We developed a manipulation platform, which combines magnetic tweezers and the incubating system, with total internal reflection fluorescence microscopy (TIRFM). Our system can generate forces up to the nN range and such forces can be used to observe and characterize cell mechanical responses to external forces. Combined with TIRFM, cell surface stress-strain phenomena can also be investigated.

Photodynamic inactivation of Staphylococcus aureus planktonic and biofilm cells by a photosensitizer, merocyanine 540 (MC 540) was investigated. For the planktonic experiments, the antimicrobial activity was enhanced with the increasing light dose, but an increase in light dose could not further improve the antimicrobial activity if the maximum excitation level attainable lay below the necessary minimum threshold level. A complete inactivation was achieved when the excitation level of MC 540 lay somewhere above the threshold level. The relationship between antimicrobial activity and the excitation level of MC 540 revealed that the more MC 540 was excited, the more S. aureus cells were killed. For the biofilm experiments, the antimicrobial activity was enhanced with the increasing light dose. No viable cells were detected when exposed to 15 /spl mu/g/ml MC 540 and a light dose of 600 J/cm/sup 2/ or 20 /spl mu/g/ml MC 540 and a light dose of 450 J/cm/sup 2/. The results from both planktonic and biofilm experiments suggest that the antimicrobial activity of photodynamic inactivation against S. aureus is closely related to the excitation level of MC 540.

We demonstrated full-field optical coherence tomography (OCT) using achromatic phase shifting with a rotating polarizer. Using this phase shifter and conventional phase shift method, we obtained en-face OCT images of an onion at different depths.

For many years, the dorsal skinfold chamber has been used for intravital observation in animal models. We have developed a chamber built using medical-grade titanium. Our design allows long-term, high resolution, and large-area observation in nude mice in vivo. Combined with two-photon fluorescence and second-harmonic generation microscopy, our chamber can be used to obtain qualitative and quantitative information for in vivo analysis of diseases such as tumors.

An evanescent wave fluorescence-based fiber-optic biosensor is developed for high sensitivity, low cost, compact design and real-time in-situ monitoring. In the research, we accomplished the fiber-optical biosensor system to monitor the reaction of IgG-Anti rabbit IgG and Biotin-Streptavidin, and the kinetic association and dissociation constant of target analytes were calculated.

We present a phantom study of NIR tomographic imaging based on the pulsed-ultrasound modulation of diffuse light in turbid media. The bottleneck of the method is the very low S/N which cannot be overcome by the conventional signal-averaging because of the slowly-varying light phase on the detector. We proposed a simple model for the detected signal and validated it experimentally. Furthermore, we proposed the frequency-doubling-and-averaging method to recover the signal, and demonstrated its feasibility through the phantom experiment.
