through study of: 1- The level of mesophilc, aerobicenterobacteriacae and coliform counts among the breast and thigh samples at 15 day and 35 days of production cycle. 2- The level of mould and yeast counts among the breast and thigh samples at 15 day and 35 days of production cycle. 3- Identification of some isolated mould and yeast.4- Determination of the antibiotic residues among the breast and thigh samples at 15 day and 35 days of production cycle. The Staistical analytical results of Mesophilc aerobic bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 34.15 X 103 ± 14.28 X 103 in breast at day 15 and was 38.64 X 103 ±19.88 X 103 in thigh at day 15. But in day 35 was 82.15 X 103 ± 24.51 X 103 in breast and was 89.91 X 103 ±28.99 X 103 in thigh at day 35 .The results of Enterobacteriaceae bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 4.20 X 102±1.70 X 102 in breast at day 15 and was 1.51 X 103±4.70 X 102 in thigh at day 15 . But in day 35 was 9.04 X 103±3.93 X 103 n breast and was 3.21 X 103±1.32 X 103 in thigh at day 35.The results of Coliforms bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 1.64 X 103 ± 1.13 X 103 in breast at day 15 and was 3.30 X 102± 6 X 10 in thigh at day 15. But in day 35 was 4.5 X 102± 1.7 X 102 in breast and was 7 X 10 ± 2 X 10 in thigh at day 35. Tht results of Moulds counts isolated from breast and thigh of broilers at different periods of experiment was 2 X 10 ± 1.0 X 10 in breast at day 15 and was 2.0 X 10 ± 1.0 X 10 in thigh at day 15. 毫无疑问,家禽业对我们的社会是非常有用的,因此家禽肉有其自身的价值,甚至是经济价值或营养价值。禽肉的特性有助于消费者获得更多的营养价值,如蛋白质、低脂肪、碳水化合物和维生素。禽肉污染对肉质及其含量的影响。因此,它影响种群,并可引起传染病。本试验是在埃及-亚历山大路私人农场获得的肉鸡上进行的。肉鸡用均衡的饮食喂养。初饲至20日龄,初饲至21日龄,初饲至试验结束。提供了水和食物。雏鸡被饲养在清洁通风良好的独立实验室内,事先用福尔马林和高锰酸钾熏蒸。地面铺上新鲜干净的麦秸碎,形成3.5 cm深的凋落物。每个房间都配备了适当数量的喂食器和饮水器具。为每组鸟提供合适的饲料和水。本研究旨在通过研究:1 .在生产周期第15天和第35天,肉鸡胸部和大腿样品中嗜温菌、好氧肠杆菌和大肠菌群的数量水平,探讨在生产周期中施用抗生素对肉鸡肉中细菌数量的影响。2-在生产周期的第15天和第35天,乳房和大腿样品中的霉菌和酵母计数水平。鉴定一些分离的霉菌和酵母菌。4-生产周期15天和35天乳房和大腿样品中抗生素残留量的测定。不同试验时期肉仔鸡胸部和大腿分离的嗜中温需氧细菌数量统计分析结果为:第15天乳房34.15 X 103±14.28 X 103,第15天大腿38.64 X 103±19.88 X 103。第35天乳房为82.15 × 103±24.51 × 103,第35天大腿为89.91 × 103±28.99 × 103。试验各时期肉仔鸡乳房和大腿分离肠杆菌科细菌数量分别为:第15天乳房为4.20 × 102±1.70 × 102,第15天大腿为1.51 × 103±4.70 × 102。第35天乳房为9.04 × 103±3.93 × 103,大腿为3.21 × 103±1.32 × 103。试验不同时期肉仔鸡胸部和大腿分离的大肠菌群数量分别为:第15天乳房1.64 × 103±1.13 × 103,第15天大腿3.30 × 102±6 × 10。第35天乳房为4.5 × 102±1.7 × 102,大腿为7 × 10±2 × 10。试验不同时期肉仔鸡胸、大腿分离霉菌数分别为:第15天乳房2 × 10±1.0 × 10,第15天大腿2.0 × 10±1.0 × 10。第35天乳房为3.0 × 10±1.0 × 10,大腿为3 × 10±1.0 × 10。试验不同时间乳腺和大腿分离酵母计数:第15天乳腺为0.04 × 103±0.02 × 103,第15天大腿为0.11 × 103±0.05 × 103。第35天乳房为0.03 X 103±0.01 X 103,大腿为0.03 X 103±0.01 X 103。

Therefore it effects the population and can cuase infectious diseases .The present experiment was conducted on the broilers that obtained from private farm located at Egypt-Alexandria road. The broiler chicks were fed on well-balanced diet. Starter diet was given till the 20th day of age after that chicks were fed on finisher diet which was given from the 21st day till the end of the experiment. Water and food were offered adlibitium. The chicks were housed in clean well ventilated separate experimental rooms, previously fumigated with formalin and potassium permanganate. The floor was bedded by fresh clean chopped wheat straw forming litter of 3.5 cm depth. Each room was provided by suitable numbers of feeders and water utensils. Each group of bird was provided by suitable feeder and water. This study aimed to investigate the effect of antibiotics administrated during broilers chicken cycle on the bacteriological count of their meat through study of: 1- The level of mesophilc, aerobicenterobacteriacae and coliform counts among the breast and thigh samples at 15 day and 35 days of production cycle. 2- The level of mould and yeast counts among the breast and thigh samples at 15 day and 35 days of production cycle. 3- Identification of some isolated mould and yeast.4- Determination of the antibiotic residues among the breast and thigh samples at 15 day and 35 days of production cycle. The Staistical analytical results of Mesophilc aerobic bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 34.15 X 103 ± 14.28 X 103 in breast at day 15 and was 38.64 X 103 ±19.88 X 103 in thigh at day 15. But in day 35 was 82.15 X 103 ± 24.51 X 103 in breast and was 89.91 X 103 ±28.99 X 103 in thigh at day 35 .The results of Enterobacteriaceae bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 4.20 X 102±1.70 X 102 in breast at day 15 and was 1.51 X 103±4.70 X 102 in thigh at day 15 . But in day 35 was 9.04 X 103±3.93 X 103 n breast and was 3.21 X 103±1.32 X 103 in thigh at day 35.The results of Coliforms bacterial counts isolated from breast and thigh of broilers at different periods of experiment was 1.64 X 103 ± 1.13 X 103 in breast at day 15 and was 3.30 X 102± 6 X 10 in thigh at day 15. But in day 35 was 4.5 X 102± 1.7 X 102 in breast and was 7 X 10 ± 2 X 10 in thigh at day 35.
