Effects of Angelica sinensis Root on Longitudinal Bone Growth Rate in Adolescent Female Rats 当归根对青春期雌性大鼠纵向骨生长速率的影响

Objectives : This study aimed to investigate the effects of Angelicae sinensis Radix on longitudinal bone growth rate in rats. We have screened traditional medicinal herbs to develop the longitudinal bone growth stimulator by wellestablished rat model. A. sinensis was identified as one of the effective herbs in the screening process. Methods : Adolescent female rats were administered A. sinensis at doses of 30 ㎎/㎏ and 300 ㎎/㎏ for 10 consecutive days. To observe the rate of longitudinal bone growth, tetracycline was injected intraperitoneally on day 8 to stain a fluorescent band on the anew formed bone. To elucidate the mode of action, we observed insulin-like growth factor-1 (IGF-1) and bone morphogenetic protein-2 (BMP-2) expression after A. sinensis administration in growth plate. Results : In the 300 ㎎/㎏ A. sinensis group, the length between the proximal endpoint of the tetracycline label and the division line between growth plate and bone was significantly increased compared with vehicle-treated control group. Height of the proximal tibial growth plate was higher in the A. sinensis group compared with control group. A. sinensis also upregulated the expressions of IGF-1 and BMP-2 in the proliferative zone and hypertrophic zone of the proximal tibial growth plate. Conclusions : A. sinensis increases longitudinal bone growth rate in rats. According to immunohistochemistry, A. sinensis increases local IGF-1 and BMP-2 expressions in the growth plate which can be considered as direct stimulation of GH on the local growth plate.
目的:研究当归对大鼠纵向骨生长速率的影响。我们通过建立的大鼠纵向骨生长刺激模型,筛选了传统中药来开发纵向骨生长刺激剂。在筛选的过程中,鉴定出白羊草为有效药材之一。方法:按30㎎/㎏、300㎎/㎏剂量给青春期雌性大鼠灌胃华刺,连续10 d。观察纵向骨生长速度,第8天腹腔注射四环素,在新形成的骨上染色荧光带。为了阐明其作用模式,我们在生长板上观察了中华按蚊给药后胰岛素样生长因子-1 (IGF-1)和骨形态发生蛋白-2 (BMP-2)的表达。结果:300㎎/㎏白羊组产水四环素标签近端至生长板与骨分割线的长度较车辆处理对照组显著增加;与对照组相比,中华按蚊组胫骨近端生长板高度增高。在胫骨近端生长板增生区和肥厚区IGF-1和BMP-2的表达上调。结论:中华黄芪可提高大鼠纵向骨生长速率。免疫组化结果显示,中华按蚊在生长板局部IGF-1和BMP-2表达升高,可认为是对生长板局部GH的直接刺激。

Quality Monitoring of Specification of Crataegi Fructus in the Korean Pharmacopoeia and Studies HPLC Standard Chromatogram 韩国药典中山楂药材质量监测及HPLC标准图谱研究

Objectives : Crataegi Fructus has been used as traditional medicines for more digestion action, amenorrhea due to blood stasis, and hyperlipemia. The aim of this study was to compare of Crataegi Fructus in South Korea collected during three years according to the standards in monographs of the Korean Pharmacopoeia Eleventh edition (KP 11). Methods : Crataegi Fructus was carried out identification test (Qualitative reaction, Thin layer chromatography), heavy metal test, and total ash registered at KP. Add to we tested loss on dry, contents of ethanol-soluble extracts, and HPLC profiling. Results : Identification test (TLC) was on comparing with ursolic acid standard solution in Rf value, all samples showed red purple spot (Rf value 0.9). Ursolic acid spot in Rf value 0.35 showed by changing mobile phase condition. Heavy metals showed contents for Pb, Cd, As, and Hg range of 0.0 ∼ 0.5 ppm, 0.0 ∼ 0.2 ppm, 0.0 ∼ 0.3 ppm, and 0.0 ∼ 0.1 ppm. Loss on drying was ranged from 5.5 to 11.9 %, total ash was between the range 2.7 ∼ 4.0 %.
目的:山楂子具有消化作用强、瘀血闭经、高脂血症等作用。本研究以韩国药典第11版(KP 11)各论标准为标准,对韩国三年间采收的山楂进行比较。方法:对山楂进行定性鉴定(薄层色谱定性反应)、重金属含量测定和总灰分测定。添加到我们测试的干燥损失,乙醇溶提取物的含量,和HPLC分析。结果:薄层鉴别法(TLC)与熊果酸标准溶液的Rf值进行比较,样品均呈红紫色斑点(Rf值为0.9)。通过改变流动相条件,在Rf值0.35处出现熊果酸斑。重金属Pb、Cd、As和Hg的含量分别为0.0 ~ 0.5 ppm、0.0 ~ 0.2 ppm、0.0 ~ 0.3 ppm和0.0 ~ 0.1 ppm。干燥损失在5.5% ~ 11.9%之间,总灰分在2.7 ~ 4.0%之间。
