The effect of forest gap disturbance on the diversity and stability of species in the regeneration layers of natural Castanopsis kawakamii forest were studied by applying biodiversity indices and improved M.Godron's stability methods in C.kawakamii natural reserve.The results showed that: forest gap disturbance improved its micro-environment,as reflected in the differences in importance values and biodiversity in the tree and shrub layers of forest gap and understory.The biodiversity indices of richness index R,domination index D,Shannon-Wiener diversity index H,and evenness-dominance-abundance index Z in the forest gap tree layer were higher than those of the forest understory,while evenness index Js and community dominance C were lower than those of the forest understory.The biodiversity indices of R,D,H,J s,and Z in the forest gap shrub layer were higher than those of the forest understory,while C were lower than those of forest understory.Biodiversity in the forest gap was richer with less evident ecological dominant species,which indicated that the population in the forest gap was developing in the direction of complexity and homogeneity.The similarity coefficients of C j,C s,and C N were relatively low,while C mH was relatively high in the tree layer of the forest gap and understory,which demonstrated that the forest gap promoted the regeneration of trees.The similarity coefficients of C j,C s,C N,and C mH were relatively high without exception and the species composition in the forest gap were basically identical with those of the understory.According to the theory of M.Godron and the simulated the optimum stability model,the logarithm model and quadratic objective function model were the best stability models in tree layer of the forest gap and understory,respectively,and the optimum stability model of the shrub layer in both the forest gap and understory was the logarithm model.The plants in the tree layer of the forest gap were stable,but were unstable in the understory.The plants in the shrub layer of both the forest gap and understory were stable.The importance value of Litsea subcoriacea was the highest in the tree layer of the forest gap and understory,which showed that it played an important role in forest regeneration and would likely have a greater impact on the regeneration of C.kawakamii population.The importance value of C.kawakamii was relatively low in the tree layer of the forest understory,which impacted its regeneration.However,forest gap disturbance improved the upward growth and importance value of C.kawakamii,which could achieve the regeneration and restoration of the C.kawakamii population effectively.

Based on long-term tracking retest data and three structure parameters(mingling intensity,neighborhood comparison and uniform angle index),we investigated the effects of four different selective cutting intensities(low intensity of 13.0% in volume,medium intensity of 29.1%,high intensity of 45.8%,and over-high intensity of 67.1%) on forest spatial structure change in a natural forest.After low and medium selective cutting forest,plot average mingling and neighborhood comparison increased while high and over-high selective cutting decreased.Selective cutting did not change the forest spatial distribution pattern.However,the variety of forest plot showed aggregated distribution,and medium selective cutting in random distribution state forest number increased considerably.After low and medium selective cutting,forest spatial structure was optimized in general.

Interspecific hybridizations between different varieties of Brassica rapa L.ssp.peki-nensis(Lour.) Olsson and Brassica oleracea var.capitata L.were carried out.The hybrids were obtained by successive bud pollination and ovule culture and tested by means of identification by cytology.Results showed that 57 plants were obtained by interspecific hybridization between B.rapa L.ssp.pekinensis and B.oleracea var.capitata L.Cytological identification and pollen characteristics survey revealed that 47 of the hybrids had the anticipated chromosomes(2n=19) and pollen fertility was aborted;while 6 hybrids had 38chromosomes and were fertile.The BC1 plants were obtained through back cross between fertile hybrids F1 and B.rapa L.ssp.pekinensis.Field investigation showed that all F1 hybrids had many characteristics from both parents,the BC1 plants had obvious heading and were similar to B.rapa L.ssp.pekinensis.

Two quadrat scales,1 m×1 m and 25 cm ×25 cm,were established to investigate the vegetation of the Jinchuan Peatland.Interspecific correlations among 9 bryophytes and 30 main vascular plants were analyzed by X 2-test,Pearson correlation,and Spearman rank correlation quantitatively.There were significantly negative associations among three main peatmosses: Sphagnum subsecundum,S.palustre,and S.capillifolium,each of which occupied different habitats.Accordingly,four ecological species groups were classified.Spearman rank correlation analysis showed that positive association amounted to 49.53% of the 741 species pairs in the large quadrats,and more than 41.14% of the 666 species pairs in the small quadrats.This study suggests that quadrat size played a crucial role in analyzing interspecific associations in peatlands,with great disparity in body size of different plant groups.It appears more suitable to analyze interspecific associations among bryophytes and among bryophytes and-vascular plants with small and large quadrats,respectively.

Heat shock proteins(Hsp),representing an important molecular chaperone in eukaryotic cells,is a common response to development,stress resistance,signal transduction and evolution of plants.The relationship between the structure and functional roles was elucidated in Hsp90 based on the generation of full-length cDNAs from Senecio scandens Buch.-Ham.ex D.Don.Sequence analysis of Hsp90-3 gene indicated that it shared 93.71% identity with Arabidopsis thaliana(GenBank accession: NP_200412.1),encoding a protein composed of 699 amino acid residues with the predicted molecular weight of 79.78 kD and theoretical isoelectric point of 5.08.Moreover,the distribution of Hsp90-3 was involved in the endomembrane system such as nuclei,peroxisomes,chloroplast thylakoid membranes,and chloroplast matrices in the present study.Three-dimensional measurement revealed that the Hsp90-3 protein was composed of three structural domains and one link region.These results suggested that Hsp90-3 played a critical role in molecular chaperone,signal transduction,transcriptional regulation and stress-response in higher plants.

Nostoc flagelliforme is a terrestrial nitrogen-fixing cyanobacterium with strong drought adaptability.We performed 2-DE analysis,gel image analysis and MALDI-TOF-TOF/MS to compare and identify differential proteins from N.flagelliforme subjected to desiccation.The results indicated that Ferritin,Dps family protein expression quantum gradually declined under desiccation stress.The Ferritin gene was cloned by designed degeneracy primer based on identified amino acid sequences.A full length of 540 bp DNA was obtained(GenBank access number: HM854287).Homology analysis showed that the N.flagelliforme Ferritin gene had high consensus regions.The secondary structures of Ferritin were made up of α helix and random coil.The RT-PCR showed that mRNA of Ferritin gradually declined under drought stress.The Ferritin gene was expressed in Escherichia coli,and a 22.4 kD heterologous protein was observed.The results laid a foundation for understanding the drought-resistant molecular mechanisms and protection mechanisms of N.flagelliforme in extreme arid environments.

To study the heat tolerance of chrysanthemum(Chrysanthemum morifolium),the electrical impedance spectroscopy(EIS) parameters of chrysanthemum'Jinba'stems and leaves under high temperature stress(25℃ to 50℃) were measured,and the relationship between electrolyte leakage(relative conductivity) and EIS parameters was analyzed.The results showed that extracellular resistance,intracellular resistance,and relaxation time of stems and leaves first increased and then decreased significantly with increasing temperature.However,the trends of distribution coefficient of relaxation time of stems and leaves showed irregular decrease under high temperature stress.The relative conductivity of chrysanthmum increased,then decreased,and finally increased significantly.Heat tolerance of chrysanthemum estimated by EIS parameters was close to that assessed by the EL method.Significant correlation between relative conductivity and extracellular resistance(stem: p0.01,leaf p0.05) and relaxation time(p0.05) of stem and leaf were found.In conclusion,extracellular resistance and relaxation time could be suitable parameters for determining heat tolerance of chrysanthemum.

Euptelea pleiospermum,a Tertiary relic and East-Asian endemic species,is a rare and threatened species in China,which requires protection.Based on stem mapping data,the spatial characteristics of E.pleiospermum in the Shennongjia Mountains were examined by nearest neighbor distance statistics.Following the pattern differences and changes within and between each life-history stage(represented by size class,seedling ≤2.5 cm,2.5 cmsapling≤7.5 cm,adult tree7.5 cm in DBH),the pattern-forming processes such as seedling establishment,recruitment and intra-specific competition were inferred and assessed.Euptelea pleiospermum was overall aggregated at the neighborhood scale.Sizes of small individuals(seedlings or saplings) showed no correlation with distances to nearest larger individuals(saplings or adult trees).Within a given radius of focal large-size individuals(saplings or adult trees),the probability of occurrence of size-identical and smaller individuals(seedlings or saplings) were approximately equal.Mean nearest neighbor distance from large-size individuals to nearest individuals in consecutive lower class(sapling vs.nearest seedling,or adult tree vs.nearest sapling) did not differ significantly from that measure within large size classes(within sapling or adult tree classes).Both summed sizes of nearest neighbor pairs and sizes of focal individuals were significantly correlated with nearest neighbor distances.Mean nearest neighbor distance within sapling class did not differ from the expected one within random-mortality-introduced seedling class,and likewise mean nearest neighbor distance within adult class did not differ from the expected one within random-mortality-introduced juvenile class(seedlings+saplings).These results suggest that(1) the aggregated distribution of E.pleiospermum may be due to the effects of seed dispersal and/or habitat heterogeneity on seedling establishment,(2) within their neighborhood established large individuals of E.pleiospermum did not appear to limit the locations and growth of smaller con-specific individuals,but neighbor-interactions may have an effect on long-term recruitment,(3) intra-specific competition existed in the E.pleiospermum population but it was not strong enough to produce distance-dependent mortality.
