A NEW RP-HPLC METHOD FOR ESTIMATION OF VERICIGUAT IN BULK DRUG AND PHARMACEUTICAL DOSAGE FORM 一种新的 RP-HPLC 方法用于估算散装药物和药物剂型中的 vericiguat

A simple, rapid, specific and accurate reverse phase high performance liquid chromatographic method has beendeveloped for the validation of Vericiguat in bulk as well as in marketed pharmaceutical dosage form. This separation wasperformed on a Symmetry ODS C18 (4.6×250mm, 5μm) column with Methanol:Phosphate Buffer (55:45) v/v asmobile phase at a flow rate of 1.0 mL min−1 with UV detection at 225 nm; the constant column temperature wasambient. The run time under these chromatographic conditions was less than 8 min. The retention time of Vericiguatwas found to be 2.252. The calibration plot was linear over the concentration range of 6 - 14 μg mL−1 with limits ofdetection and quantification values of 1.2 and 3.6 μg mL−1 respectively. The mean % assay of marketed formulation wasfound to be 99.86%, and % recovery was observed in the range of 98-102%. Relative standard deviation for theprecision study was found <2%. The developed method was simple, precise, specific, accurate and rapid, making itsuitable for estimation of Vericiguat in bulk and marketed pharmaceutical dosage form dosage form.

APPLICATIONS OF GREEN CHEMISTRY PRINCIPLES IN TEXTILE WET PROCESSING 绿色化学原理在纺织品湿加工中的应用

The conventional finishing and dyeing processes practiced in the textile industry have a huge impact on the environmentand human health. This impact is due to the use of toxic chemicals, huge amounts of water usage, discharge of theeffluents generated by the textile industry in the water bodies, inefficient treatment of the discharged, water,atmospheric emissions and high energy consumption. The aim of the present study is to discuss the applications of greenchemistry principles in eco-friendly textile wet-treatment processes. This paper discusses the use of enzymes(biochemical approach) and physical treatment processes like ultraviolet radiation, plasma, and ozone technology. In caseof dyeing it discusses the use of natural dyes and technologies like Air Dye, ultrasonic, microwave, plasma, supercriticalCO2, electrochemical and Nano-DyeTM.

Since ancient times, Anacyclus pyrethrum has been utilized as an aphrodisiac. In Ayurvedic medicine, the plant is used toboost libido and enhance sexual performance. Pyrethrins and anacyclins, among other plant substances, improve vaginalblood flow and sensation. It raises libido and promotes hormone synthesis. The plant is also claimed to have powerfulantioxidant qualities that enhance overall well-being. In addition to being used for its intended purpose, Anacycluspyrethrum is thought to provide potential health advantages. It functions as an oxygen inhibitor and has anti-inflammatoryqualities that might help in the fight against chronic illnesses, including cancer and heart disease. Additionally, someresearch has revealed that the herb may have neuroprotective qualities, making it a viable therapy for brain diseases likeAlzheimer's. The herb Anacyclus pyrethrum has a long history of use as a traditional medicine. It is a powerful naturalmedicine thanks to its aphrodisiac effects and oral health advantages. Additionally, the possible health advantages outsideof its regular use make for an intriguing research topic.

HISTOPATHOLOGICAL EFFECT OF CHLORPYRIFOS PESTICIDE ON LIVER AND KIDNEY OF FRESH WATER FISH CATLA CATLA 毒死蜱农药对淡水鱼鲶鱼肝脏和肾脏的组织病理学影响

All the natural water bodies are continuously being contaminated with toxic chemicals from industrial, agricultural and domestic activities. Chlorpyrifos is one of the most widely used organophosphate pesticides. Excessive application of pesticides from agricultural fields contaminate aquatic medium, resulting in serious damage to non-target species, including fish. The fish as a bioindicator of aquatic medium plays an important role in the monitoring of water pollution. Histopathology is an important tool for evaluating the action of any toxicant at tissue level. The focus of the present study is to measure the histological changes in liver and kidney of the fresh water fish Catla catla exposed to sub lethal concentrations of chlorpyrifos 1/15th of the 96 hour LC50 values for the period of 10 and 20 days. The fish exposed to chlorpyrifos showed mild vacuolar hepatocytes, dilation of central vein, vaculor degenerative changes, mild hyperaemia and severe hyperaemia of central vein in the liver tissue slide. The changes observed in kidney slide due to the impact of chlorpyrifos toxicity are hemorrhage, degenerative changes in between tubules, constriction of primary tubule, vacuolardegenerative changes, necrosis in epithelial lining of proximal tubules, chlorpyrifos compound induced.

DEVELOPMENT AND VALIDATION OF VISIBLE SPECTROPHOTOMETRIC METHOD FOR ESTIMATION OF ELTROMBOPAG OLAMINE IN BULK AND TABLET DOSAGE FORM 建立和验证可见分光光度法测定散剂和片剂中艾曲波帕乙醇胺的含量

A simple and sensitive visible spectrophotometric method has been developed for the quantitative estimation of Eltrombopag Olamine in its bulk and tablet dosage form. It is an orally active ethanolamine salt of Eltrombopag, a smallmolecule, nonpeptide thrombopoietin receptor agonist with megakaryopoiesis-stimulating activity. The maximum absorbance of drug was found at 425nm. Eltrombopag Olamine was found to be dissolved in Methanol so it was used as solvent for the conduction of work. Linearity range for the method was finalized as 3-18 μg/ml with R2 value of 0.9995. % RSD less than 2 which is well within indicates the method is precise and robust. The % recovery rate was found to be 100-101% indicating the method’s accuracy. The method was validated as per ICH guidelines provided and the results were found to be satisfactory. LOD & LOQ values were obtained as 0.150 & 0.456 respectively. Stability studies shows that the drug is unstable in various stress conditions like acid, base, water and oxidation.

Amorphous solid dispersion (ASD) by hot-melt extrusion (HME) is an industrially feasible approach to overcome the solubility and bioavailability limitations of poorly soluble active pharmaceutical actives. The creation of HME-based ASDs was significantly impacted by the implementation of Quality by Design (QbD). The objective of the study was to develop an ASD of posaconazole for the effective management of mucormycosis. The impact of change in levels of extra granular materials were identified as critical quality attributes (CQA's) for the development of delayed release tablet. A 23 full factorial design was employed to study the impact of independent variables HPMCAS (X1), HPC(X2) and CCS (X3) as CQA’s on the dependent variables such as hardness (Y1) and Disintegration Time (Y2) and % Drug release (Y3). The design was analyzed by using ANOVA by using MINITAB software. The influence of the extra granular components on the dissolution of tablet was closely evaluated while finalizing the optimized batch. The extrudes were also evaluated by FTIR, XRD and DSC analysis which confirmed the purity and in-situ conversion from crystalline to amorphous form ofthe drug. Systematic development of a bioavailable and stable ASD of posaconazole was achieved by studying the CQA’s at different levels by using the QbD and from the stability data, the optimized batch F4 was found to be stable up to 3 months at accelerated conditions40°C/75%Rh.

The creation of HME-based ASDs was significantly impacted by the implementation of Quality by Design (QbD). The objective of the study was to develop an ASD of posaconazole for the effective management of mucormycosis. The impact of change in levels of extra granular materials were identified as critical quality attributes (CQA's) for the development of delayed release tablet. A 23 full factorial design was employed to study the impact of independent variables HPMCAS (X1), HPC(X2) and CCS (X3) as CQA’s on the dependent variables such as hardness (Y1) and Disintegration Time (Y2) and % Drug release (Y3). The design was analyzed by using ANOVA by using MINITAB software. The influence of the extra granular components on the dissolution of tablet was closely evaluated while finalizing the optimized batch. The extrudes were also evaluated by FTIR, XRD and DSC analysis which confirmed the purity and in-situ conversion from crystalline to amorphous form ofthe drug. Many naturally occurring substances, traditionally used in the medicines contain the coumarin moiety. Coumarin is asimple scaffold widespread in nature and it is found in a number of plants as well as fungi and some bacteria. In the lastyears, it gains attention due to its ability to interact with diverse enzymes and receptors in living organisms. Coumarinbased compounds having potential not only in the medicinal chemistry but also in the cosmetic and also in fragrances. It ischemically a Benzopyran 2-one. It possess a wide range of pharmacological activities including anti-cancer, anticoagulant,antibacterial, anti-inflammatory, anti- oxidant, antidiabetic, antiparkinson’s activity and also found antihepatitis Cactivity. Its versatile range of activities makes it a versatile scaffold in a medicinal chemistry. Therefore, in this review weanalyzed a various research articles, reviews and the data found from them are collected. Here in this overview, wecollect various literature on the synthesis of possible reaction on coumarin molecule.

Obstructive Sleep Apnea (OSA) is a debilitating condition stemming from disruption to the respiratory system during sleep. At present, the nature of the relationship between OSA and mood, specifically depression and anxiety, is still unclear. The purpose of this paper is to shed some light on this relationship. There is an urgent need to better understand the roles of psychological illness in OSA. Well-designed longitudinal studies are needed to examine temporal  relationships between the OSA, and psychological diseases and further research is needed to establish the role of confounders, and effect modifiers such as gender, in any apparent relationship. Sleep clinicians are advised to consider psychological diseases a likely cause of sleepiness and fatigue. Several possible causal mechanisms linking OSA, and depression have been proposed but not established. Patients who have depression as well as OSA appear worse off than those with OSA only, and depressive symptoms persist in at least some patients in short term studies of treatment forOSA. Direct treatment of psychological diseases in OSA might improve acceptance of therapy, reduce sleepiness, and fatigue and improve quality of life, but intervention trials are required to answer this question.

Oral cancer is known for its devastating effects which need to be addressed at the molecular level to achieve the best possible outcomes. An upsurge in the COX-2 levels amongst premalignant & malignant tissues may be attributed to increased transcription along with enhanced mRNA stability. Compelling research evidence is being shown with respect to complexes which have dual COX-2/COX-1 inhibitors are beneficial in chemotherapeutic procedures of cancer.Amongst 15 compounds, we found the highest binding capacity to be -10.9 in rutaecarpine as per the ADV webserver and -11.2 in VX-809 (Lumacaftor) as per the ProdigY webserver. Highest hydrophobicity was seen in OSR, LMR & ECG weighing 389.33, 452.41 & 458.37 with the most common amino acids being Leu (338), Ser (339), Val (509), Val (335). In respect to the molar refractivity, OSR & LMR presented with 92.01 & 113.98 values, whereas the lipophilisity was found to be 2.46 & 3.08 respectively with toxicity score (assessed by toxi M score & ProTox), being 0.929 & 0.944 by ToxiM score & 4 in both compounds by ProTox class.Characteristic properties such as the hydrophobicity, hydrophilicity, molecular weight, number of H-bond acceptors, Num of H-bond donors, molar refractivity, lipophilisity (ilogP), drug likeliness and toxicity score play an important role in understanding drug-drug interactions at the molecular level which helps in determining the pharmacological actions and its application in humans. Therefore, further research in the field is recommended to comprehend the drug reactions & their outcome.

IMPACT OF HEMODIALYSIS ON HEALTH-RELATED QUALITY OF LIFE OF THE PATIENT 血液透析对患者健康相关生活质量的影响

Hemodialysis is a procedure used to clean the blood of people whose kidneys are not functioning properly. Duration of dialysis is 4-6 hours, twice or thrice a week. Hemodialysis cleans the kidneys of waste products including urea and creatinine along with free water extracorporeal, which slows the progression of kidney failure. The observational study included 80 patients who received hemodialysis.Our study was conducted on 80 patients out of which were found to be males (61.25%) and were found to be females (38.75%). The majority of the patients there (28.75%) were between the ages of 51 and 60. Among 80 patients, complications were reported in 64 (80%) patients. Commonly reported complications were weakness, muscle cramps and fever. Out of 80 patients, 16 patients (20.0%) were having long-term hypertension and 12 patients (15%) were having both hypertension and diabetes as co-morbidity. Participants in our study reported that the above-mentioned problems significantly lower the life quality in terms of the health.
