A simple and sensitive high performance liquid chromatographic determination method for free polyamines and their acetylated derivatives was developed. Polyamines were separated on an octadecyl polymer column with isocratic elution. The separated polyamines were introduced into an enzyme reactor containing immobilized acylpolyamine amidohydrolase, amine oxidase (flavin-containing) and putrescine oxidase, in which they were deacetylated and oxidizld to generate hydrogen peroxide. In competitive enzyme-immunoassays (EIAs) of insulin, mixtures of several forms of enzyme-labeled insulins prepared using glutaraldehyde1), maleimide2) or periodate 3,4) have been used . There is no detailed comparison of enzyme-labeled insulins with respect to the sensitivity of EIA of insulin. The enzyme-labeling site (s) and the number of enzyme molecules incorporated per insulin molecule may be important factors influencing the sensitivity of the EIA. Recently we have developed a method for the preparation of horseradish peroxidase (HRP ; EC 1. 11. 1. 7) -labeled porcine insulins (HRP-insulins ; GlyA1-HRPinsulin, LysB29-HRP-insulin and GlyA1, LysB29-diHRP-insulin)5) (Fig. 1). This paper surveys the immunoreactivity of the above HRP-insulins with a mouse anti-human insulin monoclonal antibody and assesses the sensitivity of each as competitors in solid-phase EIA of human insulin. Two EIA methods for human insnlin are examined : a rapid method by simultaneous addition of HRPinsulin and a highly sensitive method by delayed addition of HRP-insulin.

As indicated at the outset of this report, the critical nature of the clinical requirements when blood gas testing is indicated, coupled with the labile nature of the sample and rapidly changing patient conditions makes it imperative that the analyst be aware of more than just the analysis it self Knowledge by the analyst, of the physiology of blood gas, the interpretation of basic blood gas results, the effects of the changing patient environment and the manner in which the sample is treated in a few short minutes from collection to reporting are equally important if one is to assure timely and clinically meaningful results.

Glycation and the Development of Diabetic Retinopathy 糖化与糖尿病视网膜病变的发展

The levels of advanced-stage products of the Maillard reaction in lens proteins are considered to reflect the sum of accumulated glycation over time, because there is little turnover of lens crystallins. Thus, we investigated the relationship between the degree of diabetic complications, i. e. diabetic retinopathy and levels of advanced-stage products accumulated in the lens proteins prior to the surgical resection. The level of advanced-stage products in the lens proteins increased with the progression of diabetic retinopathy. The differences among the level in the groups of each diabetic retinopathy were significant. These results suggest that glycation measured by advanced Maillard product in the lens protein plays an important role in the etiology of diabetic complications.
