STUDY OF ABSORPTION SPECTRA OF THE COMPLEXES OF BOVINE SERUM ALBUMIN WITH HOECHST 33258 AND METHYLENE BLUE 牛血清白蛋白与赫斯特33258和亚甲基蓝配合物的吸收光谱研究

The study on the interaction of DNA-specific low-molecular compounds – groove binding material Hoechst 33258 and intercalating ligand methylene blue (MB) with serum albumin has been carried out. The absorption and differential absorption spectra of complexes of the mentioned ligands with protein were obtained. Changes of the absorption and differential absorption spectra indicate the binding of two ligands with albumin. The obtained results indicate that at the interaction with both ligands, the conformational state of the protein alters, though these changes are not similar, since in the case of MB a compactization of the protein folding occurs, while in the case of Hoechst 33258, most apparently, an unfolding of the compact structure takes place as a result of partial loss of helicity of $alpha$-structures.

RADIOSENSITIVITY OF SOFT WHEAT SEEDS TO GAMMA RADIATION 软质小麦种子对γ射线的辐射敏感性

The study of the effect of different doses of $gamma$-irradiation on the growth and development of seedlings of soft wheat seeds, resistance, and germination of seeds to irradiation has been carried out. The data show that the seeds of common wheat are resistant to ionizing radiation and up to 80% of the seeds remained viable. Under the influence of ionizing radiation, the functional and mitotic activity of the cell nucleus is disrupted, which has a significant effect on the growth parameters of the seedlings of irradiated seeds.
本文研究了不同剂量γ辐照对软质小麦种子幼苗生长发育、抗性和萌发的影响。数据显示,普通小麦的种子对电离辐射有抵抗力,高达80%的种子仍能存活。在电离辐射的影响下,细胞核的功能和有丝分裂活性被破坏,这对辐照种子幼苗的生长参数有显著影响。

The formation of Ni2+:Tryptophan (Trp) 1:1 complex, which acts as a model catalyst for decomposition of cumene hydroperoxide (ROOH) in Ni2++Trp+ROOH+H2O system, has been confirmed via kinetic study in aqueous solution at pH>7. The kinetic expression of a single catalytic decomposition reaction of ROOH under the influence of [NiTrp]+ complex was brought out. The temperature dependence of the effective rate constant of ROOH decay (Keff=Kcat[Ni2+]0[Trp]0=const) in the temperature range from 323 to 343 K can be expressed by Arrhenius equation (Eeff is in kJ/mol):  Keff=(1.87±0.02)·106exp[–(49.8±0.3)/RT], min –1.

On the basis of cyclic esters а method for producing of γ-hydroxybutyric acids hydrazides has been elaborated. The antioxidant properties of hydrazides were researched by the method of competitive reactions and it was established that they have antioxidant activity.

INCREASING OF THE SUPEROXIDE DISMUTASE TOTAL ACTIVITY IN MICROGLIAL CELLS UNDER THE TREATMENT BY $RIBES$ $NIGRUM$ L. ALCOHOL EXTRACT 枸杞醇提取物对小胶质细胞超氧化物歧化酶总活性的影响

Cell redox homeostasis is formed as a result of the balance between the accumulation of reactive oxygen species and functioning of the antioxidant enzymes or non-enzymatic antioxidants. The alcohol extracts from the leaves of Ribes nigrum L. were prepared to investigate the antioxidant activity and protective effects against both BV-2 microglial wild type (WT) cells and acyl-CoA oxidase type 1 (ACOX1) deficient cell (Acox1–/–) lines․ Our investigations showed that R. nigrum extracts possess remarkable antiradical activity in 1-diphenyl-2-picryl­hydrazyl (DPPH) with IC50 value of 32.7±0.8 µg mL–1. 3-(4,5-dimethyltrazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay showed that the sub-cytotoxic concentration of R. nigrum extract in both cases was 1 mg mL–1 The further treatment of microglial cells with the mentioned concentration of extract brought the remarkable increasing (up to 350%) of activity of the total superoxide dismutase (SOD) – the core antioxidant enzymes, under the treatment during the 48–72 h. So, relaying on the obtained data it is possible to insist that the lack of ACOX1 can be compensated by the activation of cell enzymatic antioxidant defense mechanisms.

An assessment of the ecological state of the Voghji River and its catchment basin according to chemical and biological indicators in the period 2018–2019. New taxa were discovered in phytoplankton, when contaminated with heavy metals and organic substances. A decrease in community stability and an increase in the role of Сyanoprokaryotes and green algae in plankton were observed, which can be used for phytoremediation of the degraded ecosystem of the Voghji River.

The interaction of Hoechst 33258 (H33258) and methylene blue (MB) compounds with bovine serum albumin (BSA) has been studied using the method of thermal denaturation. The obtained data showed that both ligands form complexes with BSA, moreover, MB binds to BSA stronger than H33258. Furthermore, H33258 destabilizes, while MB stabilizes the native structure of protein, leading to the decrease and increase of the denaturation temperature of BSA respectively.
采用热变性法研究了Hoechst 33258 (H33258)和亚甲基蓝(MB)化合物与牛血清白蛋白(BSA)的相互作用。结果表明,两种配体均能与BSA形成配合物,且MB对BSA的结合强于H33258。H33258使蛋白失稳,MB使蛋白的天然结构稳定,分别导致牛血清蛋白变性温度的降低和升高。

THE IMPACT OF HORSERADISH ROOTS ($ARMORACIA$ $RUSTICANA$) ON THE FUNCTIONAL STATE OF RAT BODY IN ACUTE HYPOXIA 辣根对急性缺氧大鼠机体功能状态的影响

Using the “Bioscope” hardware complex, it was shown that an oxygen deficiency leads to certain changes in the statistical parameters of the integrative state of animals both during and after hypoxia exposure. However, after 10 days of using horseradish roots (Armoracia rusticana) as an antihypoxic food supplement, in contrast to the control, significant changes in the recorded signal are observed both during and after hypoxic exposure. These changes indicate the facilitating effect of horseradish roots in conditions of oxygen deficiency.

SOIL MICROBIAL BIOMASS AND ENZYME ACTIVITIES RESPONSE TO COPPER AND MOLYBDENUM POLLUTION IN THE VICINITY OF ZANGEZUR COPPER AND MOLYBDENUM COMBINE, ARMENIA 亚美尼亚赞格祖尔铜钼联合厂附近土壤微生物生物量和酶活性对铜钼污染的响应

The aim of this study was to define a relationship between heavy metal (Cu, Mo) pollution of soil and various extracellular enzyme activities. Six enzymatic activities involved in cycles of carbon, nitrogen, phosphorus and sulfur (β-glucosidase, chitinase, leucine-aminopeptidase, acid phosphomonoesterase, alkaline phosphomonoesterase, and arylsulphatase) as well as microbial biomass were determined in soil samples collected in the surroundings of Zangezur Copper and Molybdenum Combine. The investigations showed that pollution of soil with copper and molybdenum led to a decrease in microbial biomass and soil enzymatic activity, which in turn had a negative impact on cycles of chemical elements, in particular C, P, N and S. This gives reason to conclude that the changes in soil microbial biomass and enzymatic activity may act as indicators of soil biological activity and quality.

INFLUENCE OF MILLIMETER RANGE ELECTROMAGNETIC WAVES ON HEMOLYSIS KINETICS OF RAT BLOOD ERYTHROCYTES 毫米波对大鼠红细胞溶血动力学的影响

The effect of millimeter range electromagnetic waves (MM EMW) on kinetics of acidic hemolysis of rat blood erythrocytes has been studied. It was shown that MM EMW affect the duration of erythrocyte hemolysis and this change depends on MM EMW exposure and frequency. At the irradiation by frequencies 41.8 and 42.2 GHz the acidic hemolysis occurs quicker at all durations of the irradiation compared to the control. At the irradiation by 50.3 and 51.8 GHz the hemolysis takes place quicker as well, as compared to the control at short durations of the irradiation. At the exposure 60 min the hemolysis start is delayed.
