In this work, the thermal disruption of type I collagen is investigated using second harmonic generation (SHG) microscopy. We investigated the effects of second harmonic generation images and intensity of rat tail tendon in the temperature range from 25 to 60/spl deg/C. We found a structural transition at 57/spl deg/C where a drop in second harmonic intensity is correlated to a disorganization of the collagen fibers. Our results suggest that SHG microscopy is an effective technique to monitor thermally induced collagen structural changes for biomedical applications.

For understanding the correlations between temporal and spatial photon distributions in the process of random scattering in turbid media, the Monte Carlo algorithm is used to describe various optical gating conditions. We study the distributions of transmitted photon with time-, aperture-, and angle-gating techniques. We record the optical paths of transmitted photons with different aperture sizes and angles. Time-resolved profiles and polarization states of transmitted photons are also discussed.

Due to the growing importance of optical imaging in tissue diagnosis, we use multiphoton microscopy, to characterize normal and cancerous oral tissues. We acquired both microscopic and spectroscopic data for the characterization of normal and cancerous oral tissues. Our results show that both multiphoton fluorescence and harmonic generation images may be used to distinguish both tissue types. An image of different oral tissues will be of value for clinicians in the diagnosis and treatment of oral diseases.

We imaged and found that four fungi, M.canis T.menta Fusariun Asp.flavus, can be identified using multiphoton imaging and that their luminescence is located in blue and green part of the spectrum.
