The Pacific abalone Haliotis discus hannai is a commercially important marine mollusk.To develop a rapid,economically efficient and robust approach for genetic studies in this species,twelve microsatellites were used to optimize four multiplex PCRs for the H.discus hannai,and the results obtained indicate the usefulness of the four multiplexed microsatellite systems in parentage assignment.We evaluated and validated these multiplex PCRs in 12 full-sib families.The average polymorphism information content(PIC)was 0.82.Inheritance analyses in the 144 tests showed that 4.9% microsatellite loci deviated from Mendelian ratios.The frequency of null alleles was estimated at 10.6% of all the alleles segregating based on a within-family analysis of Mendelian segregation patterns.Simulations and parentage analysis showed that with the most informative multiplex set 4,the assignment success could be 86% and 90% respectively,and 100% of the offspring were correctly allocated to their parents when two or more multiplex PCRs were used.Our results show that four multiplex PCR panels of microsatellites are possible and can be used for rapid and highly efficient parentage assignment in the Pacific abalone.

Eutrophication is the most serious environmental problem in the coastal marine pollution.Macroalgal blooms tend to have the potential for rapid absorption of nutrients and rapid proliferation.In order to investigate the physiological and biochemical responses of Ulva prolifera to nitrogen(N)and phosphorus(P)in the eutrophic seawater,the growth rate,photosynthesis and the pigments of U.prolifera were determined under different nutrients conditions and different culture densities.The experiment was divided into four treatments:the control group,+N group,+P group,and +N+P group(+N,+P represents adding NaNO3,Na2 HPO4,respectively.).The algae were pre-cultured in four nutrition conditions for 3 d.The temperature was set at 20 ℃,light intensity was approximately 6 000 lx,and photoperiod L∶ D was 12 h∶ 12 h with continuous aeration.Our results showed that either N or P supplies could increase the growth rate of U.prolifera,but much more significant enhancement was found in the N enrichment than P enrichment.When the original biomass was set as 0.1 g/L,the relative growth rates of the control group,+P group,+N group and +N+P group were 0.68,0.69,0.72 and 0.75,respectively.There was a negative relation between the relative growth rate and biomass density.The biomass doubling time significantly shortened when N and P were added.When the original biomass was 0.2 g/L,the biomass doubling time of control group,+P group,+N group and +N+P group was 1.60,1.53,1.16,and 1.03 d,respectively.The fastest biomass doubling time was found in the thalli grown at N and P enrichments condition and 0.1 g/L initial biomass density,with the period of 0.92 d.Either N enrichment or P enrichment could increase the chlorophyll and carotenoid contents,as well as the net photosynthetic rate of U.prolifera.The maximum net photosynthetic rate and pigments contents in U.prolifera were found in the culture condition with a combination of simultaneous enrichment of N and P.There was a siganificant interaction between N and P enrichment.In summary,N,P enrichments can significantly promote the growth,increase the photosynthetic pigments and photosynthetic rate of U.prolifera,which may be an important mechanism of the bloom of U.prolifera.

Seven populations of Litopenaeus vannamei were collected to evaluate the general combining ability and the specific combining ability in harvest body weight and survival with the mixed linear model and generalized linear models,combined with constrained maximum likelihood method.Incomplete diallel crosses and nest design were adopted and 130 full-sib families were obtained by artificial insemination.The results showed that the populations of UA5,UA4 and SIN were higher than others in general combining ability of harvest body weight,the magnitude were 0.40,0.32 and 0.19,respectively.The general combining ability of harvest body weight and survival in the hybrid groups of UA5×SIN,UA1×UA2,UA4×UA5,UA4×SIN and UA1×SIN,which were priority combinations in producing superior offspring,had positive advantages.The analysis of variance of combining ability showed that in different environments,the proportion of GCA and SCA variance component in body weight traits had little difference.Correlation analysis showed that there is positive correlation between the GCA of harvest weight,but negative correlation in survival.For the study of general combining ability and specific combining ability in different groups,the experiment provided the basic data for further genetic breeding of L.vannamei.

In order to understand the muscle nutrient characteristics of the sword prawn Parapenaeopsis hardwickii,the nutritional composition and quality of muscles from the wild and cultured sword prawn were investigated and analyzed by biochemical analysis methods.The results show that the crude fat content of wild group(0.89%)was significantly lower than that of cultured group(1.12%),while the crude ash content of wild group(1.44%)was significantly higher than that of cultured group(1.31%).No significant difference in moisture and crude protein contents of fresh muscles was found between wild(79.42% and 17.94%,respectively)and cultured group(79.28% and 17.95%,respectively).18 kinds of amino acids were found in muscles from wild and cultured groups,and there was no significant difference in the content of 9 kinds of amino acid between wild and cultured groups,while in other 9 kinds of amino acids,the amino acid content of cultured group was significantly higher than that of wild group.Total amino acid(TAA),total essential amino acid(EAA),total non-essential amino acid(NEAA),total delicious amino acid(DAA),and total shrimp flavor amino acid(SAA)content of cultured group(88.08,30.53,47.39,34.47,and 21.72%,respectively)were significantly higher than those of wild group(83.42,29.69,43.59,32.54,and 21.11%,respectively).The essential amino acid index(EAAI)and the ratio of branched chain amino acid to aromatic amino acid(F value)of cultured group(69.10 and 2.12,respectively)were significantly higher than those of wild group(66.41 and 2.07,respectively).The saturated fatty acids(∑SFA)and poly-unsaturated fatty acids(∑PUFA)relative content of cultured group(39.60% and 36.64%,respectively)were significantly lower than those of wild group(41.95% and 39.51%,respectively),while the mono-unsaturated fatty acids(∑MUFA)relative content of cultured group(23.76%)were significantly higher than that of wild group(18.54%).The EPA+DHA relative content of wild group(28.23%)was significantly higher than that of cultured group(25.66%),while no significant difference was observed in the EPA+DHA absolute content of muscle between wild and cultured groups.Therefore,the sword prawn is nutritional,delicious,and artificial culture did not cause abundant accumulation of fat in muscle;on the contrary,it improved protein nutritive value of the muscle.

Successive green tides that happened in South Yellow Sea in recent years have caused widespread concern.In order to provide the basis for making integrated use of green tide algae,and according to national standards,this study analyzed basic nutrients,chlorophyll,amino acids,mineral elements and heavy metals of green algae collected from Jiangsu and Shandong in 2010.The results indicated that:carbohydrates were the main nutritional components of these green tide algae,accounting for 35.82%-52.43%;the crude protein contents varied greatly,from 31.04% to 12.11%;the fat contents were low and all less than 1%;there was a significant difference in the contents of chlorophyll,the contents decreased significantly when the collection dates delayed;the amino acid contents were high and the percent age of essential amino acids was up to 37.45%,the flavor amino acids aspartic acid,glutamic acid,glycine and alanine were considerably high;these algae contained abundant mineral elements,the contents of Ca,Fe,Zn were distinctly high,but the heavy metals Pb,Hg and Cd were all lower than the limited requirements in national standards.The composition of attached samples did not change significantly.

Based on the aggregation experiments of feature-reef trough,the occurrence frequency of Sparus Macrocephalus larvae around artificial fish reefs was studied with different structures and at different background velocities.The highest frequency of fish occurrence,space volume of flow velocity of 0.7 times and0.8 times to background were considered as indexes of wake vortex range.The space volume with vertical upwelling velocity 0.1 time and 0.2 time of the upwind horizontal velocity was considered as the index of upwelling range.Based on Fluent with a turbulence model named LES(Large Eddy Simulation),a numerical trough was built and four unit-reef assemblages as an example were adopted to analyze the difference and merits of various assemblages' flow effect.The result showed it was appropriate to place 20-30 unit reefs(cube,3 meters of side length)as a 5-point symmetric mode,and 1-2 times reef distance in reef area,as this could not only bring about synergistic effect of reefs,but also optimize the regulation range of unit-reef.

Hybridization and backcrossing between Crassostrea hongkongensis and Crassostrea ariakensis were conducted in this study,and consequently,the experimental groups HH(C.hongkongensis),AA(C.ariakensis),HA(C.hongkongensis ♀×C.ariakensis ♂),AH(C.hongkongensis♂×C.ariakensis♀),HAHA(HA♀×HA♂),HHA(HH♀×HA♂),HAH(HA♀×HH♂),HAA(HA♀×AA♂),AHA(AH♀×AA♂)were produced using the method of diallel cross.The results showed that C.hongkongensis eggs can be fertilized by C.ariakensis sperm(47.33%±2.52%),but not vice versa.The fertilization can occur between HA and HH or AA.The fertilization rates of backcrossing groups were lower than those of inbred groups HH and AA.The hatching rates of the backcrossing between HA and AA were lower than those of the other groups,which were 20.33%±2.08%(AHA),55.67%±2.52%(HAA)respectively.The larval survival rate of the AHA and HAHA was 0;The survival of HA larvae was lower than HH and AA.The hybrid larvae grew faster than C.ariakensis larvae,but grew slower than C.hongkongensis.The survival rates of HA,HH,and AA juveniles were not significantly different(P0.05).The hybrid juveniles grew faster than C.hongkongensis larvae,but grew slower than C.ariakensis.In addition,the larval and juvenile growth of HHA and HA was not significantly different(P0.05).The larval survival rate was not significantly different in HHA and HA.The juvenile survival rate of HHA was lower than HA,but was higher than that of the other backcrossing groups.The results provide a basis for study on the hybridization and backcrossing between C.hongkongensis and C.ariakenis.

The arsenic speciation and the content of four toxic arsenic(including As3+,As5+,MMA and DMA)in several main economic shellfishes in our country were analyzed using HPLC-ICP-MS combined with in vitro digestion method.Results showed that S.strictus had the highest total arsenic content(5.68 mg/kg),the second was in S.Subcrenata(4.83 mg/kg),the total arsenic content in C.Farreri,C.Ostreae and H.discus hannai were much lower(2.26-2.70 mg/kg)and three of them had no significant difference.The lowest content was in R.Philippinarum and M.edulis.AsB was the main arsenic speciation in both water extraction and gastrointestinal extraction for all samples,and especially for Scapharca subcrenata,AsB was the only arsenic speciation.About 0.025,0.008 and 0.300 mg/kg As3+ were separated in the water extraction of C.farreri,R.philippinarum and S.strictus respectively.In addition,0.115 mg/kg As5+ was also detected in the water extraction of S.strictus.Different concentration of DMA was found in the water extraction of all samples except for S.subcrenata and M.edulis,with the highest content of 0.674 mg/kg in S.strictus.However,except in S.strictus,DMA was detected in the intestinal extraction,none of four toxic arsenic speciations were found in the gastrointestinal extraction of other shellfishes.Present experiments proved that the arsenic speciation in the water extraction which could reflect the real content in shellfishes and in gastrointestinal extraction which could reflect the real content under the gastrointestinal digestion was different,especially for four toxic arsenic speciation.
