One of the native catfish to Indonesia is the baung fish (Hemibagrus). Baung fish are found in rivers in Sumatra, Kalimantan and Java. The catfish population is declining, thus conservation is needed to prevent extinction. To conduct an effective conservation efforts, molecular studies are needed to confirm the fish species from the three islands. Mitochondrial DNA is one of the markers that is often used to see the lineage and kinship of animals for conservation purposes. The purpose of this study is to determine the genetic diversity of the NADH Dehydrogenase Subunit I (ND1) gene catfish from each of these locations, can be used as a genetic marker. Samples were obtained from their natural habitat, namely Magelang (5), Palembang (3), Riau (2), Samarinda (2), 2 from Sintang, and 3 from Banjarmasin. The DNA of the fish sample was then isolated and then used as a template for amplification of DNA fragments using PCR techniques. Amplikon (PCR product) was then purified by gel extraction and then sequenced to determine the DNA sequence. The potential of DNA sequences as catfish genetic markers was proven by analyzing genetic diversity between species using the MEGA version 7.0 program (Kumar et al., 2016). The sequencing results 972 nucleotides composing the ND gene, and found differences in 268 nucleotide sites and 47 amino acid sites. Based on the nucleotide sequence of the ND1 gene, catfish from the Progo river (Magelang, Central Java), the Musi river (Palembang, Sumatra), the Kampar river (Riau), the Kapuas river (Sintang, Kalimantan), the Martapura river (Banjarmasin), the Mahakam river (Kalimantan) is included in the Hemibagrus sp.; catfish from the Elo river (Magelang, Central Java) belongs to the genus Mystus sp.; catfish from the Bengawan Solo river (Bojonegoro, East Java) belongs to the Pangasius sp.

One of the effort to provide superior rams is to pay attention to the provision of feed that has good substances and is able to improve the quality of sperm. Based on studies, snail flour has a fairly high protein content and snails can be an alternative source of protein in additional feed. This study aims to improve the quality of sheep sperm using an additional feed of snail flour (Achatina fulica) which can increase the use value and economic value. Eight sheep used in this research were divided into one group as the control and two groups as the treatment. The control group was given standard sheep forage, while the treatment groups were given standard sheep forage added with flour snail of 5% and 10%. Sperm collection using castration method. Parameters observed were motility, viability, and concentration of sperms. The results of the study showed that the addition of snail flour as additional feed for sheep affected sperm quality with a significant increase in concentration parameters, while motility and viability did not show a significant increase

Fauna 2, Karangmalang, Yogyakarta 55281 ABSTRACTAvian Pathogenic Escherichia coli (APEC) is a pathogen that causes colibacillosis in poultry, including salpingitis, omphalitis, cellulitis, swollen head syndrome, coligranuloma yolk sac inflammation, and air sacs inflammation. APEC is a zoonotic strain which spread through raw meat and processed meat products of animals and birds. In this research, the isolation and the identification of Escherichia coli were done by using selective media MacConkey, Kligger Iron Agar, and Gram staining. To analyse genetopically, polymerase chain reaction (PCR) was used to detect 16SrRNA genes, vt1 genes, and vt2 genes. Thirty one (55,36%) isolates of 56 specimens collected from quail were detected as Escherichia coli. The detection of APEC strains towards 31 Escherichia coli isolates were done by using polymerase chain reaction (PCR) with vt1 and vt2 specific primer. From the results showed that 32,26% (10/31) was APEC strains and 67.74% was non-APEC strains. From 10 isolates, 90% had vt1 gene and 10% had vt2 gene. Escherichia coli isolates were found in eyes (32,26%), infraorbital sinus fluid (32,26%), nasal fluid (16,20%), also in lungs, air sacs, ascites, and heart for 3,2% each. The isolates could not be found in the specimens from the skull. These isolates are zoonotic and have an impact on human health. Keywords: APEC, colibacillosis, quail, vt1 and vt2

Milk products consumption has become a new trend of healthy lifestyle among students in order to meet appropriate daily nutrients. This study was aimed to measure the total number of microbes found in milk drinks sold around the IPB Dramaga and Cilibende and to determine factors that influence it. Samples were collected from 13 milk vendors within radius of 2 kilometers from outermost point of campus and taken with 3 repetitions. Calculation of the total number of microbes was done by using the plate count method in accordance with SNI 2897-2008 about testing method of microbial contamination in meat, eggs, and milk, as well as processed products and SNI 01-6366-2000 about the maximum limit of microbial contamination in food. Risk factors’ data was taken by interviewing milk drinks seller using structured questionnaire. Factors that influenced microbial contamination were analyzed using Chi-Square test. The results showed that 74.4% of milk drinks samples had a high total number of microbes. The average total number of microbes in Dramaga and Cilibende samples was 1.0 x 105 ± 1.4 x 105 cfu / ml and 1.1 x 107 ± 1.2 x 107 cfu/ml. The total number of microbes are significantly influenced (p<0.05) by equipments used, the cleanliness of the equipment, the cleanliness of the table, washing hands practice, and the distance from the crowd. The high number of microbial contamination in milk drinks could damage the quality of milk and shorten the shelf life.

Back leg paresis is muscle weakness that can cause the inability to walk or support the back of the body. A domestic cat, female, black tabby hair colour with a bodyweight of 2.9 kg has weakness in both hind legs so it cannot lift the back of the body properly but there is still a reflex when pressing using tweezers sirurgis on the gluteus muscle tendon. The flexor reflexes on the digits are still present but slightly. From the physical examination, it was concluded that there was a disturbance in the segment IV spinal cord, precisely at L7-S1. Cats also experience problems with defecation, namely the patient's stool is watery inconsistency. The results of the X-Ray examination showed that the segments of the os. lumbar, os. femur, os. tibia, os. fibula, os. metatarsal to digit there are no fractures and also the visceral organs do not experience changes and abnormalities. However, there is a bias picture, namely compression on the 7th segment (S4-L5, L6 and L7). The diagnosis was that the cat had disorders of the spinal cord segments IV (L4-S2) and V, to be precise, from L7 to S2. Electroacupuncture therapy at points BL-18, BL-19, SP-6, ST-36 and LIV-3 was carried out for three weeks, namely twice in the first week and once in the second and third weeks. Oral drug therapy in the form of Neurobion® tablets is taken orally during therapy.

There is a transition period that dairy cattle always pass through in their reproduction cycle, making them more stressed. Stress can simply be identified by the ratio of neutrophil (N) and lymphocyte (L) at the haematologic state of dairy cattle. The purpose of this study is to evaluate stress conditions in dairy cattle’s transition period based on neutrophil and lymphocyte ratio. Twenty samples of dairy cattle’s blood in prepartum and postpartum condition are being analyzed with a hematology analyzer. The result of this study shows that the average of neutrophil has increased insignificantly from prepartum to postpartum period as 4.14 103/µL ± 1.51 to 4.27 103/µL ± 2.08 or 1.03% with a normal interval of neutrophil is 1.7-6.0 103/µL. However, the average of lymphocytes decreased insignificantly from 3.23 103/µL ± 1.13 to 3.03 103/µL ± 1.23 or 0.93% with a normal interval of lymphocyte is 1.8-8.1 103/µL. Then, the average of neutrophil and lymphocyte ratio has increased insignificantly from 1.35 103/µL ± 0.46 to 1.47 103/µL ± 0.60 or 1.09%. Based on this result, the ratio of neutrophils per lymphocyte still in the normal range. In the transition period, only a few dairy cattle have a stressful condition. The stress condition in dairy cattle can be identified by increasing neutrophil (neutrophilia) and reducing lymphocyte (lymphocytopenia).
