ABSTRACTRecent trends towards greater fresh market use of ‘Bartlett’ pears has increased the need to extend its storage life to prolong the packing and marketing season in the United States Pacific Northwest region. Sixteen and 38%, respectively, of control fruit developed senescence disorders following 5 and 6 months of storage at −1.1°C. Commercial standard controlled atmosphere (CA) conditions (O2 at 1.5 kPa and CO2 < 1 kPa) or edible coating (Semperfresh™, SF) prevent the appearance of senescence disorders for 5 months, but 9% and 16% of fruit, respectively, developed senescence disorders after 6 months. The combination of CA+SF completely inhibited senescence disorders for 6 months. ABSTRACTBurrowing nematode (Radapholus similis) is a major threat to banana (Musa spp.) production throughout the world. All commercial banana cultivars are lacking of R. similis resistance. The conferred resistance available in non-commercial banana genotypes opens ways to develop new hybrids with R. similis resistance by conventional or molecular-breeding techniques. Understanding the mechanism of resistance is the primary step that can provide Musa breeders and molecular biologists with the necessary principles to create nematode-resistant plants. Phenols and lignin have been ascribed as post-infectional resistant mechanism in banana. However, information on anti-nematode compounds that are presented in resistant banana plants before challenge by R. similis is limited. Therefore, the roots of some resistant/tolerant genotypes were tested for anti-nematode bioactivity by root amendment study. This study reports on in vitro regeneration of Abutilon indicum plantlets through callus mediated organogenesis. The leaf explants implanted on Murashige and Skoogs (MS) medium supplemented with 4.52 µM 2, 4-Dicholorophenoxy acetic acid (2,4-D) and 8.88 µM 6 Benzyladenine (BA) showed highest response (70.3%) for callus proliferation, but these callus did not showed any morphogenetic differentiation on the same medium even after 12 weeks. Whereas, subsequent sub-culture of this green proliferated callus on MS medium added with 2.68µM α-Napthalene acetic acid (NAA), 8.88µM BA and 543 µM Adenine sulphate showed the highest frequency (62.2%) of multiple shoot-buds production and also elongation of shoots. Well developed shoots were efficiently rooted in vitro on half strength MS medium supplemented with 7.38 µM Indole-3-butyric acid (IBA). Seventy per cent of in vitro regenerated plantlets were successfully established in garden and were morphologically alike to the donor plants. The genetic homogeneity of these in vitro regenerated plantlets was also affirmed by inter simple sequence repeat (ISSR) analysis using eight ISSR primers. This standardised in vitro organogenesis protocol supplements a good platform for the conservation of A. indicum germplasms and also caters for the needs of the herbal industry.

ABSTRACTIn this study, we examined the covering effect of a new kind of infrared ray (IR)-cutting net to improve the low yield of summer-sown spinach crops in greenhouse. This IR-cutting net gives about 61% transmittance of visible light and approx. 50–54% absorbance of IR from 700 to 2600 nm. The utilisation of IR-cutting net in summer could favour plant growth due to lowering greenhouse temperatures. The globe temperatures when shielded by the IR-cutting net were approx. 6–8oC lower than without the net, an effect nearly equal to that of a 50% shade net. Although the plant growth was significantly reduced when plants were grown without a net, the crop productivity was intensified more by covering with the IR-cutting net than the 50% shade net. Mean fresh weight and yield for the IR-cutting net treatment was approx. 1.6 and 1.3 times larger, respectively, than for the 50% shade net. This IR-cutting net gives about 61% transmittance of visible light and approx. 50–54% absorbance of IR from 700 to 2600 nm. The utilisation of IR-cutting net in summer could favour plant growth due to lowering greenhouse temperatures. The globe temperatures when shielded by the IR-cutting net were approx. 6–8oC lower than without the net, an effect nearly equal to that of a 50% shade net. Although the plant growth was significantly reduced when plants were grown without a net, the crop productivity was intensified more by covering with the IR-cutting net than the 50% shade net. Mean fresh weight and yield for the IR-cutting net treatment was approx. 1.6 and 1.3 times larger, respectively, than for the 50% shade net. SummaryUsing an embryo rescue technique, we obtained an F1 plantlet whose female parent was a purple chilli pepper (Capsicum annuum 7033) and whose male parent was a green chilli pepper (C. chinense 7020).We verified the origin of this interspecific hybrid using a peroxidase isozyme assay and random amplified polymorphic DNA (RAPD) markers. Most of the morphological features of the F1 plant, particularly the purple colour of its foliage, flowers and immature fruit, were intermediate between those of its parents. Cytological analysis indicated that the meiotic chromosomes of the pollen mother cells of the F1 plant exhibited abnormal behaviour, resulting in a marked reduction in the proportion of fertile pollen compared to both its parents. Sequence analysis of the internal transcribed spacer of nuclear ribosomal DNA demonstrated that, compared to the parental sequences, the F1 hybrid had 186 polymorphic sites, 139 of which (74.7%) were inherited from the male parent, and 23 (12.4%) were inherited from the ...
